Proteins present within the cell layer and those released in the cell medium from in vitro cultured normal human dermal fibroblasts were separated and characterized in terms of their isoelectric point and molecular weight, by two-dimensional (2-D) gel electrophoresis. All spots in the synthetic gel were firstly analyzed by the Melanie 3 software and compared with those of breast cancer cells, colorectal epithelial cells, HL60, lymphoma cells, and platelets, already available on-line. From the identification of 144 spots from both the cell layer and the medium, we were able to recognize 89 different proteins, since a certain number of spots represented different isoforms of the same molecule. Identifications were performed by matching with on-line 2-D databases, and by matrix assisted laser-desorption/ionization-time of flight-mass spectrometry (MALDI-TOF-MS), in order to confirm the identification by matching, or to identify new proteins. The procedure we used allows (i) to design a highly reproducible reference map of the proteome of adult human normal fibroblasts in culture, (ii) to evaluate protein species produced in the cell layer as well as those released in the culture medium, and (iii) to compare data from gel matching with those obtained by MS. This work represents an essential step for a better knowledge of mesenchymal cells, given the widespread use of this cell type in both clinical and experimental investigations.  相似文献

The spectra of some 9-chloro-6-nitroacridines with methoxy substituents in position 1,4; 2,4; 3,4; 2,3; and ethoxy substituents in positions 1, 2, 3, were recorded. The influence of electron releasing alkoxy groups and electron withdrawing nitro groups on chemical shifts of protons in the 9-chloroacridinic cycle was discussed and PMR parameters were determined, comparatively with those of acridine and 9-chloroacridine. Good agreement between calculated and experimental values is observed. The spectrum of 9-chloro-1,2,3-triethoxyacridine shows for the four protons of the ringC. (H-5, H-6, H-7, H-8) anAMNX coupling pattern whose parameters were determined.  相似文献

The distortion of the charge cloud around a uniformly charged, dielectric, rigid sphere that translates and rotates in an unbounded binary, symmetric electrolyte at zero Reynolds number is examined. The zeta potential of the particle ζ is assumed small relative to the thermal voltage scale. It is assumed that the equilibrium structure of the cloud is slightly distorted, which requires that the Péclet numbers characterizing distortion due to particle translation, , and rotation, , are small compared to unity. Here, a is radius of the particle; D is the ionic diffusion coefficient; and , where U and Ω are the rectilinear and angular velocities of the particle, respectively. Perturbation expansions for small and are employed to calculate the nonequilibrium structure of the cloud, whence the force and torque on the particle are determined. In particular, we predict that the sphere experiences a force orthogonal to its directions of translation and rotation. This “lift” force arises from the nonlinear distortion of the cloud under the combined actions of particle translation and rotation. The lift force is given by . Here, ε is the permittivity of the electrolyte; is the Debye length; and is a negative function that decreases in magnitude with increasing . The lift force implies that an unconstrained particle would follow a curved path; an electrokinetic analog of the inertial Magnus effect. Finally, the implication of the lift force on cross‐streamline migration of an electrophoretic particle in shear flow is discussed.  相似文献

A simple observation about the action for geodesics in a stationary spacetime with separable geodesic equations leads to a natural class of slicings of that spacetime whose orthogonal geodesic trajectories represent the world lines of freely falling fiducial observers. The time coordinate function can then be taken to be the observer proper time, leading to a unit lapse function, although the time coordinate lines still follow Killing trajectories to retain the explicitly stationary nature of the coordinate grid. This explains some of the properties of the original Painlevé-Gullstrand coordinates on the Schwarzschild spacetime and their generalization to the Kerr-Newman family of spacetimes, reproducible also locally for the Gödel spacetime. For the static spherically symmetric case the slicing can be chosen to be intrinsically flat with spherically symmetric geodesic observers, leaving all the gravitational field information in the shift vector field.  相似文献

Snake venom hyaluronidases known as “spreading factor” are not extensively studied. Recently, it is argued that beyond its role as a spreading factor, venom hyaluronidase (HYL) deserves to be explored as a possible therapeutic target for inhibiting the systemic distribution of venom/toxins and also for minimizing local tissue destruction. In this context, in the present study, a low-molecular weight HYL has been isolated from Bungarus caeruleus (Indian krait) venom by single step chromatography on HPLC system. The apparent molecular weight determined by SDS-PAGE is 14 ± 2 kDa, as confirmed by zymogen study and LC–MS as well. The enzyme had optimal pH 6 and temperature 37°C. The Michaelis–Menten constant (Km) was found to be 8.48 µg/mL at 37°C. The activity of purified enzyme was completely inhibited by Ba2+ metal ion and N-acetyl imidazole group-specific agents. This work yielded a highly active HYL from B. caeruleus the first one to be isolated. Further studies on its pharmacological actions will be interesting to develop lead molecules for better management of snakebite.  相似文献

Selective monofunctionalization of substrates with distant, yet equally reactive functional groups is difficult to achieve, as it requires the second functional group to selectively modulate its reactivity once the first functional group has reacted. We now show that mechanically interlocked catalytic rings can effectively regulate the reactivity of stoppering groups in rotaxanes over a distance of about 2 nm. Our mechanism of communication is enabled by a unique interlocked design, which effectively removes the catalytic rings from the substrates by fast dethreading as soon as the first reaction has taken place. Our method not only led to a rare example of selective monofunctionalization, but also to a “molecular if function”. Overall, the study presents a way to get distant functional groups to communicate with each other in a reaction‐history‐dependent manner by creating linkers that can ultimately perform logical operations at the molecular level.  相似文献

The emission pattern in the v(perp)-v(par) plane of intermediate mass fragments with Z = 3--7 (IMF) has been studied in the collision 116Sn+ 93Nb at 29.5A MeV as a function of the total kinetic energy loss of the reaction. This pattern shows that for peripheral reactions most IMF's are emitted at velocities intermediate between those of the projectile- and target-like products. Coulomb trajectory calculations show that these IMF's are produced in the interaction zone in a short time interval at the end of the target-projectile interaction.  相似文献
