Summary A novel approach to solid phase extraction, based on the use of a highly selective molecularly imprinted polymer, is presented.
The versatility of this type of sorbent for solid phase extraction was demonstrated in a model batch-wise pre-concentration
of sameridine prior to gas chromatography. Problems associated with leakage of remaining imprint molecules during the desorption
phase could be eliminated by the use of a close structural analogue of sameridine as the imprint species. It was found that
a major benefit of the imprinted polymer was its specificity, which lead to distinctly cleaner chromatographic traces and

The stability of Ochratoxin A during its extraction using different extraction techniques has been evaluated. Microwave-assisted extraction and pressurised liquid extraction, in addition to two other reference methods of extraction, i.e. ultrasound-assisted and magnetic stirring-assisted extraction, were evaluated. The effect of extraction temperature using the cited techniques was checked.The results show that Ochratoxin A can be extracted using microwave-assisted extraction at temperatures up to 150 °C without degradation. Pressurised liquid extraction can be used at temperatures up to 100 °C, for extraction times of less than 30 min. Further, both ultrasound-assisted extraction and magnetic stirring extraction can be applied at temperatures up to 65 °C.High-performance liquid chromatography combined with fluorescence detection using a Chromolith RP-18e column at a flow rate of 5 mL min−1 was used to quantify the Ochratoxin A. The retention time for the Ochratoxin A was 1.3 min. The limits of detection (LOD) and of quantification (LOQ) were 0.03 and 0.10 μg L−1, respectively. 相似文献

通过PCR扩增出猪传染性胃肠炎病毒(TGEV)聚合酶部分片段,反向插入逆转录病毒表达载体pLXSN中,用脂质体法将重组质粒plxas-pol转染PA317细胞,经抗生素G418筛选出稳定的产毒细胞克隆,分别扩大培养后,取其上清液感染小鼠成纤维细胞NIH3T3,细胞克隆产生的重组病毒效价达9.0×105CFU/mL。用高效价假病毒感染IBRS2细胞,再经抗生素G418筛选,提取细胞克隆总RNA,经RT-PCR证明plxas-pol整合入IBRS2细胞。以TGEV感染IBRS2细胞和具有抗性的IBRS2细胞所产生的细胞病变为指标,证明该反义RNA对病毒有明显抑制作用,抑制率约为80%。用2×103和4×102TCID50/mL剂量的TGEV感染时,引起细胞死亡的时间分别为21 h和27 h,与对照组相比,抗性细胞系可明显延迟因病毒感染引起细胞死亡的时间。 相似文献

Uptake of trace elements into fish otoliths is governed by several factors such as life histories and environment in addition to stock and species differences. In an attempt to elucidate the elemental signatures of rare earth elements (REEs) in otoliths, a solid phase extraction (SPE) protocol was used in combination with electrothermal vaporization (ETV) as a sample introduction procedure for the determinations by inductively coupled plasma quadrupole mass spectrometry (ICP-MS). Effects of various parameters, such as carrier gas flow rate, atomization temperature and chemical modification, were examined for optimization of the conditions by ETV-ICP-MS. Atomization was achieved at 2800 °C. Lower temperatures (i.e. 2600 °C) resulted in severe memory problems due to incomplete atomization. Palladium was used as a chemical modifier. It was found that an increase in Pd concentration up to 0.5 μg in the injection volume (70 μl) led up to four-fold enhancement in the integrated signals. This phenomenon is attributed to the carrier effect of Pd rather than the stabilization since no significant losses were observed for high temperature drying around 700 °C even in the absence of Pd. Preconcentration was performed on-line at pH 5 by using a mini-column of Toyopearl AF-Chelate 650M chelating resin, which also eliminated the calcium matrix of otolith solutions. After preconcentration of 6.4 ml of solution, the concentrate was collected in 0.65 ml of 0.5% (v/v) HNO3 in autosampler cups, and then analyzed by ETV-ICP-MS. The method was validated with the analysis of a fish otolith certified reference material (CRM) of emperor snapper, and then applied to samples. Results obtained from otoliths of fish captured in the same habitat indicated that otolith rare earth element concentrations are more dependent on environmental conditions of the habitat than on species differences. 相似文献

In the present paper a procedure is proposed for the determination of traces of Cd, Co, Mn and Cr in petroleum industry produced water by inductively coupled plasma optical emission spectrometry. The procedure is based on cloud point extraction of these metals, as their dithizonate complexes, into the surfactant-rich phase of octylphenoxypolyethoxyethanol surfactant (Triton X-114). Extractions were carried out in solutions with salinities between 10‰ and 70‰. Since residual salinity in the surfactant-rich phase caused differences in its transport to the plasma, yttrium was used as an internal standard to correct for this effect. The simultaneous metal extraction procedure was optimized by response surface methodology using a Doehlert design and desirability function. Enhancement factors of 21, 21, 9 and 19, along with limits of quantification of 0.093, 0.20, 0.73 and 1.2 μg L− 1, and precision expressed as relative standard deviation (n = 8, 20.0 μg L− 1) of 5.8, 1.2, 1.7 and 5.7% were obtained for Cd, Co, Mn and Cr, respectively. There is a high correlation between molecular surface area (TSA) of triorganotin and triorganolead compounds and their toxicity towards a bacterium (Escherichia coli) and an alga (Selenastrum capricornutum). Parallel attempts to correlate other Group IVA organometals incorporating silicon or germanium were unsuccessful. It was further demonstrated, however, that a high correlation was obtainable between certain series of compounds with the same organic substituent but different metal centers involving all Group IVA elements. In both instances, the inability to obtain a quantitative structure-activity relationship (QSAR) for all systems studied appears to be a function of the solubility of the compounds. While organotin TSA values have been found to correlate well with their toxicities toward various organisms, this study clearly suggests that this type of QSAR can be readily extended to include other organometal systems, provided that there is no solubility problem and the toxicity is a function of the hydrophobicity of the organometal compounds. 相似文献
