Neutron irradiation of Cp2MoCl2 for 24 h afforded the radiotracer Cp299MoCl2 which was characterised by UV–Vis spectroscopy and thin layer chromatography. Binding experiments with the thiol containing protein human serum albumin (HSA) or calf thymus DNA, were monitored for 99Mo using a gamma counter. Under the conditions investigated, molar ratios of binding of 0.2:1 (Cp2MoCl2:DNA) and 9.4:1 (Cp2MoCl2:HSA) were calculated. The results are consistent with in vitro coordination studies that have shown strong preferential interaction of Cp2MoCl2 with thiols versus other donor sites in biomolecules including DNA.  相似文献

We developed a new mammalian cell-based luciferase reporter gene assay for androgenic and antiandrogenic activities of chemicals and environmental samples. Environmental samples usually have a complex matrix that may contain the constituents acting as androgen receptor (AR) agonists, AR antagonists or aryl hydrocarbon receptor (AhR) agonists. AhR agonists are known to elicit the antiandrogenic effect through cross-talk between AR and AhR signal transduction pathways. In this study, PC3/AR human prostate carcinoma cells were transiently transfected with a prostate-specific antigen (PSA) promoter-driven luciferase expression plasmid. The cells were treated with a test compound or an environmental sample for 24 h at 37 degrees C and then measured for luciferase activity. The luciferase activity was induced by dihydrotestosterone (DHT) in a concentration-dependent manner in a concentration range from 10 fM to 1 nM. R1881, a synthetic androgen receptor agonist, induced luciferase activity and its inductive effects was additive to that of DHT. The luciferase activity was not induced by cortisol, a glucocorticoid, progesterone, a progestin, and 17beta-estradiol, an estrogen in a concentration range of up to 1 microM. DHT-induced luciferase activity was reduced by bicalutamide and cyproterone acetate, AR antagonists, and also by benzo[a]pyrene, an aryl hydrocarbon receptor agonist, through AhR-mediated pathways. All of these findings indicate that the present assay system correctly responds to AR agonists, AR antagonists and AhR agonist and, therefore, it is a powerful tool for the sensitive and selective screening of chemicals and environmental samples for their androgenic and antiandrogenic activities. We developed the first assay system, in which the expression of luciferase was driven by the promoter of a prostate-specific antigen gene, a typical human androgen-regulated gene.  相似文献

The magnetic structure and ordering temperatures of three intermetallic compounds which crystallize in the tetragonal ThCr2Si2 structure, TbCr2Si2, HoCr2Si2 and ErCr2Si2, have been determined by neutron diffraction, differential scanning calorimetry and magnetization measurements. The Cr-sublattice orders anti-ferromagnetically with Néel temperatures of 758 K for TbCr2Si2, 718 K for HoCr2Si2 and 692 K for ErCr2Si2. Chromium atoms located at 4d crystallographic sites are aligned anti-parallel along the c-axis, with GZCr magnetic modes. In contrast with metallic bcc Cr, the refined room temperature value of the ordered Cr moment is anomalously large for all three compounds. The effects of vanadium substitution on the iron magnetism in YFe12-x V x compounds (x~2–4) have been studied by57Fe Mössbauer spectroscopy. The preference of vanadium atoms for the 8i site of these ThMn12-structure type compounds is confirmed. The rapid collapse of the iron sublattice magnetization with increasing vanadium concentration is analysed in terms of the ‘magnetic valence’ model.  相似文献

Selective chemical reactions that are orthogonal to the diverse functionality of biological systems have become important tools in the field of chemical biology. Two notable examples are the Staudinger ligation of azides and phosphines and the Cu(I)-catalyzed [3 + 2] cycloaddition of azides and alkynes ("click chemistry"). The Staudinger ligation has sufficient biocompatibility for performance in living animals but suffers from phosphine oxidation and synthetic challenges. Click chemistry obviates the requirement of phosphines, but the Cu(I) catalyst is toxic to cells, thereby precluding in vivo applications. Here we present a strain-promoted [3 + 2] cycloaddition between cyclooctynes and azides that proceeds under physiological conditions without the need for a catalyst. The utility of the reaction was demonstrated by selective modification of biomolecules in vitro and on living cells, with no apparent toxicity.  相似文献

A study of the Nd-Fe-Ti phases at the Fe-rich end of the phase diagram has been carried out using57Fe Mössbauer spectroscopy, scanning electron microscopy (SEM) and energy dispersive X-ray analysis (EDAX). In particular,57Fe Mössbauer spectroscopy has been used to characterize the phases present in these samples. In addition to the two well-known 217 and 112 phases, twonovel phases have been identified, with compositions denoted by3–29 and9–94, which are closely linked to the orthorhombic ScFe6Ga6-type and the hexagonal TbCu7-type structures, respectively. The possible crystal structures of these new phases will be discussed in terms of the dumbbell substitution sequence in the CaCu5 structure.  相似文献

Nanotechnology promises to enhance the functionality and sensitivity of miniaturized analytical systems. For example, nanoscale transport systems, which are driven by molecular motors, permit the controlled movement of select cargo along predetermined paths. Such shuttle systems may enhance the detection efficiency of an analytical system or facilitate the controlled assembly of sophisticated nanostructures if transport can be coordinated through complex track networks. This study determines the feasibility of complex track networks using kinesin motor proteins to actively transport microtubule shuttles along micropatterned surfaces. In particular, we describe the performance of three basic structural motifs: (1) crossing junctions, (2) directional sorters, and (3) concentrators. We also designed track networks that successfully sort and collect microtubule shuttles, pointing the way towards lab-on-a-chip devices powered by active transport instead of pressure-driven or electroosmotic flow.  相似文献

The promotion energies, ionization potentials, electron affinities, and orbital electronegativities for the valence states of importance in atoms of the first two rows of the periodic system have been evaluated from analytical Hartree-Fock functions for the corresponding groundstates.
