Assessment of aptamer-steroid binding using stacking-enhanced capillary electrophoresis

The binding affinity of 17β-estradiol with an immobilized DNA aptamer was measured using capillary electrophoresis. Estradiol captured by the immobilized DNA was injected into the separation capillary using pH-mediated sample stacking. Stacked 17β-estradiol was then separated using micellar electrokinetic capillary chromatography and detected with UV-visible absorbance. Standard addition was used to quantify the concentration of estradiol bound to the aptamer. Following incubation with immobilized DNA, analysis of free and bound estradiol yielded a dissociation constant of 70 ± 10 μM. The method was also used to screen binding affinity of the aptamer for estrone and testosterone. This study demonstrates the effectiveness of capillary electrophoresis to assess the binding affinity of DNA aptamers.     

Multiple periodic solutions for a nonlinear suspension bridge equation

We investigate nonlinear oscillations in a fourth-order partialdifferential equation which models a suspension bridge. Previouswork establishes multiple periodic solutions when a parameterexceeds a certain eigenvalue. In this paper, we use Leray-Schauderdegree theory to prove that if the parameter is increased further,beyond a second eigenvalue, then additional solutions are created.     

Pervaporation-gas chromatography coupling for slurry samples. Determination of acetaldehyde and acetone in food

The use of pervaporation for the removal of volatile species from slurry samples, with a full automated introduction of the sample into the lower chamber of the pervaporation unit prior to their individual separation and determination by gas chromatography-flame ionisation detection, is presented for the first time. For this purpose, the upper chamber of the pervaporator is situated in the loop of an HPLC injection valve and the only requirement of the experimental setup for being used with slurries is to have adequately sized diameters for the units of the dynamic manifold assisting the donor chamber in order to avoid clogging by the suspended particles. The method developed was applied to the determination of acetaldehyde and acetone in food samples with different solids contents, such as yoghurt, Actimel and different kind of juices.     

Computerized data analysis of amino acids in physiologic fluids.

A Fortran IV computer program is described which identifies, computes, and statistically evaluates amino acid concentrations determined by an automatic amino acid analyzer in physiologic fluids. The program accepts retention time and intergrated peak areas from two calibration standards for identification and computational reference. Two internal standards are used to compensate for variations in injector performance and ninhydrin decay. Calibration standard responses are statistically treated to assist in detection of equipment malfunction. The statistical data base is automatically explanded through inclusion of data from normal patients. Amino acid concentrations are printed with appropriate mean and standard deviation values in a format acceptable as a final report.     

Experiments with longitudinally polarized electrons in a storage ring using a siberian snake

We report on first measurements with polarized electrons stored in a medium-energy ring and with a polarized internal target. Polarized electrons were injected at 442 MeV (653 MeV), and a partial (full) Siberian snake was employed to preserve the polarization. Longitudinal polarization at the interaction point and polarization lifetime of the stored electrons were determined with laser backscattering. Spin observables were measured for electrodisintegration of polarized 3He, with simultaneous detection of scattered electrons, protons, neutrons, deuterons, and 3He nuclei, over a large phase space.     

Phosphine-catalyzed Heine reaction

Aziridines are important synthetic intermediates which readily undergo ring-opening reactions. It is demonstrated that electron-rich phosphines are efficient catalysts for the regioselective rearrangement of N-acylaziridines to oxazolines. The reactions occur in excellent yield under neutral conditions. Evidence is provided for an addition/elimination mechanism by generation of a phosphonium intermediate. Similar intermediates may be useful for the development of alternate aziridine ring-opening processes and stereoselective synthesis with enantiopure phosphines.     

DNA vaccine containing the mycobacterial hsp65 gene prevented insulitis in MLD-STZ diabetes

Background  

Our group previously demonstrated that a DNA plasmid encoding the mycobacterial 65-kDa heat shock protein (DNA-HSP65) displayed prophylactic and therapeutic effect in a mice model for tuberculosis. This protection was attributed to induction of a strong cellular immunity against HSP65. As specific immunity to HSP60 family has been detected in arthritis, multiple sclerosis and diabetes, the vaccination procedure with DNA-HSP65 could induce a cross-reactive immune response that could trigger or worsen these autoimmune diseases.
