Synthesis of fused triazolothienopyrimidine derivatives

Reactions of substituted 1-(thieno[2,3-d]pyrimidin-2-yl)-4-arylthiosemicarbazides with methyl iodide led to the formation of fused triazolothienopyrimidine derivatives.     

X-ray crystallographic investigation of the adduct of 1-demethylamino-2-(4-bromophenyl-2H-isoindole with N-phenylmalemide

Cyclocondensation of [1-Cyano-2-(methylsulfanyl)vinyl]triphenylphosphonium Iodide with Amidine Nucleophiles

The substituted vinylphosphonium salt (E)-MeSCH=C(CN)P⁺Ph₃I⁻ readily cyclizes under the action of benzamidine and 3-amino-s-triazole, but it does not enter cyclocondensation with 2-aminopyridine. The structure of the cyclization product with 3-amino-s-triazole was confirmed by its transformation to 7-imino-6-(triphenylphosphoranylidene)-6,7-dihydro-s-triazolo[1,5]pyrimidine which was identified by X-ray diffraction. Synthesis of functionalized m-bistrifluoromethylbenzenes via cyclocondensation of 1,1,1,5,5,5-hexafluoroacetylacetone with enamines

The reaction of 1,1,1,5,5,5-hexafluoroacetylacetone with push-pull enamines having a methyl group at the α-position was investigated. It was found that the reaction is sensitive both to the structure of enamines and to reaction conditions. As a result, a set of bistrifluoromethyldialkylanilines and ethyl bistrifluoromethylsalicylate was prepared. Plausible mechanisms and factors influencing the course of the reaction are discussed.     

Synthesis of 4-Benzylsulfonyl-5-polyfluoroalkyl-ν-triazoles from 1,1-Dihydropolyfluoroalkyl Benzyl Sulfones

The reaction of 1,1-dihydropolyfluoroalkyl benzyl sulfones with sodium azide or trimethylsilyl azide in the presence of tertiary amines gives ammonium salts of 4-benzylsulfonyl-5-polyfluoroalkyl--triazoles. The proposed reaction scheme includes the participation of catalytic amounts of azide ion in the cyclization of the formed intermediate vinyl azides to triazoles. X-ray analysis of 1,4-diazabicyclo[2.2.2]octane salt of 4-benzylsulfonyl-5-(-H-hexafluoropropyl)--triazole has been carried out.     

Synthesis and Heterocyclization of 3-Arylaminothiocrotonanilides

Reactions of acetylthioacetanilide with arylamines in acetic acid in the presence of sodium acetate give 3-arylaminothiocrotonanilides in good yields. When treated with -bromoacetophenone in acetone, these products are converted to substituted 4-hydroxy-²-thiazolinium bromides, one of which was dehydrated to obtain the corresponding thiazolium bromide. The structure of the heterocyclization products was confirmed by single crystal X-ray diffraction and NMR study of 2-acetonylidene-3,4-diphenyl-2,3-dihydrothiazole formed by dehydration of the corresponding ²-thiazolinium salt with simultaneous hydrolysis.     

A new stereoselective synthesis of 2-amino-4,5-dihydrothiophene-3-carbonitrile derivatives

A new stereoselective method for the synthesis of trans-isomers of 2-amino-4-aryl-5-benzoyl-4,5-dihydrothiophene-3-carbonitriles was proposed. The method involves base-catalyzed reactions of phenacyl thiocyanate with 3-(het)aryl-2-cyanoprop-2-enethioamides. (4R,5S/4S,5R)-2-Amino-5-benzoyl-4-(2-chlorophenyl)-4,5-ihydrothiophene-3-carbonitrile was structurally characterized by X-ray diffraction analysis. Published in Russian in Izvestiya Akademii Nauk. Propanal in the Syntheses of Alkyl-substituted 3-Cyano-2-piperidone, 3-Cyano-2,5,6,7-tetrahydropyrindin-2(1H)-one, and 3-Cyanopyridine-2(1H)-thione

Condensation of propanal with cyanothioacetamide and morpholine gave 3-cyano-4-ethyl-5-methyl-6-morpholino-2-piperidone, the structure of which was studied by X-ray analysis. Reaction of propanal with cyanothioacetamides and cycloalkanone enamines gave 3-cyano-4-ethyl-2,5,6,7-tetrahydropyrindin-2(1H)-one and 3-cyano-4-ethyl-5,6-hexamethylenepyridine-2(1H)-thione. The latter was used for the preparation of substituted 2-benzyloxycarbonylmethylthiopyridine and 3-amino-2-benzyloxycarbonylthieno[2,3-b]pyridine. __________ Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 7, pp. 1053–1059, July, 2005.
