The anticancer activity of terretonin N (1) and butyrolactone I (2), obtained from the thermophilic fungus Aspergillus terreus TM8, was intensively studied against prostate adenocarcinoma (PC-3) and ovary adenocarcinoma (SKOV3) human cell lines. According to this study, both compounds showed potent cytotoxicity towards ovarian adenocarcinoma cells (SKOV3) with IC50 1.2 and 0.6 μg/mL, respectively. With respect to metastatic prostate cells (PC-3), the two compounds 1 and 2 showed a significantly promising cytotoxicity effect with IC50 of 7.4 and 4.5 μg/mL, respectively. The tested fungal metabolites showed higher rates of early and late apoptosis with little or no necrotic apoptotic pathway in all treated prostate adenocarcinoma (PC-3) and ovary adenocarcinoma (SKOV3) human cell lines, respectively. The results reported in this study confirmed the promising biological properties of terretonin N (1) and butyrolactone I (2) as anticancer agents via the induction of cellular apoptosis. However, further studies are needed to elucidate the molecular mechanism by which cellular apoptosis is induced in cancer cells.  相似文献

This work describes the fabrication of long chain alkyl methacrylate monolithic materials for use as stationary phases in capillary liquid chromatography. After capillary inner wall surface activation with 3-(trimethoxysilyl)propyl methacrylate, monoliths were formed by copolymerization of either lauryl or stearyl methacrylate (LMA or SMA) with ethylene dimethacrylate (EDMA) as crosslinker, in the presence of azobisisobutyronitrile (AIBN) as initiator and a mixture of porogenic solvents including water, 1-propanol and 1,4-butanediol. The composition of the polymerization mixture was changed in terms of monomer, crosslinker and porogen ratio composition, in order to compare the influence of these parameters. The monoliths were prepared in 320 ??m i.d. and 200 mm length capillaries. The column morphology was characterized by optical microscopy and scanning electron microscopy (SEM). Total porosity and permeability of each column were calculated using uracil as unretained material by measuring the pressure drop across the columns as a function of linear velocity. The microglobule average size for each column was also determined using Hagen?CPoiseuille equation and compared with the SEM images. As expected, a decrease of the porogen to monomer ratio corresponded to smaller microglobules and a lower total porosity. The columns were then chromatographically evaluated; good results were obtained when these capillaries were used to separate mixtures of phenols, aromatics and drug compounds.  相似文献

The identification of ignitable liquids is very important and challenging aspect in arson crime investigations. The detection of gasoline and diesel fuel components using solid phase micro-extraction prior to gas chromatography–mass spectrometry for the forensic analysis of fire debris has been carried out. Previous works show that the absorption characteristics of the substrate are one of the most important factors in determining the evaporation rate of the accelerants. In order to determine the presence of the fuel residues, four of the most common substrate materials were tested in this work; wool, cotton, silk and polyester. The obtained results indicate that both gasoline and diesel fuel accelerants persisted longer on wool and silk than on the other selected substrates. Such information illustrates the influence of fuel persistence times after extinguishing and the best materials to be scanned for ignitable liquids at the fire scene.  相似文献

A monolithic capillary column containing a composite of metal–organic framework MIL‐53(Al) incorporated into hexyl methacrylate‐co‐ethylene dimethacrylate was prepared to enhance the separation of mixtures of small aromatic compounds by using capillary liquid chromatography. The addition of 10 mg/mL MIL‐53(Al) microparticles increased the micropore content in the monolithic matrix and increased the Brunauer–Emmett–Teller surface area from 26.92 to 85.12 m2/g. The presence of 1,4‐benzenedicarboxylate moieties within the structure of MIL‐53(Al) as an organic linker greatly influenced the separation of aromatic mixtures through π–π interactions. High‐resolution separation was obtained for a series of alkylbenzenes (with resolution factors in the range 0.96–1.75) in less than 8 min, with 14 710 plates/m efficiency for propylbenzene, using a binary polar mobile phase of water/acetonitrile in isocratic mode. A reversed‐phase separation mechanism was indicated by the increased retention factor and resolution as the water percentage in the mobile phase increased. A stability study on the composite column showed excellent mechanical stability under various conditions. The higher resolution and faster separation observed at increased temperature indicated an exothermic separation, whereas the negative values for the free energy change of transfer indicated a spontaneous process.  相似文献

Hendra virus (HeV) belongs to the paramyxoviridae family of viruses which is associated with the respiratory distress, neurological illness, and potential fatality of the affected individuals. So far, no competitive approved therapeutic substance is available for HeV. For that reason, the current research work was conducted to propose some novel compounds, by adopting a Computer Aided Drug Discovery approach, which could be used to combat HeV. The G attachment Glycoprotein (Ggp) of HeV was selected to achieve the primary objective of this study, as this protein makes the entry of HeV possible in the host cells. Briefly, a library of 6000 antiviral compounds was screened for potential drug-like properties, followed by the molecular docking of short-listed compounds with the Protein Data Bank (PDB) structure of Ggp. Docked complexes of top two hits, having maximum binding affinities with the active sites of Ggp, were further considered for molecular dynamic simulations of 200 ns to elucidate the results of molecular docking analysis. MD simulations and Molecular Mechanics Energies combined with the Generalized Born and Surface Area (MMGBSA) or Poisson–Boltzmann and Surface Area (MMPBSA) revealed that both docked complexes are stable in nature. Furthermore, the same methodology was used between lead compounds and HeV Ggp in complex with its functional receptor in human, Ephrin-B2. Surprisingly, no major differences were found in the results, which demonstrates that our identified compounds can also perform their action even when the Ggp is attached to the Ephrin-B2 ligand. Therefore, in light of all of these results, we strongly suggest that compounds (S)-5-(benzylcarbamoyl)-1-(2-(4-methyl-2-phenylpiperazin-1-yl)-2-oxoethyl)-6-oxo-3,6-dihydropyridin-1-ium-3-ide and 5-(cyclohexylcarbamoyl)-1-(2-((2-(3-fluorophenyl)-2-methylpropyl)amino)-2-oxoethyl)-6-oxo-3,6-dihydropyridin-1-ium-3-ide could be considered as potential therapeutic agents against HeV; however, further in vitro and in vivo experiments are required to validate this study.  相似文献

This work describes the preparation of polymer based monolithic materials and their use as stationary phases in capillary liquid chromatography. Multi-walled carbon nanotubes (MWCNT) were incorporated into a mixture containing benzyl methacrylate (BMA) and ethylene dimethacrylate (EDMA) as co-monomers. The optimized porogenic mixture was a ternary solution composed of cyclohexanol, 1,4-butandiol and butanol which resulted in a stable and homogeneous suspension. Six capillary columns with increasing amounts of MWCNT, from 0 to 0.4 mg mL(-1), were prepared by thermal polymerization in 0.32 mm (i.d.) and 150 mm length fused silica tubing. The chromatographic evaluation showed that the synthesized monolithic beds were mechanically stable while their porosity and permeability increased with the MWCNT content. The prepared capillary columns were tested for the separation of mixtures of ketones and phenols at an optimum flow rate of 2 μL min(-1). The results showed that incorporation of MWCNT slightly affected the retention while it enhanced the column efficiency by increasing the column efficiency by a factor of up to 9. This effect corresponded also to an improved resolution and full separation of the solutes.  相似文献

Since their discovery in 1991 by the Japanese scientist “Sumio Iijima”, carbon nanotubes have been of great interest, both from a fundamental point of view and for future applications. Different types of carbon nanotubes can be produced in various ways. Economically feasible large-scale production and purification techniques are still under development. Carbon nanotubes are discussed in this review in terms of history, types, structure, synthesis and characterisation methods. Carbon nanotubes have attracted the fancy of many scientists worldwide.
