Synthesis of mesoionic[1,2,3]triazolo[5,1-d][1,2,5]triazepines

Intramolecular cyclization of 1-amino-3-phenacyl-4-carbohydrazido-1,2,3-triazolium-5-olates has been shown to take place via selective interaction of the carbonyl group with the terminal amino function of the hydrazido group to form a 1,2,5-triazepine ring. Minor products, resulting from the interaction of the α-nitrogen atom of the hydrazido group with the carbonyl function, having a N-amino-pyridazine structure were also detected and isolated. A general method for the synthesis of novel mesoionic 2-amino-7-aryl-4-oxo-2,4,5,8-tetrahydro[1,2,3]triazolo[5,1-d][1,2,5]triazepin-9-ium-3-olates was developed.     

High-speed microwave-promoted hetero-Diels-Alder reactions of 2(1H)-pyrazinones in ionic liquid doped solvents

Inter- and intramolecular hetero-Diels-Alder reactions in a series of functionalized 2(1H)-pyrazinones were investigated under controlled microwave irradiation. The cycloaddition reactions were efficiently performed in sealed tubes, utilizing either a combination of 1,2-dichloroethane and a thermally stable ionic liquid, or 1,2-dichlorobenzene as reaction medium. In all cases, a significant rate-enhancement using microwave flash heating as compared to thermal heating was observed.     

A microwave-assisted click chemistry synthesis of 1,4-disubstituted 1,2,3-triazoles via a copper(I)-catalyzed three-component reaction

A microwave-assisted three-component reaction was used to prepare a series of 1,4-disubstituted-1,2,3-triazoles from corresponding alkyl halides, sodium azide, and alkynes. This procedure eliminates the need to handle organic azides, as they are generated in situ, making this already powerful click process even more user-friendly and safe.     

Synthesis of novel dendrimers containing pyrimidine units

A novel convergent approach to dendritic macromolecules is described in which 4,6-dichloro-2-(4-methoxyphenyl)-pyrimidine is used as the building block. The nucleophilic aromatic substitution reaction at this AB₂-monomer was used as the key step in the propagation of the dendrons. Different core reagents were used to form the dendrimers, including a 5,15-bis(pyrimidyl)porphyrin core. Fourth-generation dendrons and third-generation dendrimers could be synthesized. The presented dendrimers are promising candidates to be used in applications where a more rigid structure and a larger resistance towards the applied conditions is required.     

Influence of the coordination number of nitrogen on the structure of 6aλ4-thia-1,3,4,6-tetraazapentalenic systems. Crystal structure analyses of 3-(2-pyridylimino)-3H-[1,2,4]thiadiazolo[4,3-a]pyridine and its 1-methylated salt

Structural data were obtained by X-ray crystallography for the title compounds which show that they are essentially planar and exhibit an approximately linear N2-S1-N8 arrangement. In compound 3 the separation between the sulfur atom and the pyridine nitrogen atom (2.61 Å) is larger than the Huggins constant energy distance (2.58 Å), suggesting that there is little or no bonding between them. The methylated salt 4 , on the contrary, has a closer S…N(pyridine) distance (2.19 Å) with an estimated bond dissociation energy of 6 kcal/mole.     

The regioselectivity of the formation of 2-pyrazolylthiazoles and their precursors from the reaction of 2-hydrazinothiazoles with 4,4,4-trifluoro-1-hetaryl-1,3-butanediones

Reaction of 2-hydrazinothiazoles 1 with 1-thienyl- and 1-furyl-1,3-butanediones 2a,b in methanol in the presence of hydrochloric acid mainly leads to a mixture of pyrazoles 3 and pyrazolines 4 or pyrazoles 3 and 5 in strong acidic conditions. Isomeric hydrazones 6 and pyrazolines 4 were formed and isolated in these reactions in the absence of hydrochloric acid. It has been shown that the regioselectivity in the reaction of diketones 2 with hydrazine 1 is governed by both the concentration of acid and the nature of substituents in the 1,3-diketones 2. Cyclization of hydrazones 6 is shown to occur under milder conditions than dehydration for pyrazolines 4. The new heterocyclic compounds were prepared and fully characterized by NMR spectra and by X-ray analysis for 3c.     

1,2,3-Thiadiazole derivatives with a nearly linear N-S…O grouping. X-ray crystal structure analysis of four methylated products of 4-phenyl-1,2,3-thiadiazole-5-carbaldoxime

The title oxime 6 was methylated under different conditions and yielded four monomethylated products 7-10 and two bismethylated products 11 and 12 which were easily distinguished by their ¹³C nmr spectra. In view of the potential thiapentalene character of 8, 9, 10 and 11 , their X-ray crystal structures were determined. The structural properties of the nitroso compound 9 are in accordance with a thiapentalene structure, whereas those of the other compounds deviate in the order 10 < 11 < < 8 .     

The application of "click chemistry" for the decoration of 2(1H)-pyrazinone scaffold: generation of templates

The "click chemistry" approach has been explored on the 2-(1H)-pyrazinone scaffold for the generation of pharmacologically interesting heterocyclic moieties. Huisgen 1,3-dipolar cycloaddition has been evaluated as the key step for the construction of the 1,2,3-triazole ring at the C-3 position of 2-(1H)-pyrazinones. Two different pathways have been successfully evaluated: (1) via C-C or C-O linkage of the acetylenic part to the C-3 position of the 2-(1H)-pyrazinone scaffold or (2) via azide introduction in the C-3 position. The subsequent application of "click chemistry" resulted in the formation of hitherto unknown skeletons. Microwave irradiation has successfully been applied in different steps of the sequence.     

N-Confused Calix

Next to the "normal" calix[4]pyrrole 1, the N-confused calix[4]pyrrole 2 is formed in substantial amounts (up to 22 % yield) as side product in the acid-catalyzed condensation reaction of ketones and pyrrole. In some cases, doubly N-confused calix[4]pyrroles are also formed.
