Simultaneous quantification of leukotrienes and hydroxyeicosatetraenoic acids in cell culture medium using liquid chromatography/tandem mass spectrometry

Leukotrienes (LTs) and hydroxyeicosatetraenoic acids (HETEs) are important bioactive lipid mediators that participate in various pathophysiological processes. To advance understanding of the mechanisms that regulate these mediators in physiological and pathological processes, an analytical method using liquid chromatography/tandem mass spectrometry for the simultaneous quantification of LTB₄, LTC₄, LTD₄, LTE₄, 5‐HETE_, 8‐HETE, 12‐HETE and 15‐HETE in cell culture media was developed. A Supel?‐Select HLB solid‐phase extraction cartridge was used for sample preparation. The compounds were separated on a C₁₈ column using gradient elution with acetonitrile–water–formic acid (20:80:0.1, v/v/v) and acetonitrile–formic acid (100:0.1, v/v). The calibration curves of LTB₄, LTD₄, LTE₄ and HETEs were linear in the range of 0.025–10 ng/mL, and the calibration curve of LTC₄ was linear in the range of 0.25–10 ng/mL. Validation assessment showed that the method was highly reliable with good accuracy and precision. The stability of LTs and HETEs was also investigated. Using the developed method, we measured LTs and HETEs in the culture supernatant of the human mast cell line HMC‐1. The present method could facilitate investigations of the mechanisms that regulate the production, release and signaling of LTs and HETEs. Copyright © 2014 John Wiley & Sons, Ltd.     

Stable all-optical formation of Bose–Einstein condensate using pointing-stabilized optical trapping beams

By stabilizing the beam pointing of optical trapping beams, we have succeeded in stable formation of Bose–Einstein condensate (BEC) of ⁸⁷Rb with all-optical method. The thermal effect of acousto-optic modulator (AOM) is usually one of the most serious problems to induce beam-pointing instability, especially for high power CO₂ laser. By passing the beam through AOM twice, we have improved the beam pointing from about 4.8 mrad to less than 0.4 mrad, which has been experimentally confirmed to be small enough to stably form BEC at the crossed region of CO₂ lasers as well as to perform experiments using an optical lattice which might have been affected by beam-pointing instability. Inflation model with lower multipoles of the CMB suppressed

The recent observation of the cosmic microwave background anisotropy by the WMAP confirmed that the lower multipoles are considerably suppressed. From the standpoint of the cosmic variance, it is nothing but a statistical accident. Alternatively, one can attribute the deficit of fluctuation on the large scale to the cosmic history, which might be explained in the context of the inflationary physics. In this Letter, we show that it is possible to explain the suppressed lower multipoles in the hybrid new inflation model.     

Optical properties of impurity atoms in pressurized superfluid helium

We report on the optical properties of alkali atoms (Cs and Rb) in the pressurized superfluid helium. We observed excitation and emission spectra at various pressures from the saturated vapor pressure to about 25 atm. The theoretical calculations on the basis of the atomic bubble model have also been worked out. The qualitative agreement between the theoretical and experimental results with respect to the peak shift, the linewidth, and their pressure dependence is achieved in the framework of the spherical atomic bubble model. TheD ₂ excitation spectra with the double peaks are interpreted qualitatively in terms of the Jahn-Teller effect, indicating the existence of the nontotally symmetrical density distribution of the surrounding helium atoms.     

High-throughput characterization of local conductivity of Nd0.9Ca0.1Ba2Cu3O7−δ thin film by the low-temperature scanning microwave microscope

We developed a scanning microwave microscope (SμM) designed for high-throughput electric-property screening as well as for rapid construction of electronic phase diagrams at low temperatures. As a sensor probe, we used a high-Qλ/4 coaxial cavity resonator to which a thin needle with ball-tip end was attached. The sensor module was mounted on the low-temperature XYZ stage, which allowed us to map out the change of resonance frequency and quality factor due to the local tip-sample interaction at low temperatures. From the measurements of combinatorial thin films, such as Ti_1−xCo_xO_2−δ and Nd_0.9Ca_0.1Ba₂Cu₃O_7−δ (NCBCO), it was demonstrated that this SμM system has enough performance for the high-throughput characterization of sample conductance under variable temperature conditions.     

THE DESIGNS OF HIGH EFFICIENCY LAUNCHER OF QUASI-OPTICAL MODE CONVERTER FOR HIGH POWER GYROTRONS

A high efficiency launcher of quasi-optical (QO) mode converters for high power gyrotrons have been designed and tested. A helical cut launcher radiates the RF power via its straight cut onto the first phase correcting mirror. The launchers have been optimized for the TE_31.8 mode at 170 GHz and TE_22.6 mode at 110 GHz by numerically optimizing a launcher surface. The helical cut of the launcher has been optimized by taking the taper angle into account. Further more, the amplitude of the surface perturbation have been optimized for improved focusing in order to reduce the diffraction losses at the helical cut. Low power measurement shows a good agreement with the design. High efficiency characteristics of the design have also been calculated on the assumption of frequency downshift due to the thermal expansion of the cavity and stepwise frequency tuning by changing the operating mode. Besides, the possibility of high efficiency launcher for higher mode is discussed, and these results give the prospect to high efficiency long pulse gyrotrons.     

Direct determination of benzamides in serum by column-switching high-performance liquid chromatography.

A column-switching high-performance liquid chromatographic method with fluorescence detection was developed for the simultaneous determination of four benzamide-type anti-psychotic drugs: sulpiride, tiapride, sultopride and metoclopramide in human serum. In this method, a TSKgel Super-ODS column was used as an analytical column, and a TSKgel G 2000SW was prepared as a pretreatment column. Under the optimized analytical conditions, four benzamide-type anti-psychotic drugs were eluted within 18 min. The detection limits (S/N = 3) for sulpiride, tiapride, sultopride and metoclopramide are 1 ng/ml, 4 ng/ml, 2 ng/ml and 0.5 ng/ml, respectively. Finally, the method was applied to the determination of sulpiride in human serum samples obtained after a single oral dose of sulpiride.
