Oligonucleotides modified by a 2′-deoxy-2′-(N-methoxyamino) ribonucleotide react readily with aldehydes in slightly acidic conditions to yield the corresponding N-(methoxy)oxazolidine-linked oligonucleotide-conjugates. The reaction is reversible and dynamic in slightly acidic conditions, while the products are virtually stable above pH 7, where the reaction is in a ‘‘switched off-state’’. Small molecular examinations have demonstrated that aldehyde constituents affect the cleavage rate of the N-(methoxy)oxazolidine-linkage. This can be utilized to adjust the stability of this pH-responsive cleavable linker for drug delivery applications. In the present study, Fmoc-β-Ala-H was immobilized to a serine-modified ChemMatrix resin and used for the automated assembly of two peptidealdehydes and one aldehyde-modified peptide nucleic acid (PNA). In addition, a triantennary N-acetyl-d-galactosamine-cluster with a β-Ala-H unit has been synthesized. These aldehydes were conjugated via N-(methoxy)oxazolidine-linkage to therapeutically relevant oligonucleotide phosphorothioates and one DNA-aptamer in 19–47% isolated yields. The cleavage rates of the conjugates were studied in slightly acidic conditions. In addition to the diverse set of conjugates synthesized, these experiments and a comparison to published data demonstrate that the simple conversion of Gly-H to β-Ala-H residue resulted in a faster cleavage of the N-(methoxy)oxazolidine-linker at pH 5, being comparable (T0.5 ca 7 h) to hydrazone-based structures.  相似文献

Numerical time-domain-diffusion simulations were used for studying the diffusion behavior of tracer molecules in rock matrix with homogeneous and heterogeneous porosity. As the heterogeneous sample in these simulations, a 3D tomographic image of altered tonalite was used, in which the mineral components and the pores resolved by X-ray microtomography were represented by their respective intragranular porosities determined previously by the 14C-PMMA method. The apparent diffusion coefficient of a tracer in altered tonalite was determined experimentally, and was then used in the simulations. In the altered tonalite analyzed, inclusion of heterogeneity in the porosity increased the diffusion coefficient by 16 %. Altered and pristine feldspar was the main mineral component in the sample (72 %), and it also provided the dominant contribution to tracer diffusion, explaining alone 52 % of the diffusion coefficient. The large pores resolved by microtomography (6 %) and altered and pristine mica (22 %) gave an equal contribution to the diffusion coefficient. The simulation method applied was also validated by comparing the results to both an analytical and a numerical solution to the diffusion equation in a homogenous medium. In addition, the method was compared to discrete-time random-walk simulations in the case of randomly placed overlapping spheres.  相似文献

Magnetic resonance first-pass imaging of a bolus of contrast agent is well adapted to distinguish normal and hypoperfused areas of the myocardium. In most cases, the signal intensity-time curves in user defined regions of interest (ROI) are studied. As image acquisition is ECG-gated, the images are acquired at the same moment in the cardiac cycle, and the basic shape of the heart is similar from one view to the next. However, superficial respiratory motion can displace the heart in the short-axis plane. The aim of this study is to correct for the respiratory motion of the heart by performing an automatic realignment of the myocardial ROI based on a method tracking the movement of the lung-myocardium interface. Visual and quantitative analyses performed on 120 curves show a very good concordance between two automatic methods and the manual one.  相似文献

The suitability of ion-selective electrode for the determination of ammonia in pyrolysis gases of fossil fuels was studied. The ammonia was absorbed into acidic solution and two kinds of determination methods were carried out. The ammonia was either measured directly from the acid solution, or ammonia was first released into the gas phase and then determined (air gap method) by the ammonia selective electrode. The electrode functioned well in both cases, but the linear calibration range was rather narrow, slightly more than one tenfold. The quantitative detection limit in the water phase was 5 x 10(-6)M (0.085 ppm) NH(3) and in gas phase operation solutions above 5 x 10(-4)M (8.5 ppm) NH(3) it was possible to measure quantitatively. The applications were carried out with Finnish energy peat samples and a coal sample.  相似文献

Studies of the release of critical ash-forming elements from combustion of biomass are typically conducted with small sample masses under well controlled conditions. In biomass combustion on a grate, secondary recapture and release reactions in the fuel-bed may affect the overall release and partitioning of these elements. Earlier work by the authors on the release of K, Cl, and S from a high-chlorine biomass (corn stover) in a lab-scale setup is, in the present work, supplemented with novel results from a bench-scale fixed bed reactor and a 100 kW moving grate pilot facility. The results from the bench-scale reactor indicate that S and K release are not significantly affected by secondary reactions, while Cl is partly recaptured by secondary reactions in the char. A linear increase in K-release was observed from 50% at 906 °C to almost 80 wt.% at 1234 °C when firing only corn stover. A similar release profile was observed for Cl, from 65% to nearly 100%. Complete release of S was achieved at 1234 °C with a linear increase from 70% at 906 °C. Co-combustion of corn stover with low-Cl wood chips served to increase the bed temperature, resulting in complete and close to complete release of Cl and S, respectively. An increase in the relative K-release was observed when increasing the wood chip fraction from 40% to 100% (energy basis). Pilot scale flue gas results indicate that the share of Cl released as HCl decreases towards 0% as the share of wood chips is increased towards 100%. Hence, co-combustion of corn stover with wood chips is expected to decrease the absolute release of KCl due to the lower feedstock quantity of Cl, however, increase the relative release of Cl as KCl.  相似文献
