A Preparative Method for Crosslinking Proteins to DNA in Nuclei by Single-pulse UV Laser Irradiation
Abstract— Photocrosslinking of proteins to DNA by single-pulse UV laser has been used only in analytical experiments, carried out with reconstituted complexes of a single DNA binding protein and a labeled target sequence. Here we propose a large-scale technique for irradiation of nuclei, generating preparative quantities of covalently linked protein-DNA complexes for further analysis of the partner molecules. The use of a flow cuvette allows a milligram of DNA in either nuclei or chromatin to be irradiated by a single pulse within few minutes. The efficiency of crosslinking varies from 6 to 12% of the total nuclear proteins. The presence of histones and other chromosomal proteins in the crosslinked protein-DNA complexes was demonstrated by using specific antibodies. The irradiation procedure can be fully automated using a microcomputer. 相似文献

A quasi-steady rolling problem with nonlocal friction, for porous rigid-plastic, strain-rate-sensitive and strain hardening materials, is considered. A variational formulation is derived, consisting of a variational inequality and two evolution equations, coupling the velocity, strain hardening and relative density variables. The convergence of a variable stiffness parameters method is proved, and existence and uniqueness results are obtained. This study is devoted to investigation of the micro- and mesostructure (including fractal properties) of porous zirconia glasses synthesized by precipitation from zirconium n-propoxide solutions in the presence of different hydrolyzing-agent (H2O) quantities at different temperatures. Analysis of small-angle neutron, ultra-small-angle neutron and X-ray scattering, scanning electron microscopy allows concluding that the synthesized glasses are complex systems with a three-level hierarchical fractal structure. It is revealed that both the temperature of synthesis and the H2O concentration in the initial solution significantly affect the structural characteristics of the glasses. 相似文献
