Selective chemical reactions that are orthogonal to the diverse functionality of biological systems have become important tools in the field of chemical biology. Two notable examples are the Staudinger ligation of azides and phosphines and the Cu(I)-catalyzed [3 + 2] cycloaddition of azides and alkynes ("click chemistry"). The Staudinger ligation has sufficient biocompatibility for performance in living animals but suffers from phosphine oxidation and synthetic challenges. Click chemistry obviates the requirement of phosphines, but the Cu(I) catalyst is toxic to cells, thereby precluding in vivo applications. Here we present a strain-promoted [3 + 2] cycloaddition between cyclooctynes and azides that proceeds under physiological conditions without the need for a catalyst. The utility of the reaction was demonstrated by selective modification of biomolecules in vitro and on living cells, with no apparent toxicity. 相似文献
We describe a phase retrieval approach for intensity point-spread functions of high-numerical-aperture optical systems such as light microscopes. The method calculates a generalized pupil function defined on a spherical shell, using measured images at several defocus levels. The resultant pupil functionsreproduce measured point-source images significantly better than does an ideal imaging model. Availability of pupil function information will facilitate new approaches to aberration correction in such systems. 相似文献
Measurements of synchrotron radiation emitted by 30-MeV runaway electrons in the TEXTOR-94 tokamak show that the runaway population decays after switching on neutral beam injection (NBI). The decay starts only with a significant delay, which decreases with increasing NBI heating power. This delay provides direct evidence of the energy dependence of runaway confinement, which is expected if magnetic modes govern the loss of runaways. Application of the theory by Mynick and Strachan [Phys. Fluids 24, 695 (1981)] yields estimates for the "mode width" (delta) of magnetic perturbations: delta<0.5 cm in Ohmic discharges, increasing to delta = 4.4 cm for 0. 6 MW NBI. 相似文献

Summary: Coexisting bicontinuous morphologies, one ordered and one disordered, are investigated in a macrophase‐separated poly(styrene‐block‐isoprene) diblock copolymer/homopolystyrene (SI/hS) blend. Two‐phase behavior is attributed to the relatively high hS/S mass ratio (0.92). According to its crystallographic signature and channel coordination as discerned from three‐dimensional (3D) images generated by transmission electron microtomography (TEMT), the ordered morphology is classified as gyroid. The 3D local and global topological characteristics of both bicontinuous morphologies as measured directly from TEMT images are reported. The disordered morphology is further compared with molecular‐field simulations to ascertain the spatial distribution of the constituent species within the blend, thereby demonstrating the utility of high‐resolution 3D imaging coupled with molecular‐level simulations.
Incorporation of a block copolymer into a thin polymer film is observed to alter both the rate and mechanism by which the film dewets from an immiscible polymer substrate. Films with little or no copolymer dewet by classical nucleation and growth of circular holes, and the dewetting rate decreases with increasing copolymer concentration. Increasing the copolymer content at constant film thickness generates copolymer micelles that adsorb/aggregate along the polymer/polymer interface and promote nonclassical dewetting fluctuations similar in appearance to spinodal dewetting. At higher copolymer concentrations, dewetting proceeds after a lengthy induction period by the nucleation and growth of flower-shaped holes suggestive of film pinning or viscous fingering. Atomic force microscopy of the polymer/polymer interface after removal of the top film by selective dissolution reveals substantial structural development due to copolymer self-assembly. 相似文献
