The two new diarylheptanoids, cymodienol (1), and cymodiene (2), obtained from specimens of the sea grass Cymodocea nodosa, collected from the coastal areas of central Greece, are the first members of this class isolated from marine organisms. The chemical structures of the two metabolites were assigned on the basis of their NMR and MS spectroscopic data, including information obtained by 1D- and 2D-NMR experiments. Metabolite 2 possesses an unprecedented skeleton that might be biosynthetically related to metabolite 1. Cymodienol (1) was found to exhibit significant cytotoxic activity against two lung cancer cell lines. 相似文献

Hypoxia is an important contributing factor to the development of drug‐resistant cancer, yet few nonperturbative tools exist for studying oxygenation in tissues. While progress has been made in the development of chemical probes for optical oxygen mapping, penetration of such molecules into poorly perfused or avascular tumor regions remains problematic. A click‐assembled oxygen‐sensing (CAOS) nanoconjugate is reported and its properties demonstrated in an in vitro 3D spheroid cancer model. The synthesis relies on the sequential click‐based ligation of poly(amidoamine)‐like subunits for rapid assembly. Near‐infrared confocal phosphorescence microscopy was used to demonstrate the ability of the CAOS nanoconjugates to penetrate hundreds of micrometers into spheroids within hours and to show their sensitivity to oxygen changes throughout the nodule. This proof‐of‐concept study demonstrates a modular approach that is readily extensible to a wide variety of oxygen and cellular sensors for depth‐resolved imaging in tissue and tissue models. 相似文献

Several novel squalamine-related polyaminosterols are reported. The synthesis of 7α-N-[3N-(4-aminobutyl)aminopropyl]aminocholestanol I, 6α-N-[3N-(4-aminobutyl)aminopropyl]aminocholestanol II, 7α and 7β-N-[3N-(4-aminobutyl)aminopropyl]aminocholesterol (III and IV), was accomplished from cholesterol, they provide the first examples in which spermidine is introduced in the B steroidal ring. These molecules showed comparable antibacteria and fungi activities to squalamine, and were cytotoxic on a human non-small cell bronchopulmonary carcinoma line (NSCLC-N6). Therefore, these molecules with antibiotic and cytotoxic activities are promising for immune-compromised patients in cancer chemotherapy. 相似文献

A series of chiral aryl alkyl carbinols, dissolved in MBBA and other nematic solvents, induce cholesteric structures. The handedness of the induced mesophases and the twisting power of the dopant alcohols are studied by means of CD and Grandjean-Cano microscopic techniques. The investigation points out the possibility of obtaining information on the stereochemistry of the dopant by studying the characteristics of the induced helices.Two different types of H- bonds between alcohols and MBBA are discussed in connection with a possible model of induction. 相似文献

Oxygen monitoring has been a topic of exhaustive study given its central role in the biochemistry of life. The ability to quantify the physiological distribution and real‐time dynamics of oxygen from sub‐cellular to macroscopic levels is required to fully understand the mechanisms associated with both normal physiology and disease states. This Review will present the most significant recent advances in the development of oxygen‐sensing materials and techniques, including polarographic, nuclear medicine, magnetic resonance, and optical approaches, that can be applied specifically for the real‐time monitoring of oxygen dynamics in cellular and tissue environments. As some of the most exciting recent advances in synthetic methods and biomedical applications have been in the field of optical oxygen sensors, a major focus will be on the development of these toolkits. 相似文献

Three new pentacyclic triterpenoids: (20R)-3-oxolupan-30-al (1), (20S)-3-oxolupan-30-al (2) and (20R)-28-hydroxylupen-30-al-3-one (3), along with (20S)-3beta-hydroxylupan-30-al (4), the latter previously described as a constituent of an epimeric mixture, were isolated from Acacia mellifera. In addition, the known metabolites 30-hydroxylup-20-(29)-en-3-one (5), 30-hydroxylup-20-(29)-en-3beta-ol (6), atranorin, methyl 2,4-dihydroxy-3,6 dimethyl benzoate, sitosterol-3beta-O-glucoside and linoleic acid were found in the analyzed plant species for the first time. The structures of the new metabolites were elucidated by extensive spectroscopic analyses and their relative stereochemistry was determined by NOESY experiments. The new metabolite 3 exhibited significant cytotoxic activity against the NSCLC-N6 cell line, derived from a human non-small-cell bronchopulmonary carcinoma. 相似文献

The first synthesis of the naturally occurring cyclic peptide axinellin A has been achieved. Cyclization and subsequent deprotection of linear precursors containing either a t-butyl protected Thr residue or a Thr(ΨMe,Mepro) derivative gave a cyclic peptide, identical in all respects to the naturally occurring material, with the exception that the synthetic peptide does not exhibit the cytotoxic activity reported for the natural product. 相似文献

Three new brominated diterpenes (1, 2, 8), along with six previously reported metabolites (3-7, 9), were isolated from the organic extract of Sphaerococcus coronopifolius, collected in Palaiokastritsa bay at the west coasts of Corfu Island. The structures of the new natural products, as well as their relative stereochemistry, were established by means of spectral data analyses, including 2D experiments. The absolute stereochemistry of 2 and 4 as well as the structure revision of the previously reported metabolite 3 were established by X-ray crystallographic analyses. The cytotoxicity of the isolated metabolites was evaluated against the NSCLC-N6-L16 and A549 human lung cancer cell lines. 相似文献
