Crystallization of Arthrobacter sp. strain 1C N-(1-D-carboxyethyl)-L-norvaline dehydrogenase and its complex with NAD+

The novel NAD+-linked opine dehydrogenase from a soil isolate Arthrobacter sp. strain 1C belongs to an enzyme superfamily whose members exhibit quite diverse substrate specificites. Crystals of this opine dehydrogenase, obtained in the presence or absence of co-factor and substrates, have been shown to diffract to beyond 1.8 ? resolution. X-ray precession photographs have established that the crystals belong to space group P21212, with cell parameters a = 104.9, b = 80.0, c = 45.5 ? and a single subunit in the asymmetric unit. The elucidation of the three-dimensional structure of this enzyme will provide a structural framework for this novel class of dehydrogenases to enable a comparison to be made with other enzyme families and also as the basis for mutagenesis experiments directed towards the production of natural and synthetic opine-type compounds containing two chiral centres.     

Palladium‐catalyzed amination with benzophenone imine as a new,safe and practical alternative to nitration for the synthesis of 7‐amino‐2,1,3‐benzothiadiazoles

Palladium‐catalyzed amination of 7‐bromo‐4‐methyl‐2,1,3‐benzothiadiazole ( 7 ) with benzophenone imine as an ammonia equivalent is described as a new, safe and practical alternative to nitration for the synthesis of 7‐amino‐4‐methyl‐2,1,3‐benzothiadiazole ( 1 ) in high yield. This methodology was successfully scaled‐up in the pilot plant on 14.0‐kg scale of 7 and was also utilized for the synthesis of 7‐amino‐4,6‐dimethyl‐2,1,3‐ben‐zothiadiazole ( 12 ) by the amination of 7‐bromo‐4,6‐dimethyl‐2,1,3‐benzothiadiazole ( 10 ).     

Stereoselective total syntheses of (±)-warburganal and (±)-isotadeonal

A short, stereoselective synthesis of (±)-warburganal ($\text{1}$), its C-9 epimer ($\text{2}$) and the related sesquiterpene dialdehyde (±)-isotadeonal is presented.     

Selective Benzylic Bromination of 2-Methylnaphthalene

A practical synthesis of 2-(bromomethyl)naphthalene (1), by selective benzylic bromination of 2-methylnaphthalene (2), with bromine in heptane in the presence of lanthanum acetate hydrate is described.     

Lithiation of N‐protected‐dihydro‐1,4‐benzoxaziness

Lithiation of N‐protected‐2,3‐dihydro‐1,4‐benzoxazines is described. Lithiation of N‐(tert‐butoxycarbonyl)‐2,3‐dihydro‐1,4‐benzoxazine ( 1 ) with BuLi/TMEDA occurred in the α‐position to nitrogen on the heterocyclic ring, leading to the unexpected ring‐opened product 3 . On the other hand, lithiation of N‐methyl‐2,3‐dihydro‐1,4‐benzoxazine ( 4 ) took place at the oxygen‐adjacent ortho‐position of the aromatic ring.     

Reductive alkylation of thioureas: a highly practical synthesis of unsymmetrical N,N′-disubstituted thioureas

A highly practical synthesis of unsymmetrical N,N′-disubstituted thioureas by the reductive alkylation of N-monosubstituted thioureas with aldehydes is described. N-Monosubstituted thioureas can in turn be synthesized by the reductive amination of thiourea with an appropriate aldehyde. This reductive alkylation methodology was also extended to carbamates.     

A General Synthesis of Functionalized Primary t-Alkylamines

A simple and efficient γ-acetamidopropylation of different nucleophiles using oxazine 3 has been developed. Deacetylation of acetamides with titanium(IV) isopropoxide/diphenylsilane is also described.     

A new reaction of N-aryl-2-pyrimidinamines with triphosgene

A new reaction of N-aryl-2-pyrimidinamines with triphosgene to afford N-aryl-N-(2-pyrimidinyl)-2-pyrimidinamine is described.
