The novel NAD+-linked opine dehydrogenase from a soil isolate Arthrobacter sp. strain 1C belongs to an enzyme superfamily whose members exhibit quite diverse substrate specificites. Crystals of this opine dehydrogenase, obtained in the presence or absence of co-factor and substrates, have been shown to diffract to beyond 1.8 ? resolution. X-ray precession photographs have established that the crystals belong to space group P21212, with cell parameters a = 104.9, b = 80.0, c = 45.5 ? and a single subunit in the asymmetric unit. The elucidation of the three-dimensional structure of this enzyme will provide a structural framework for this novel class of dehydrogenases to enable a comparison to be made with other enzyme families and also as the basis for mutagenesis experiments directed towards the production of natural and synthetic opine-type compounds containing two chiral centres.  相似文献

We investigate the influence of isotopic substitution and solvation of N-methylacetamide (NMA) on anharmonic vibrational coupling and vibrational relaxation of the amide I and amide II modes. Differences in the anharmonic potential of isotopic derivatives of NMA in D2O and DMSO-d6 are quantified by extraction of the anharmonic parameters and the transition dipole moment angles from cross-peaks in the two-dimensional infrared (2D-IR) spectra. To interpret the effects of isotopic substitution and solvent interaction on the anharmonic potential, density functional theory and potential energy distribution calculations are performed. It is shown that the origin of anharmonic variation arises from differing local mode contributions to the normal modes of the NMA isotopologues, particularly in amide II. The time domain manifestation of the coupling is the coherent exchange of excitation between amide modes seen as the quantum beats in femtosecond pump-probes. The biphasic behavior of population relaxation of the pump-probe and 2D-IR experiments can be understood by the rapid exchange of strongly coupled modes within the peptide backbone, followed by picosecond dissipation into weakly coupled modes of the bath.  相似文献

Here are reported composites made by crosslinking unsaturated units in canola, sunflower, or linseed oil with sulfur to yield CanS , SunS , and LinS , respectively. These plant oils were selected because the average number of crosslinkable unsaturated units per triglyceride vary from 1.3 for canola to 1.5 for sunflower and 1.8 for linseed oil. The remeltable composites show compressive strengths that increase with increasing unsaturation number from CanS (9.3 MPa) to SunS (17.9 MPa) to LinS (22.9 MPa). These values for SunS and LinS are competitive when compared with the value of 17 MPa required for residential building using traditional Portland cement. The plant oil composites are recyclable over many cycles and can retain up to 100% of strength after 24 hr in oxidizing acid under conditions where Portland cement is dissolved in under 30 min. Infusion of the composites into premade cement blocks affords them with significantly improved acid resistance as well. This work thus provides a simple, nearly 100% atom economical route to convert plant oils and waste sulfur to composites having enhanced performance over commercial structural materials.  相似文献

The practical application of adsorbents with the desired separation properties depends not only upon the adsorption characteristics of the material but also upon the mechanical properties of the packed bed. The packed bed, the vessel surrounding the bed and any internal structure that supports the bed are subjected to both static and cyclic loads during an adsorption process. In order to properly design the vessel and its internal structure, the bulk mechanical properties (most particularly the elastic properties) of the adsorbent bed must be known. The primary focus of this study was to determine the elastic properties of adsorbent beds packed with activated alumina, synthetic molecular sieve 13X or natural zeolite clinoptilolite. The bulk modulus of elasticity was found to be a linear function of applied stress for each of these materials in a range of stresses lower than the bulk crush strength. The Poisson’s ratio for the packed bed was also deduced from these results.  相似文献

Intercalates of o-, m-, and p-toluidine into α-Zr(HPO4)2 · H2O were prepared and characterized by powder X-ray diffraction, thermogravimetric analysis and infrared spectroscopy. As follows from IR, toludine molecules are protonated in the interlayer space. Toluidine molecules are arranged in a bimolecular way in the intercalates containing more than 1.5 toluidine molecules per Zr atom. On the other hand, a monolayer of the toluidine molecules is supposed in the intercalates with less than one toluidine molecule per Zr atom.  相似文献

The free energy as a function of the reaction coordinate (rc) is the key quantity for the computation of equilibrium and kinetic quantities. When it is considered as the potential of mean force, the problem is the calculation of the mean force for given values of the rc. We reinvestigate the PMCF (potential of mean constraint force) method which applies a constraint to the rc to compute the mean force as the mean negative constraint force and a metric tensor correction. The latter allows for the constraint imposed to the rc and possible artefacts due to multiple constraints of other variables which for practical reasons are often used in numerical simulations. Two main results are obtained that are of theoretical and practical interest. First, the correction term is given a very concise and simple shape which facilitates its interpretation and evaluation. Secondly, a theorem describes various rcs and possible combinations with constraints that can be used without introducing any correction to the constraint force. The results facilitate the computation of free energy by molecular dynamics simulations.  相似文献
