The mechanism of the unfolding of human serum albumin by urea was studied using d.c. polarography. It was found that this reaction is a complex process which cannot be described in terms of a two-state transition model. As well as the Brdi?ka catalytic current we have also studied the reduction current of disulfide groups in native and denatured human serum albumin. The number of cystine residues accessible for electrode reduction in native and denatured protein was calculated. On the basis of these results a scheme for the unfolding of human serum albumin by urea is proposed.  相似文献

The enthalpies of formation and total energies of a series of 1- and 2-substituted tetrazoles in the gas phase and in solution were calculated by MNDO and AM1 semiempirical methods and by a set of nonempirical procedures. The effect of solvent on the relative stability of N-substituted tetrazoles was estimated in terms of the PCM, SM5, and SCRF models. The possibility for isomerization of N-substituted tetrazoles, depending on the substituent in position 1 or 2, was studied at the MP2/6-31G*//HF/6-31G* level. According to the results of nonempirical calculations, 2-substituted tetrazoles are more stable in the gas phase, in keeping with the experimental data for the corresponding isomers. The solvent nature is an important factor affecting isomerization of N-substituted tetrazoles: Rise in the solvent polarity leads to displacement of the equilibrium toward 1-substituted isomers.  相似文献

57Fe Mössbauer spectroscopy is used to study neutron irradiation induced changes in the short-range order of Fe73.5Nb3Cu1Si13.5B9 alloy. The samples are investigated in both amorphous and nanocrystalline states. Neutron irradiation leads to an increase of the standard deviation of a hyperfine field distribution (HFD), implying rearrangement of the atoms towards disordering. Simultaneously, changes in the average value of the hyperfine field and a net magnetic moment position occur as a consequence of a spin reorientation, atom mixing and microscopic stress centres which are introduced by neutron irradiation.  相似文献

This study aimed to investigate matrix metalloproteinase (MMP) activity in human dentin using in-situ and gelatin zymography, after at-home and in-office bleaching, related to their clinical exposure times. Dentin specimens (n = 5) were treated with 35% hydrogen peroxide (50 min per session/4 sessions), 10% carbamide peroxide (180 min/21 sessions), or no treatment. All were subjected to in-situ zymography. Dentin slices were, subsequently, obtained, covered with fluorescein-conjugated gelatin, and examined with confocal laser-scanning microscopy. The fluorescence intensity was quantified and statistically analyzed using one-way ANOVA and Bonferroni tests (α = 0.05). Furthermore, gelatin zymography was performed on protein extracts obtained from dentin powder (N = 8 teeth), treated with hydrogen peroxide or carbamide peroxide, with different exposure times (10/50 min for hydrogen peroxide; 252/1260 min for carbamide peroxide). The results of the in-situ zymography showed no statistical differences between the bleached specimens and the control group, with a medium level of gelatinolytic activity expressed in the dentin tubules. The results of gelatin zymography showed an increased expression of pro-MMP-9 in carbamide peroxide groups. The expression of pro-MMP-2 decreased in all the experimental groups. The bleaching treatments performed on the enamel of sound teeth do not influence dentinal enzymatic activity. However, when unprotected dentin tissue is bleached, matrix metalloproteinases are more expressed, particularly when carbamide peroxide is used, proportional to the exposure time.  相似文献

The problem of how “near” we can come to a n-coloring of a given graph is investigated. I.e., what is the minimum possible number of edges joining equicolored vertices if we color the vertices of a given graph with n colors. In its generality the problem of finding such an optimal color assignment to the vertices (given the graph and the number of colors) is NP-complete. For each graph G, however, colors can be assigned to the vertices in such a way that the number of offending edges is less than the total number of edges divided by the number of colors. Furthermore, an Ω(epn) deterministic algorithm for finding such an n-color assignment is exhibited where e is the number of edges and p is the number of vertices of the graph (e?p?n). A priori solutions for the minimal number of offending edges are given for complete graphs; similarly for equicolored Km in Kp and equicolored graphs in Kp.  相似文献
