We have developed a new algorithm to identify proteins by means of peptide mass fingerprinting. Starting from the matrix-assisted laser desorption/ionization-time-of-flight (MALDI-TOF) spectra and environmental data such as species, isoelectric point and molecular weight, as well as chemical modifications or number of missed cleavages of a protein, the program performs a fully automated identification of the protein. The first step is a peak detection algorithm, which allows precise and fast determination of peptide masses, even if the peaks are of low intensity or they overlap. In the second step the masses and environmental data are used by the identification algorithm to search in protein sequence databases (SWISS-PROT and/or TrEMBL) for protein entries that match the input data. Consequently, a list of candidate proteins is selected from the database, and a score calculation provides a ranking according to the quality of the match. To define the most discriminating scoring calculation we analyzed the respective role of each parameter in two directions. The first one is based on filtering and exploratory effects, while the second direction focuses on the levels where the parameters intervene in the identification process. Thus, according to our analysis, all input parameters contribute to the score, however with different weights. Since it is difficult to estimate the weights in advance, they have been computed with a generic algorithm, using a training set of 91 protein spectra with their environmental data. We tested the resulting scoring calculation on a test set of ten proteins and compared the identification results with those of other peptide mass fingerprinting programs. 相似文献

In the construction of a large area neutron detector(neutron wall)that is used to detect neutrons at GeV energies,the performances of all the sampling paddle modules prepared for the neutron wall areinvestigated with a specially designed test bench.Tested by cosmic rays,an average intrinsic time resolution of 222.5 ps is achieved at the center of the modules.The light attenuation length and the effective speed of the light in the module are also investigated. 相似文献

Cosmic-ray data of 90 M events have been collected and used for calibration, alignment as well as detector tuning. A special tracking algorithm for the BESⅢ muon counter is developed and verified with Monte-Carlo simulation and then further confirmed with the cosmic-ray data. The obtained strip resolutions are in good agreement with the design values. A new alignment approach for the BESⅢ muon counter is confirmed with the cosmic-ray data and proposed to be used in future analysis of experimental data. 相似文献

Time-resolved photoluminescence (TRPL) was applied to investigate the transient process in GaP1-xNx (x = 0.12%) alloy. The filling, transferring and decay processes among nitrogen pairs are directly observed. The NN4 pair, either not present or only a small obscure peak under a proper excitation condition in the steady-state photoluminescence spectrum, is well resolved by TRPL. 相似文献

Stable isotope labels are routinely introduced into proteomes for quantification purposes. Full labeling of cells in varying biological states, followed by sample mixing, fractionation and intensive data acquisition, is used to obtain accurate large‐scale quantification of total protein levels. However, biological processes often affect only a small group of proteins for a short time, resulting in changes that are difficult to detect against the total proteome background. An alternative approach could be the targeted analysis of the proteins synthesized in response to a given biological stimulus. Such proteins can be pulse‐labeled with a stable isotope by metabolic incorporation of ‘heavy’ amino acids. In this study we investigated the specific detection and identification of labeled proteins using acquisition methods based on Precursor Ion Scans (PIS) on a triple‐quadrupole ion trap mass spectrometer. PIS‐based methods were set to detect unique immonium ions originating from labeled peptides. Different labels and methods were tested in standard mixtures to optimize performance. We showed that, in comparison with an untargeted analysis on the same instrument, the approach allowed a several‐fold increase in the specificity of detection of labeled proteins over unlabeled ones. The technique was applied to the identification of proteins secreted by human cells into growth media containing bovine serum proteins, allowing the preferential detection of labeled cellular proteins over unlabeled bovine ones. However, compared with untargeted acquisitions on two different instruments, the PIS‐based strategy showed some limitations in sensitivity. We discuss possible perspectives of the technique.
