We use agarose gel electrophoresis to characterize how the monovalent catioinic surfactant cetyltrimethylammonium bromide (CTAB) compacts double-stranded DNA, which is detected as a reduction in electrophoretic DNA velocity. The velocity reaches a plateau at a ratio R = 1.8 of CTAB to DNA-phosphate charges, i.e., above the neutralization point, and the complexes retain a net negative charge at least up to R = 200. Condensation experiments on a mixture of two DNA sizes show that the complexes formed contain only one condensed DNA molecule each. These CTAB-DNA globules were further characterized by time-resolved measurements of their velocity inside the gel, which showed that CTAB does not dissociate during the migration but possibly upon entry into the gel. Using the Ogston-model for electrophoresis of spherical particles, the measured in-gel velocity of the globule is quantitatively consistent with CTAB having two opposite effects, reduction of both the electrophoretic charge and DNA coil size. In the case of CTAB the two effects nearly cancel, which can explain why opposite velocity shifts (globule faster than uncomplexed DNA) have been observed with some catioinic condensation agents. Dissociation of the complexes by addition of anionic surfactants was also studied. The DNA release from the globule was complete at a mixing ratio between anionic and cationic surfactants equal to 1, in agreement with equilibrium studies. Circular DNA retained its supercoiling, and this demonstrates a lack of DNA nicking in the compaction-release cycle which is important in DNA transfection and purification applications.  相似文献

Using a family of higher degree polynomials as a bridge, together with complex surgery techniques, we construct a homeomorphism between any two limbs of the Mandelbrot set of equal denominator. Induced by these homeomorphisms and complex conjugation, we obtain an involution between each limb and itself, whose fixed points form a topological arc. All these maps have counterparts at the combinatorial level relating corresponding external arguments. Assuming local connectivity of the Mandelbrot set we may conclude that the constructed homeomorphisms between limbs are compatible with the embeddings of the limbs in the plane. As usual we plough in the dynamical planes and harvest in the parameter space.  相似文献

This work describes a synthetic approach where a non-planar aromatic heterocyclic [7]helicene is compressed to yield a hetero[8]circulene containing an inner antiaromatic cyclooctatetraene (COT) core. This [8]circulene consists of four benzene rings and four heterocyclic rings, and it is the first heterocyclic [8]circulene containing three different heteroatoms. The synthetic pathway proceeds via a the flattened dehydro-hetero[7]helicene, which is partially a helicene and partially a circulene: it is non-planar and helically chiral as helicenes, and contains a COT motif like [8]circulenes. The antiaromaticity of the COT core is confirmed by nucleus independent chemical shift (NICS) calculations. The planarization from a helically π-conjugated [7]helicene to a fully planar heterocyclic [8]circulene significantly alters the spectroscopic properties of the molecules. Post-functionalization of the [7]helicenes and the [8]circulenes by oxygenation of the thiophene rings to the corresponding thiophene-sulfones allows an almost complete fluorescence emission coverage of the visible region of the optical spectrum (400–700 nm).  相似文献

α‐Conotoxins are disulfide‐rich peptides that target nicotinic acetylcholine receptors. Recently we identified several α‐conotoxins that also modulate voltage‐gated calcium channels by acting as G protein‐coupled GABAB receptor (GABABR) agonists. These α‐conotoxins are promising drug leads for the treatment of chronic pain. To elucidate the diversity of α‐conotoxins that act through this mechanism, we synthesized and characterized a set of peptides with homology to α‐conotoxins known to inhibit high voltage‐activated calcium channels via GABABR activation. Remarkably, all disulfide isomers of the active α‐conotoxins Pu1.2 and Pn1.2, and the previously studied Vc1.1 showed similar levels of biological activity. Structure determination by NMR spectroscopy helped us identify a simplified biologically active eight residue peptide motif containing a single disulfide bond that is an excellent lead molecule for developing a new generation of analgesic peptide drugs.  相似文献

Lipid vesicles can be connected by membrane nanotubes to build networks with promising bioanalytical properties. Here we characterize electrophoretic transport in such membrane tubes, with a particular eye to how their soft-material nature influences the intratube migration. In the absence of field, the tube radius is 110 +/- 26 nm, and it remains in this range during electrophoresis even though the applied electric field causes a slight decrease in the tube radius (approximately 6-11%). The electrophoretic velocity of the membrane wall (labeled with quantum dots) varies linearly with the field strength. Intratube migration is studied with latex spheres of radii 15, 50, 100, and 250 nm. The largest particle size does not enter the tube at fields strengths lower than 1250 V/m because the energy cost for expanding the tube around the particles is too high. The smaller particles migrate with essentially the same velocity as the membrane at low fields. Above 250 V/cm, the 15 nm particles exhibit an upward deviation from linear behavior and in fact migrate faster than in free solution whereas the 100 nm particles deviate downward. We propose that these nonlinear effects arise because of lipid adsorption to the particles (dominating for 15 nm particles) and a pistonlike compression of the solvent in front of the particles (dominating for 100 nm). As expected from such complexities, existing theories for a sphere migrating in a rigid-wall cylinder cannot explain our velocity results in lipid nanotubes.  相似文献

Degradation of the residual strength of a glass-fiber-reinforced polyester composite of a lay-up typical of the wind rotor blade material is studied at low-cycle fatigue. A gradual reduction of the residual strength is observed as expected for GRP, accompanied by an increasing scatter of strength. The residual strength model based on the strength-life equal rank assumption yields an accurate approximation of experimental data. The strength reduction at a stress level corresponding to high-cycle fatigue (N>10 6 cycles) appears to correlate well with the test results at higher stress levels, which indicates that the strength degradation at the design stress level can be evaluated using low-cycle tests. Assuming that the parameters of the strength degradation model do not depend on the applied stress level, the residual strength data obtained in low stress level tests of comparatively short duration can be used to estimate the average fatigue life at the same stress thus reducing the total test time. Data on the ablative photodecomposition of three condensation polymers [polyimide, poly(ethylene terephthalate), and polycarbonate] and one addition polymer (polymethyl methacrylate) by laser pulses at 193 nm are presented. The etch depth/pulse is a linear function of the number of pulses at constant laser fluence. It varies with the logarithm of the fluence in a linear manner at different fluences. The etch depth is independent of the atmosphere above the film, whether it is air at 1 atm, or a vacuum. The etching of PMMA at fluences > 100 mJ/cm2 is believed to follow a mechanism different from the process at lower fluences. Etching of polyimide and polyethylene terephthalate at 248 and 308 nm is also reported. The mechanism of etching by laser radiation may receive greater contribution from a thermal process with increasing wavelength. This is manifested in the etch depth versus log fluence plot by sharp changes in slope.  相似文献

To evaluate the interaction between 17β-trenbolone (TB) and 17α-ethinylestradiol (EE2), male eelpout, Zoarces viviparus, was exposed for 21 days (April to May 2008) to 5 ng l−1 EE2 and 5 or 20 ng l−1 TB, separately or in combination in a flow-through SW system. The effects on hepatosomatic (HSI) and gonadosomatic index (GSI), plasma vitellogenin (Vtg) concentration, gonadal histology, hepatic and testicular Vtg mRNA and estrogen receptor (ERα) mRNA expression were investigated. No effects on HSI were observed. A significant decrease was observed in the GSI of all males exposed to EE2 (<0.7%) when compared to controls (1.4%). Histological alterations and immature stages were observed in the testis of all exposed males; however, males exposed to EE2 were the most affected. Increased tubule number and proportionally decreased tubule diameter were observed in the testis of all EE2 groups. No effects in Vtg mRNA expression were observed in the testis; however, a significant decrease in testis ERα mRNA was observed in males exposed to 20 ng l−1 TB. The groups exposed to EE2 showed a significant increase in plasma Vtg (>300-fold), hepatic Vtg mRNA (>450-fold), and ERα mRNA (>100-fold) when compared to controls. This study shows that lower concentrations of 17β-trenbolone are unable to counteract the EE2 estrogenic effects when the exposure is simultaneous.   相似文献
