The main protease (Mpro) is a major protease having an important role in viral replication of the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), the novel coronavirus that caused the pandemic of 2020. Here, active Mpro was obtained as a 34.5 kDa protein by overexpression in E. coli BL21 (DE3). The optimal pH and temperature of Mpro were 7.5 and 37 °C, respectively. Mpro displayed a Km value of 16 μM with Dabcyl-KTSAVLQ↓SGFRKME-Edans. Black garlic extract and 49 polyphenols were studied for their inhibitory effects on purified Mpro. The IC50 values were 137 μg/mL for black garlic extract and 9–197 μM for 15 polyphenols. The mixtures of tannic acid with puerarin, daidzein, and/or myricetin enhanced the inhibitory effects on Mpro. The structure–activity relationship of these polyphenols revealed that the hydroxyl group in C3′, C4′, C5′ in the B-ring, C3 in the C-ring, C7 in A-ring, the double bond between C2 and C3 in the C-ring, and glycosylation at C8 in the A-ring contributed to inhibitory effects of flavonoids on Mpro.  相似文献

Hydrogels are materials with the ability to swell in water through the retention of significant fractions of water within their structures. Owing to their relatively high degree of biocompatibility, hydrogels have been utilized in a host of biomedical applications. In an attempt to determine the optimum conditions for hydrogel synthesis by the free-radical polymerization of sorbitan methacrylate (SMA), the hydrogel used in this study was well polymerized under the following conditions: 50% (w/v) SMA as monomer, 1% (w/w) alpha, alpha'-azo-bis(isobutyro-nitrile) as thermal initiator, and 1% (w/w) ethylene glycol dimethacrylate as cross-liking agent. Under these conditions, the moisture content of the polymerized SMA hydrogel was higher than in the other conditions. Moreover, the moisture content of the poly(SMA) hydrogel was also found to be higher than that of the poly(methyl methacrylate [MMA]) hydrogel. When the Fourier transform-infrared spectrum of poly(SMA) hydrogel was compared with that of poly(MMA) hydrogel, we noted a band at 1735-1730/cm, which did not appear in the Fourier transform-infrared spectrum of poly(MMA). The surface of the poly(SMA) hydrogel was visualized through scanning electron microscopy, and was uniform and clear in appearance.  相似文献

Liquid crystal displays had a humble beginning with wrist watches in the seventies. Continued research and development in this multi-disciplinary field have resulted in displays with increased size and complexity. After three decades of growth in performance, LCDs now offer a formidable challenge to the cathode ray tubes (CRT). A major contribution to the growth of LCD technology has come from the developments in addressing techniques used for driving matrix LCDs. There are several approaches like passive matrix addressing, active matrix addressing and plasma addressing to drive a matrix display. Passive matrix LCD has a simple construction and uses the intrinsic non-linear characteristic of the LCD for driving. Departure from conventional line by line addressing of a passive matrix has resulted in improved performance of the display. Orthogonal functions have played a crucial role in the development of passive matrix addressing. Simple orthogonal functions that are useful for driving a matrix LCD are introduced. The basics of driving several rows simultaneously (multi-line addressing) are discussed by drawing analogies from multiplexing in communication. The impact of multi-line addressing techniques on the performance of the passive matrix LCDs in comparison with the conventional technique will be discussed.  相似文献

Characterization of Polyurethane Rubber at High Deformation Rates   总被引:1,自引:0,他引:1  
Polyurethane rubber materials have widespread usage in large-deformation energy absorption and dissipation applications. Accurate design modeling with these materials requires an appropriate constitutive material model that accounts for both static (low strain rate) and dynamic (high strain rate) responses. A common modeling approach is the use of hyper-viscoelastic formulations, which couple quasi-static hyperelastic with dynamic viscoelastic responses and describe the material response over a range of deformation rates. In this work the effectiveness of two models, the Modified Quasi-Linear Viscoelastic and Non-Linear Hyper-Viscoelastic, are investigated to describe the high-rate behaviour of two different grades of polyurethane rubber. From quasi-static, uniaxial compression tests, a Rivlin hyperelastic formulation was found to describe the low-rate response well. High-rate, uniaxial compressions test were performed using a Polymeric Split Hopkinson Pressure Bar (PSHPB), supported by high-speed photography. In general, it was found that the Modified Quasi-Linear Viscoelastic model did not fit the experimental data well due to its limited non-linear terms, while the Non-Linear Hyper-Viscoelastic provided very good agreement.  相似文献

A supramolecular strategy for detecting specific proteins in complex media by using hyperpolarized 129Xe NMR is reported. A cucurbit[6]uril (CB[6])‐based molecular relay was programmed for three sequential equilibrium conditions by designing a two‐faced guest (TFG) that initially binds CB[6] and blocks the CB[6]–Xe interaction. The protein analyte recruits the TFG and frees CB[6] for Xe binding. TFGs containing CB[6]‐ and carbonic anhydrase II (CAII)‐binding domains were synthesized in one or two steps. X‐ray crystallography confirmed TFG binding to Zn2+ in the deep CAII active‐site cleft, which precludes simultaneous CB[6] binding. The molecular relay was reprogrammed to detect avidin by using a different TFG. Finally, Xe binding by CB[6] was detected in buffer and in E. coli cultures expressing CAII through ultrasensitive 129Xe NMR spectroscopy.  相似文献
