Thermodynamic study of transthyretin association (wild‐type and senile forms) with heparan sulfate proteoglycan: pH effect and implication of the reactive histidine residue

The tetramer destabilization of transthyretin into monomers and its fibrillation are phenomena leading to amyloid deposition. Heparan sulfate proteoglycan (HSPG) has been found in all amyloid deposits. A chromatographic approach was developed to compare binding parameters between wild‐type transthyretin (wtTTR) and an amyloidogenic transthyretin (sTTR). Results showed a greater affinity of sTTR for HSPG at pH 7.4 compared with wtTTR owing to the monomeric form of sTTR. Analysis of the thermodynamic parameters showed that van der Waals interactions were involved at the complex interface for both transthyretin forms. For sTTR, results from the plot representing the number of protons exchanged vs pH showed that the binding mechanism was pH‐dependent with a critical value at a pH 6.5. This observation was due to the protonation of a histidine residue as an imidazolium cation, which was not accessible when TTR was in its tetrameric structure. At pH >6.5, dehydration at the binding interface and several contacts between nonpolar groups of sTTR and HSPG were also coupled to binding for an optimal hydrogen‐bond network. At pH <6.5, the protonation of the His residue from sTTR monomer when pH decreased broke the hydrogen‐bond network, leading to its destabilization and thus producing slight conformational changes in the sTTR monomer structure. Numerical methods for finding multiple eigenvalues of matrices depending on parameters

Summary. Let be a square matrix dependent on parameters and , of which we choose as the eigenvalue parameter. Many computational problems are equivalent to finding a point such that has a multiple eigenvalue at . An incomplete decomposition of a matrix dependent on several parameters is proposed. Based on the developed theory two new algorithms are presented for computing multiple eigenvalues of with geometric multiplicity . A third algorithm is designed for the computation of multiple eigenvalues with geometric multiplicity but which also appears to have local quadratic convergence to semi-simple eigenvalues. Convergence analyses of these methods are given. Several numerical examples are presented which illustrate the behaviour and applications of our methods. A versatile protocol for the preparation of substituted 1- and 2-naphthyl piperazines from aminonaphthols

Aminonaphthols are easily transformed into a variety of 1- and 2-naphthyl piperazines using a sequence of diazotization, iodide substitution and Pd(0) catalyzed coupling reactions.     

Local intersections of plane algebraic curves

We determine the maximum punctual order of contact between two plane algebraic curves, of which one is reduced. We prove that, generically on the reduced curve, this quantity is always strictly bounded by the product of the degrees of the curves.

     

Doppler cooling a single Ca+ ion with a violet extended-cavity diode laser

We present a scheme for employing a violet extended-cavity diode laser in experiments with single, trapped ions. For this the grating-stabilised laser is spatially and spectrally filtered and referenced to a Fabry–Pérot cavity. We measure an upper limit to the line width by observing a 305-kHz FWHM beat note with the second harmonic of a titanium sapphire laser. The laser is subsequently used to optically cool a single ⁴⁰Ca⁺ ion close to the Doppler limit. PACS 03.67.Lx; 32.80.Pj; 42.55.Px     

Characterization of supercommuting matrices

For a given n-by-n matrix A, we consider the set of matrices which commute with A and all of whose principal submatrices commute with the corresponding principal submatrices of A. The properties of this set are examined, with particular attention to its dimension.     

RECONSTRUCTION OF BINARY RELATIONS FROM THEIR RESTRICTIONS OF CARDINALITY 2, 3, 4 and (n - 1) II

We shall prove here that any binary relation on a base E with cardinality n > 6 is reconstructible from its restrictions of cardinality 2, 3, 4 and (n - 1). This proof needs results of part I of this paper where we characterize any pair of relations R, R' which are 2-, 3- and 4-hypomorphic. As a corollary we obtain that any binary relation is (n - 4)-reconstructible (when n > 9).     

SELECTIVE DEGRADATION OF AMINO ACIDS PHOTOSENSITIZED BY TRYPTOPHAN IN POLYPEPTIDE STRUCTURES

Polypeptides containing a basic amino acid close to their single tryptophan residue were irradiated with monochromatic 302 nm radiation. Tryptophan photolysis was monitored by absorption and fluorescence spectroscopy. Amino acid loss was evaluated by amino acid analysis. Only the protonated residues adjacent to tryptophan in the sequence were destroyed upon tryptophan excitation. This reaction is probably due essentially to direct interaction between the excited tryptophan and the neighbouring residue without electron solvation.
