The Tn antigen (GalNAc-α-1-O-Thr/Ser) is a well-known tumor-associated carbohydrate determinant. The use of glycopeptides that incorporate this structure has become a significant and promising niche of research owing to their potential use as anticancer vaccines. Herein, the conformational preferences of a glycopeptide with an unnatural Tn antigen, characterized by a threonine decorated with an sp2-iminosugar-type α-GalNAc mimic, have been studied both in solution, by combining NMR spectroscopy and molecular dynamics simulations, and in the solid state bound to an anti-mucin-1 (MUC1) antibody, by X-ray crystallography. The Tn surrogate can mimic the main conformer sampled by the natural antigen in solution and exhibits high affinity towards anti-MUC1 antibodies. Encouraged by these data, a cancer vaccine candidate based on this unnatural glycopeptide and conjugated to the carrier protein Keyhole Limpet Hemocyanin (KLH) has been prepared and tested in mice. Significantly, the experiments in vivo have proved that this vaccine elicits higher levels of specific anti-MUC1 IgG antibodies than the analog that bears the natural Tn antigen and that the elicited antibodies recognize human breast cancer cells with high selectivity. Altogether, we compile evidence to confirm that the presentation of the antigen, both in solution and in the bound state, plays a critical role in the efficacy of the designed cancer vaccines. Nanoparticles are widely studied as carrier vehicles in biological systems because their size readily allows access through cellular membranes. Moreover, they have the potential to carry cargo molecules and as such, these factors make them especially attractive for intravenous drug delivery purposes. Interest in protein-based nanoparticles has recently gained attraction due to particle biocompatibility and lack of toxicity. However, the production of homogeneous protein nanoparticles with high encapsulation efficiencies, without the need for additional cross-linking or further engineering of the molecule, remains challenging. Herein, we present a microfluidic 3D co-flow device to generate human serum albumin/celastrol nanoparticles by co-flowing an aqueous protein solution with celastrol in ethanol. This microscale co-flow method resulted in the formation of nanoparticles with a homogeneous size distribution and an average size, which could be tuned from ≈100 nm to 1 μm by modulating the flow rates used. We show that the high stability of the particles stems from the covalent cross-linking of the naturally present cysteine residues within the particles formed during the assembly step. By choosing optimal flow rates during synthesis an encapsulation efficiency of 75±24 % was achieved. Finally, we show that this approach achieves significantly enhanced solubility of celastrol in the aqueous phase and, crucially, reduced cellular toxicity. 相似文献

An analysis is made on the absorption distribution within a cylindrical active medium lamp pumped by means of fused silica light-guides. The performances of the proposed pump cavities are analysed through a non-sequential 3D ray-trace program and are compared to those of three reflective chambers: an elliptical cavity, a close-coupled cavity and a diffusive close-wrap cavity. Two light-guides cavities with eight and nine guides, respectively, were built and tested. High coupling efficiency and a reasonable uniform absorption distribution were registered. An overall laser efficiency of 1% was measured. The proposed cavities have permitted the tailoring of the absorption distribution within the laser rod. 相似文献

We investigate the spin-orbit (SO) interaction in two-dimensional electron gases in quantum wells with two subbands. From the 8x8 Kane model, we derive a new intersubband-induced SO term which resembles the functional form of the Rashba SO but is nonzero even in symmetric structures. This follows from the distinct parity of the confined states (even or odd) which obliterates the need for asymmetric potentials. We self-consistently calculate the new SO coupling strength for realistic wells and find it comparable to the usual Rashba constant. Our new SO term gives rise to a nonzero ballistic spin-Hall conductivity, which changes sign as a function of the Fermi energy (epsilonF) and can induce an unusual Zitterbewegung with cycloidal trajectories without magnetic fields. 相似文献
