The application of the recently introduced dispersive liquid–liquid microextraction (DLLME) for the separation and determination of an inorganic selenite [Se(IV)] derivative by means of a gas chromatography–electron-capture detection system has been studied. The selenium derivative was extracted with the DLLME technique using a mixture of ethanol (disperser solvent) and chlorobenzene (extraction solvent). The influences of the various analytical parameters on the derivatization reaction and microextraction procedure have been evaluated and optimized. Under the optimum conditions, an enrichment factor of 122 was obtained for only 5.00 mL of the water sample. The calibration graph was linear in the range of 0.015–10 μg L?1 with a detection limit of 0.005 μg L?1. The relative standard deviation for ten replicate measurements of 2 μg L?1 of selenium was 4.1%. The method was applied to the determination of selenium in environmental surface water samples with satisfactory recovery.  相似文献

A simple and powerful microextraction technique was used for determination of selenium in water samples using dispersive liquid-liquid microextraction (DLLME) followed by graphite furnace atomic absorption spectrometry (GF AAS). DLLME and simultaneous complex formation was performed with rapid injection of a mixture containing ethanol (disperser solvent), carbon tetrachloride (extraction solvent) and ammonium pyrrolidine dithiocarbamate (APDC, chelating agent) into water sample spiked with selenium. After centrifuging, fine droplets of carbon tetrachloride, which were dispersed among the solution and extracted Se-APDC complex, sediment at the bottom of the conical test tube. The concentration of enriched analyte in the sedimented phase was determined by iridium-modified pyrolitic tube graphite furnace atomic absorption spectrometry. The concentration of selenate was obtained as the difference between the concentration of selenite after and before pre-reduction of selenate to selenite. Some effective parameters on extraction and complex formation, such as extraction and disperser solvent type and their volume, extraction time, salt effect, pH and concentration of chelating agent were optimized. Under the optimum conditions, the enrichment factor of 70 was obtained from only 5.00 mL of water sample. The calibration graph was linear in the range of 0.1-3 μg L− 1 with detection limit of 0.05 μg L− 1. The relative standard deviation (RSDs) for ten replicate measurements of 2.00 μg L− 1 of selenium was 4.5%. The relative recoveries of selenium in tap, river and sea water samples at spiking level of 2.00 μg L− 1 were 106, 96 and 98%, respectively.  相似文献

We present a binding free energy function that consists of force field terms supplemented by solvation terms. We used this function to calibrate the solvation model along with the binding interaction terms in a self-consistent manner. The motivation for this approach was that the solute dielectric-constant dependence of calculated hydration gas-to-water transfer free energies is markedly different from that of binding free energies (J. Comput. Chem. 2003, 24, 954). Hence, we sought to calibrate directly the solvation terms in the context of a binding calculation. The five parameters of the model were systematically scanned to best reproduce the absolute binding free energies for a set of 99 protein-ligand complexes. We obtained a mean unsigned error of 1.29 kcal/mol for the predicted absolute binding affinity in a parameter space that was fairly shallow near the optimum. The lowest errors were obtained with solute dielectric values of Din = 20 or higher and scaling of the intermolecular van der Waals interaction energy by factors ranging from 0.03 to 0.15. The high apparent Din and strong van der Waals scaling may reflect the anticorrelation of the change in solvated potential energy and configurational entropy, that is, enthalpy-entropy compensation in ligand binding (Biophys. J. 2004, 87, 3035-3049). Five variations of preparing the protein-ligand data set were explored in order to examine the effect of energy refinement and the presence of bound water on the calculated results. We find that retaining water in the final protein structure used for calculating the binding free energy is not necessary to obtain good results; that is the continuum solvation model is sufficient. Virtual screening enrichment studies on estrogen receptor and thymidine kinase showed a good ability of the binding free energy function to recover true hits in a collection of decoys.  相似文献

Despite considerable advantages of the railway track over other means of transportation, noise pollution is the main adverse consequence of railway transportation. The basic cause of railway noise is rail corrugation. Although characteristics of railway noise have been considerably studied in the literature, rail corrugation effects on rolling noise generation in tangent tracks and the curves have not been sufficiently investigated. This research addresses the limitations of the current understanding of the rolling noise generation by investigating rail corrugation effects on rolling noise in tangent tracks and curves. This research was made based on the results obtained form a thorough field investigation carried out in a railway line which includes tangents tracks and sharp curves. A track geometry recording car was used to measure rail corrugations. For this purpose, an indirect method was developed in this research to obtain rail corrugation patterns from the data recorded by the track recording car. The effectiveness of the new method was shown. The induced noises were recorded using two particular types of microphones and implementing the method suggested by the ISO 3095 Standard. The rolling noise signal was distinguished from the total noise, using Butterworth Band-Pass Filtering. The role of rail corrugations in the rolling noise was discussed. Correlations were made between various types of corrugations and the induced noises. The results were presented and discussed in the spatial and frequency scales. Results obtained have led to new findings in rail corrugation effects on rolling noise generation. This research paves a way toward a better understanding of rolling noise sources and the parameters influencing the noise.  相似文献

Dispersive liquid-liquid microextraction and gas chromatography-flame photometric detection (DLLME-GC-FPD) were performed for the speciation of butyl and phenyltin compounds in water samples after derivatization with sodium tetraethylborate (NaBEt4). Some important parameters, such as pH, amount of NaBEt4, derivatization time, kind and volume of extraction and disperser solvents, extraction time and salt effect were investigated and optimized. High enrichment factors (825-1036) and low detection limits (0.2-1 ng L(-1)) were obtained under the optimum conditions. The calibration graphs were linear in the range of 0.5-1000 ng L(-1) (as Sn) for the target analytes. The relative standard deviations (RSDs) for the extraction of 20 ng L(-1) (as Sn) of butyl and phenyltin compounds varied from 2.3 to 5.9% (n=7) and from 4.1 to 8.8% (n=7) with and without using internal standard, respectively. Seawater and river water samples were successfully analyzed using the proposed method and the relative recoveries of the studied compounds in the water samples, at spiking levels of 10.0 and 100 ng L(-1) (as Sn) were obtained to be 82.5-104.7%.  相似文献

Macitentan is an endothelin receptor antagonist commonly used in the treatment of pulmonary arterial hypertension (PAH). A novel, rapid, simple and sensitive UPLC–MS/MS method was developed and validated for pharmacokinetic study and the determination of macitentan in PAH patients. Macitentan and bosentan, which are used as internal standards, were detected using atmospheric pressure chemical ionization in positive ion and multiple reaction monitoring mode by monitoring the mass transitions m/z 589.1 → 203.3 and 552.6 → 311.5, respectively. Chromatographic separation was performed on a reverse‐phase C18 column (5 μm, 4.6 × 150 mm) with an isocratic mobile phase, which consisted of water containing 0.2% acetic acid–acetonitrile (90:10, v/v) at a flow rate of 1 mL/min. Retention times were 1.97 and 1.72 min for macitentan and IS, respectively. The calibration curve with high correlation coefficient (0.9996) was linear in the range 1–500 ng/mL. The lower limit of quantitation and average recovery values were determined as 1 ng/mL and 89.8%, respectively. This method is the first UPLC–MS/MS method developed and validated for the determination of macitentan from human plasma. The developed analytical method was fully validated for linearity, selectivity, specificity, accuracy, precision, sensitivity, stability, matrix effect and recovery according to US Food and Drug Administration guidelines. The developed method was applied successfully for pharmacokinetic study and the determination of macitentan in PAH patients.  相似文献

Significant advances have led to receptor induced-fit and conformational selection models for describing bimolecular recognition, but a more comprehensive view must evolve to also include ligand shape and conformational changes. Here, we describe an example where a ligand's "structural hinge" influences potency by inducing an "L-shape" bioactive conformation, and due to its solvent exposure in the complex, reasonable conformation-activity-relationships can be qualitatively attributed. From a ligand design perspective, this feature was exploited by successful linker hopping to an alternate "structural hinge" that led to a new and promising chemical series which matched the ligand bioactive conformation and the pocket bioactive space. Using a combination of X-ray crystallography, NMR and modeling with support from binding-site resistance mutant studies and photoaffinity labeling experiments, we were able to derive inhibitor-polymerase complexes for various chemical series.  相似文献

A novel method for preconcentration is described for chromium speciation at microgram per liter to sub-microgram per liter levels. It is based on selective complex formation of both Cr(VI) and Cr(III) followed by dispersive liquid–liquid microextraction and determination by microsample introduction-flame atomic absorption spectrometry. Effects influencing complex formation and extraction (such as pH, temperature, time, solvent, salinity and the amount of chelating agent) have been optimized. Enrichment factors up to 275 and 262 were obtained for Cr(VI) and total Cr, respectively. The calibration graph is linear from 0.3 to 20 µg L?1, and detection limits are 0.07 and 0.08 µg L?1 for Cr(VI) and total Cr, respectively. Relative standard deviations (RSDs) were obtained to be 2.0% for Cr(VI) and 2.6% for total Cr (n?=?7).  相似文献

Dispersive liquid-liquid microextraction (DLLME) technique was successfully used as a sample preparation method for graphite furnace atomic absorption spectrometry (GF AAS). In this extraction method, 500 μL methanol (disperser solvent) containing 34 μL carbon tetrachloride (extraction solvent) and 0.00010 g ammonium pyrrolidine dithiocarbamate (chelating agent) was rapidly injected by syringe into the water sample containing cadmium ions (interest analyte). Thereby, a cloudy solution formed. The cloudy state resulted from the formation of fine droplets of carbon tetrachloride, which have been dispersed, in bulk aqueous sample. At this stage, cadmium reacts with ammonium pyrrolidine dithiocarbamate, and therefore, hydrophobic complex forms which is extracted into the fine droplets of carbon tetrachloride. After centrifugation (2 min at 5000 rpm), these droplets were sedimented at the bottom of the conical test tube (25 ± 1 μL). Then a 20 μL of sedimented phase containing enriched analyte was determined by GF AAS.Some effective parameters on extraction and complex formation, such as extraction and disperser solvent type and their volume, extraction time, salt effect, pH and concentration of the chelating agent have been optimized. Under the optimum conditions, the enrichment factor 125 was obtained from only 5.00 mL of water sample. The calibration graph was linear in the rage of 2-20 ng L−1 with detection limit of 0.6 ng L−1. The relative standard deviation (R.S.D.s) for ten replicate measurements of 20 ng L−1 of cadmium was 3.5%. The relative recoveries of cadmium in tap, sea and rivers water samples at spiking level of 5 and 10 ng L−1 are 108, 95, 87 and 98%, respectively. The characteristics of the proposed method have been compared with cloud point extraction (CPE), on-line liquid-liquid extraction, single drop microextraction (SDME), on-line solid phase extraction (SPE) and co-precipitation based on bibliographic data. Therefore, DLLME combined with GF AAS is a very simple, rapid and sensitive method, which requires low volume of sample (5.00 mL).  相似文献
