The interaction of Rose Bengal (RB) with hydroxypropyl-α-cyclodextrin (HP-α-CD), hydroxypropyl-β-cyclodextrin (HP-β-CD) and hydroxypropyl-γ-cyclodextrin (HP-γ-CD) has been studied in water and in acetate buffer at pH 4.5 by UV–Vis absorption, fluorescence spectroscopy and Induced Circular Dichroism at 298 K. Evidence of the complex formation between the RB and all HP-CDs have been obtained both in water and in buffer. Binding constants and stoichiometry of RB/HP-CD complexes in water have been determined by applying the modified Benesi-Hildebrand equation to the fluorescence measurements.  相似文献

Photosystem II is a multisubunit membrane complex which performs the water oxidation process in the higher plants. Core dimers and monomers of photosystem II have been isolated from thylakoid membranes by sucrose density gradient centrifugation. Lipids extracted from different photosystem II-enriched fractions obtained from spinach thylakoids have been analysed by thin layer chromatography. Cardiolipin is enriched throughout the purification of photosystem II complexes; in particular dimers contained two times more cardiolipin than their monomeric counterparts.  相似文献

Equilibrium constants and standard molar enthalpies of reaction were determined by titration calorimetry for the reaction of 1-butanol with 2-hydroxypropyl-b-cyclodextrin (HP-b-CD) in aqueous solution at different concentrations of NaCl (0-1.9 M). The standard molar free energy and entropy changes associated to the complexation were calculated from the corresponding equilibrium constants, K, and standard enthalpies determined experimentally. In NaCl solutions the inclusion complexes ButOH/HP-b-CD are more stable than in water and their stability increases at increasing NaCl concentration; otherwise, the standard molar enthalpy associated to the formation of the complexes does not change with the increasing of salt concentration. The dependence of K on NaCl concentration were used to evaluate the number of water molecules displaced from the hydration shells of HP-b-CD and ButOH in forming complexes. T-2 and HT-2 toxins are mycotoxins produced by several Fusarium species that are commonly found in various cereal grains, including oats, barley, wheat and maize. Intake estimates indicate that the presence of these mycotoxins in the diet can be of concern for public health. In this work, the inclusion processes occurring between fluorescent anthracene-derivatives of T-2 and HT-2 toxins and different cyclodextrin (CD) molecules were investigated in aqueous solutions by means of UV–Vis absorption, fluorescence emission and dynamic light scattering. Binding constant values and chemico-physical parameters were calculated. It was found that β-CDs give stronger inclusion reactions with both T-2 and HT-2 derivatives, as stated by important emission intensity increments. Such interactions were found to be fundamentally enthalpy-driven. Among β-CDs, the effect of the methylation at hydroxyl groups was tested: as a result, the di-methyl form of β-CD was found to induce the best fluorescence intensity enhancements.  相似文献

The interactions of Rose Bengal (RB) with alpha-cyclodextrin (alpha-CD), hydroxypropyl-beta-cyclodextrin (HP-beta-CD), hydroxypropyl-gamma-cyclodextrin (HP-gamma-CD), heptakis(2,3,6-tri-O-methyll)-beta-cyclodextrin (TM-beta-CD) were studied in aqueous solutions of 0.1 M KClO(4) and 0.1 M LiClO(4) by vis absorption, fluorescence spectroscopy as well as electrochemical measurements at 298 K. The spectrophometric results indicate that RB is included in all beta- and gamma-CDs forming complexes with a stoichiometry 1:1 whose stability is slightly higher in KClO(4) than in LiClO(4) solutions. The complex stability constants determined for salt-containing CD solutions are lower than those for water solutions. The complexation of RB with beta- and gamma-CD and the differences between the complexes obtained in the presence of the two salts were confirmed by an electrochemical study.  相似文献

The interactions between chlorophyll a and two beta-cyclodextrins, that have the same cavity size but different substituents, were studied in aqueous solutions. These supramolecular host-guest complexes were examined by a combination of UV/vis absorption, circular dichroism, NMR, and steady-state and time-resolved fluorescence measurements. The results indicate that all cyclodextrins solubilize the pigment mainly in monomeric form in water. The pigment forms 1:1 complexes with the heptakis(2,3,6-tri-O-methyl)-beta-cyclodextrin and 1:2 complexes with the hydroxypropyl-beta-cyclodextrin. In such complexes the methyl groups of the cyclodextrin inner cavity are involved in the interaction with the pigment as evidenced by NMR measurements. We also measured the luminescence of singlet oxygen photosensitized by chlorophyll a in the inclusion complexes.  相似文献

In this work, we have studied the interactions between two different cyclodextrins (CDs) and chlorophyll a (Chl a) in the presence of electrolyte by means of absorption, fluorescence spectroscopy, circular dichroism and cyclic voltammetry. The results obtained indicate that the presence of both CDs gives rise to an increase of Chl a solubility in water. In particular, heptakis-(2,3,6-tri-O-methyl)-beta-cyclodextrin (TRIMEB) favours the dissolution of Chl a monomer in aqueous solution, whereas the presence of hydroxypropyl-beta-cyclodextrin (beta-HP-CD) promotes the pigment aggregation.  相似文献

Ochratoxin A (OTA) is a nephrotoxic mycotoxin produced by several fungal species, mainly Aspergillus ochraceus, A. carbonarius and Penicillium verrucosum. It contaminates many foodstuffs, particularly cereals and their derivatives, coffee, beer, wine and cocoa, and represents a serious health threat both to humans and animals. Spectroscopic properties of OTA solutions depend on the pH, solvent polarity and can be influenced by the presence of cyclodextrins (CDs). In this work, the effect of β-CD on spectroscopic properties of OTA in aqueous solutions has been investigated by means of absorption and steady-state fluorescence at different pHs (range 3.5–9.5). Binding constants of OTA/β-CD inclusion complexes have been determined by applying modified Benesi-Hildebrand equation. A 1:1 stoichiometry of OTA/β-CD complexes has been observed at all tested pHs.  相似文献

In this work studies on rapid inhibitory interactions between heavy metals and photosynthetic materials at different organization levels were carried out by optical assay techniques, investigating the possibility of applications in the heavy metal detection field. Spinach chloroplasts, thylakoids and Photosystem II proteins were employed as biotools in combination with colorimetric assays based on dichlorophenol indophenole (DCIP) photoreduction and on fluorescence emission techniques. It was found that copper and mercury demonstrated a strong and rapid photosynthetic activity inhibition, that varied from proteins to membranes, while other metals like nickel, cobalt and manganese produced only slight inhibition effects on all tested photosynthetic materials. By emission measurements, only copper was found to rapidly influence the photosynthetic material signals. These findings give interesting information about the rapid effects of heavy metals on isolated photosynthetic samples, and are in addition to the literature data concerning the effects of growth in heavy metal enriched media.  相似文献

The regeneration method of Khorana [J. Biol. Chem. 262 (1987) 9271] has been modified in order to study the effect of endogenous archaeabacterial lipids and, in particular, of glycocardiolipin (GlyC) in the refolding and chromophore regeneration of bacteriorhodopsin (BR). BR refolding and chromophore regeneration could be obtained in the presence of endogenous lipid mixtures containing or not containing glycocardiolipin; however, the kinetics of bacteriorhodopsin regeneration in the presence of glycocardiolipin was faster than in its absence. These results show for the first time that the interaction of glycocardiolipin with bacteriorhodopsin favours its refolding from the denaturated state and the chromophore regeneration.  相似文献
