The influence of ionic strength on the complexes formed by natural bovine serum albumin (BSA), pH 5.4 (near the isoelectric point), and sodium dodecyl sulfate (SDS) in aqueous buffered (sodium acetate) solution was investigated by using surface tension, fluorescence and small angle X-ray scattering (SAXS) techniques. Ionic strength was varied by changing sodium acetate buffer concentration from 0.020 to 0.5 M. Surface tension revealed that SDS:BSA saturation binding occurs at psp = 42 +/- 2 mM, independent of the solution ionic strength. Further, SAXS curves are consistent with the necklace and bead model, where micelle-like aggregates are randomly distributed along the partial unfolded protein. Micelle-like aggregates grow from small spheres at 10 mM SDS to small ellipsoids (upsilon = 1.3 , ratio between the largest and the shortest axes) near psp, in good agreement with micellar aggregation numbers obtained by fluorescence, independent of salt concentration. Protein-bound micelles stop growing above psp and further SDS addition induces free-micelle formation.  相似文献

The interactions between octyl--D-glucoside and glycine in water have been investigated by surface tension, viscosity, and density measurements. The results show that the -amino acid causes an unexpected lowering of the critical micellar concentration of octyl--D-glucoside. Such a finding has been interpreted in temss of dipole-dipole interactions between the hydrophilic site of the surfactant and the peptidic cosluttes. From three to seven amino acid molecules have been estimated to be coordinated with each glucoside unity in the micellar state. The research has been extended to glycine oligopeptides and L-lysine. The latter compound has effects similar to those observed with glycine whereas diglycine and triglycine show weaker effects on the micellization process.  相似文献

The palladium-catalyzed desilylation-arylation of substituted vinylsilanes by p-iodoanisole in the presence of bidentate phosphine ligands is described. Apart from enhancing the rate of the reaction considerably, heteroatom-based functional groups in the vinylsilane moiety have a profound influence on the regiochemistry. A catalytic cycle for the chelation-controlled desilylation-arylation reaction involving five- and six-membered chelate rings is proposed.  相似文献

An analytical approach based on high-performance capillary electrophoresis (CE) in conjunction with electrospray ionization quadrupole time-of-flight tandem mass spectrometry (ESI-QTOF-MS/MS) has been developed for providing the basis to obtain new insights into the domain structure of the glycosaminoglycan (GAG) moiety of proteoglycans. The feasibility and performance of the off-line CE/ESI-QTOF-MS approach in GAG oligosaccharide analysis were assessed by screening a chondroitin/dermatan sulfate (DS) oligosaccharide mixture obtained from bovine aorta by enzymatic depolymerization by chondroitin B lyase. The CS/DS mixture was analyzed by CE using 50 mM ammonium acetate, pH 12.0, dissolved in aqueous methanol (2:3; v/v), as a CE carrier. Structural identification of the GAG components was achieved using off-line CE/nanoESI-QTOF-MS and-MS/MS experiments. ESI-QTOF instrumental parameters were found to play an important role in the MS screening of the CE-separated GAG species. By optimizing the ESI conditions, oligosaccharides differing in chain length and degree of sulfation could be detected. The building block composition, the size of the carbohydrate chain, as well as structural features of the repeating HexA-GalNAc, HexA-GalNAc(S) units, have been determined using MS/MS by applying collision-induced dissociation at low energies. Cleavage of GAG chains by chondroitin B lyase occurs with formation of structural markers useful for identification of IdoA-containing domains.  相似文献

In the discussion that follows some of the more recent progress in the area of anion binding by synthetic polyamine receptors is presented, with emphasis given to work undertaken by the authors’ groups. A continuing theme in these studies has been the relationship between receptor structure and its anion extraction properties.Systematic solvent extraction and structural studies for halide and perrhenate complexes with polyamines of tripodal, macrocyclic and macrobicyclic architecture that contain both aromatic moieties and four to eight amine functions have been performed in order to derive relevant structure-binding/extractability relationships. The results demonstrate that the binding and extraction behaviour of the polyamines towards halides and perrhenate is a complex function of their structural features, degree of protonation and lipophilic properties. The extraction is characterized by the preferred formation of mono- and diprotonated amine species in the organic phase. X-ray structure studies of iodide and perrhenate complexes with open-chain tetraamino derivatives and octaamino cryptands in different protonation states lead to the conclusion that in the first case only limited chelation of the anion occurs and in the second only highly protonated species are able to encapsulate the anion. The structural patterns observed are strongly influenced by the presence of water molecules in the crystals.  相似文献

The mechanical properties of hybrid materials consisting of polystyrene (PS), which was cross-linked with different proportions of the multifunctional cluster Zr6O4(OH)4(methacrylate)12 (Zr6) were investigated. With the help of (micro)indentation and scratch testing, the influence of the Zr6 clusters on mechanical properties, such as hardness, stiffness, creep, craze initiation, and scratch resistance was shown. There was only a slight increase in hardness and in indentation modulus with higher cluster loadings. While this observation was in agreement with the compression behaviour of the materials, the tensile properties showed a much stronger dependence on the Zr6 content. With increasing cluster loading, an increase of craze initiation stress, as determined by ball indentation experiments, was found. Performing scratch testing with constant load, a reduction of pile-up and a stronger recovery were observed for the hybrid materials compared to the neat PS. Scratch tests with a constant increase of load showed an increase of the critical load for crack opening during scratching.  相似文献

The copper‐catalyzed atom transfer radical polymerization (ATRP) of poly(propylene glycol) methacrylate (PPGM) in solution to produce linear and starlike polymers is reported, using methylethyl ketone as the solvent and a temperature of 80 °C. The ATRP system used was efficient for polymerization of the functionalized monomer without protecting hydroxyl end groups of monomer. The polymerizations were consistent with “living” or controlled processes, as revealed by the linear evolution of molecular weight with conversion. Increasing the [M]0:[I]0 ratio resulted in increasing molecular weights, whereas the polydispersity indices remained low (Mw/Mn < 1.4) even at high conversion. Decreasing the [CuBr]0:[I]0 ratio resulted in lower conversions, slightly larger polydispersities, and decreased molecular weights, likely resulting from a lower initiation efficiency. Polymers were characterized by 1H and 13C NMR; molecular weights of polymers with low degrees of polymerization were estimated by end‐group analysis from 13C NMR spectra obtained using distortionless enhancement by polarization transfer and the gated decoupling techniques. © 2001 John Wiley & Sons, Inc.
