Rhodium-catalyzed hydrogenation of 1,3-enynes 1a-8a and 1,3-diynes 9a-13a at ambient temperature and pressure in the presence of ethyl (N-tert-butanesulfinyl)iminoacetate and ethyl (N-2,4,6-triisopropylbenzenesulfinyl)iminoacetates, respectively, results in reductive coupling to afford unsaturated alpha-amino acid esters 1b-13b in good to excellent yields with exceptional levels of regio- and stereocontrol. Further hydrogenation of the diene containing alpha-amino acid esters 1b-8b using Wilkinson's catalyst at ambient temperature and pressure results in regioselective reduction to afford the beta,gamma-unsaturated alpha-amino acid esters 1c-8c in good to excellent yields. Exhaustive hydrogenation of the unsaturated side chains of the Boc- and Fmoc-protected derivatives of enyne and diyne coupling products 14b-16b occurs in excellent yield using Crabtree's catalyst at ambient temperature and pressure providing the alpha-amino acid esters 14d-16d, which possess saturated side chains. Finally, cross-metathesis of the Boc-protected reductive coupling product 14b with cis-1,4-diacetoxy-2-butene proceeds readily to afford the allylic acetate 14e. Isotopic labeling studies that involve reductive coupling of enyne 1a and diyne 9a under an atmosphere of elemental deuterium corroborate a catalytic mechanism in which oxidative coupling of the alkyne and imine residues is followed by hydrogenolytic cleavage of the resulting metallacycle. A stereochemical model accounting for the observed sense of asymmetric induction is provided. These studies represent the first use of imines as electrophilic partners in hydrogen-mediated reductive carbon-carbon bond formation.  相似文献

The BubR1 mitotic-checkpoint protein monitors proper attachment of microtubules to kinetochores, and links regulation of chromosome-spindle attachment to mitotic-checkpoint signaling. Thus, disruption of BubR1 activity results in a loss of checkpoint control, chromosomal instability caused by a premature anaphase, and/or the early onset of tumorigenesis. The mechanisms by which deregulation and/or abnormalities of BubR1 expression operate, however, remain to be elucidated. In this study, we demonstrate that levels of BubR1 expression are significantly increased by demethylation. Bisulfite sequencing analysis revealed that the methylation status of two CpG sites in the essential BubR1 promoter appear to be associated with BubR1 expression levels. Associations of MBD2 and HDAC1 with the BubR1 promoter were significantly relieved by addition of 5-aza-2'-deoxycytidine, an irreversible DNA methyltransferase inhibitor. However, genomic DNA isolated from 31 patients with colorectal carcinomas exhibited a +84A/G polymorphic change in approximately 60% of patients, but this polymorphism had no effect on promoter activity. Our findings indicate that differential regulation of BubR1 expression is associated with changes in BubR1 promoter hypermethylation patterns, but not with promoter polymorphisms, thus providing a novel insight into the molecular regulation of BubR1 expression in human cancer cells.  相似文献

This paper proposes an alternative robust adaptive high-gain fuzzy observer design scheme and its application to synchronization and secure communication of chaotic systems. It is assumed that their states are immeasurable and their parameters are unknown. The structure of the proposed observer is represented by Takagi–Sugeno fuzzy model and has the integrator of the estimation error. It improves the performance of high-gain observer and makes the proposed observer robust against noisy measurements, uncertainties and parameter perturbations as well. Using Lyapunov stability theory, an adaptive law is derived to estimate the unknown parameters and the stability of the proposed observer is analyzed. Some simulation result of synchronization and secure communication of chaotic systems is given to present the validity of theoretical derivations and the performance of the proposed observer as an application.  相似文献

2.2.2-Cryptand(1+) salts of the [Sb(2)Se(4)](2)(-), [As(2)S(4)](2)(-), [As(10)S(3)](2)(-), and [As(4)Se(6)](2)(-) anions have been synthesized from the reduction of binary chalcogenide compounds by K in NH(3)(l) in the presence of the alkali-metal-encapsulating ligand 2.2.2-cryptand (4,7,13,16,21,24-hexaoxa-1,10-diazabicyclo[8.8.8]hexacosane), followed by recrystallization from CH(3)CN. The [Sb(2)Se(4)](2)(-) anion, which has crystallographically imposed symmetry 2, consists of two discrete edge-sharing SbSe(3) pyramids with terminal Se atoms cis to each other. The Sb-Se(t) bond distance is 2.443(1) ?, whereas the Sb-Se(b) distance is 2.615(1) ? (t = terminal; b = bridge). The Se(b)-Sb-Se(t) angles range from 104.78(4) to 105.18(5) degrees, whereas the Se(b)-Sb-Se(b) angles are 88.09(4) and 88.99(4) degrees. The (77)Se NMR data for this anion in solution are consistent with its X-ray structure (delta 337 and 124 ppm, 1:1 intensity, -30 degrees C, CH(3)CN/CD(3)CN). Similar to this [Sb(2)Se(4)](2)(-) anion, the [As(2)S(4)](2)(-) anion consists of two discrete edge-sharing AsS(3) pyramidal units. The As-S(t) bond distances are 2.136(7) and 2.120(7) ?, whereas the As-S(b) distances range from 2.306(7) to 2.325(7) ?. The S(b)-As-S(t) angles range from 106.2(3) to 108.2(3) degrees, and the S(b)-As-S(b) angles are 88.3(2) and 88.9(2) degrees. The [As(10)S(3)](2)(-) anion has an 11-atom As(10)S center composed of six five-membered edge-sharing rings. One of the three waist positions is occupied by a S atom, and the other two waist positions feature As atoms with exocyclic S atoms attached, making each As atom in the structure three-coordinate. The As-As bond distances range from 2.388(3) to 2.474(3) ?. The As-S(t) bond distances are 2.181(5) and 2.175(4) ?, and the As-S(b) bond distance is 2.284(6) ?. The [As(4)Se(6)](2)(-) anion features two AsSe(3) units joined by Se-Se bonds with the two exocyclic Se atoms trans to each other. The average As-Se(t) bond distance is 2.273(2) ?, whereas the As-Se(b) bond distances range from 2.357(3) to 2.462(2) ?. The Se(b)-As-Se(t) angles range from 101.52(8) to 105.95(9) degrees, and the Se(b)-As-Se(b) angles range from 91.82(7) to 102.97(9) degrees. The (77)Se NMR data for this anion in solution are consistent with its X-ray structure (delta 564 and 317 ppm, 3:1 intensity, 25 degrees C, DMF/CD(3)CN).  相似文献

Psoriasis is one of the most common immune-mediated chronic inflammatory skin diseases. However, little is known about the molecular mechanism underlying the immunological circuits that maintain innate and adaptive immune responses in established psoriasis. In this study, we found that the Pellino1 (Peli1) ubiquitin E3 ligase is activated by innate pattern-recognition receptors (PRRs), such as Toll-like receptors (TLRs), and is highly upregulated in human psoriatic skin lesions and murine psoriasis-like models. Increased Peli1 expression is strongly correlated with the immunopathogenesis of psoriasis by activating hyperproliferation of keratinocytes in the S and G2/M phases of the cell cycle and promoting chronic skin inflammation. Furthermore, Peli1-induced psoriasis-like lesions showed significant changes in the expression levels of several T helper 17 (Th17)-related cytokines, such as IL-17a, IL-21, IL-22, IL-23, and IL-24, indicating that overexpression of Peli1 resulted in the sequential engagement of the Th17 cell response. However, the overexpression of Peli1 in T cells was insufficient to trigger psoriasis, while T cells were indispensable for disease manifestation. In summary, our findings demonstrate that Peli1 is a critical cell cycle activator of innate immunity, which subsequently links Th17 cell immune responses to the psoriatic microenvironment.Subject terms: Chronic inflammation, Immunoproliferative disorders  相似文献

The effect of excess surfactants (oleic acids) in a colloidal solution on the adsorption behavior of 9.5-nm-sized, sterically stabilized iron oxide (γ-Fe2O3) nanoparticles on hydrogen terminated Si (Si:H) substrates during a dip-coating process is examined. While the particle coverage follows a type of Langmuir adsorption isotherm as initially increasing and subsequently saturating with increasing particle concentration, it also critically depends on the excess surfactant concentration in the solution. For instance, it is noted that by adding the oleic acids from 0.06 to 2.80 × 1018 ml−1 in the solution with 4.65 × 1013 ml−1 particle concentration, the coverage is gradually reduced from 0.42 to 0.25. In addition, increasing surfactant concentration distinctly changes the morphology of a self-assembled particle layer from densely distributed smaller clusters to sparsely connected, larger ones with enlarged space. The reduced coverage and enlarged cluster size with increasing oleic acid concentration are explained by the reduced interaction energy between particle and substrate and the increased capillary force between particles.  相似文献

In this paper, we study stochastic aggregation properties of the financial model for the N‐asset price process whose dynamics is modeled by the weakly geometric Brownian motions with stochastic drifts. For the temporal evolution of stochastic components of drift coefficients, we employ a stochastic first‐order Cucker‐Smale model with additive noises. The asset price processes are weakly interacting via the stochastic components of drift coefficients. For the aggregation estimates, we use the macro‐micro decomposition of the fluctuations around the average process and show that the fluctuations around the average value satisfies a practical aggregation estimate over a time‐independent symmetric network topology so that we can control the differences of drift coefficients by tuning the coupling strength. We provide numerical examples and compare them with our analytical results. We also discuss some financial implications of our proposed model.  相似文献
