为探讨平阳霉素碘油乳剂(PLE)治疗肝海绵状血管瘤(HCH)的临床疗效及并发症的防治,对22例经CT平扫及增强证实的肝海绵状血管瘤,作超选择肝动脉插管,灌注PLE,然后用明胶海绵栓塞.结果表明,介入治疗后病人症状改善,病灶有不同程度缩小.其中18例3个月缩小50%以上,15例1年B超复查瘤体消失.认为经肝动脉灌注PLE是治疗HCH安全有效的方法.  相似文献

采用亲水相互作用色谱-四极杆/静电场轨道阱高分辨质谱系统,建立了肝组织中胶原蛋白水解物羟脯氨酸(Hyp)的快速定量检测方法。将正常及四氯化碳肝纤维化模型小鼠的肝组织样品酸水解,经过滤、稀释后,采用Hypersil GOLD HILIC色谱柱(100 mm×2.1 mm,3μm)分离,以水-乙腈(28∶72,v/v)为流动相进行等度洗脱,最后用配有电喷雾离子源的四极杆/静电场轨道阱高分辨质谱在正离子模式下进行检测。结果表明,Hyp在0.78~100.00μg/L范围内呈良好的线性关系,相关系数(R2)为0.998 3,方法的定量限为0.78μg/L,样品加标回收率为97.4%~100.9%,相对标准偏差为1.4%~2.0%(n=6)。此外,该方法与传统的氯胺T法进行比较,发现两种方法的检测结果相关性良好,Pearson相关系数为0.927;较氯胺T法,该法具有操作简便、准确度高的优点。该方法可用于肝组织中Hyp的快速定量分析。  相似文献

G-actin prepared from pork liver and purified to give a product of 95–98% purity was polymerized to F-actin in a rotational dynamic viscometer. The rheological properties were investigated during the course of polymerisation and after polymerisation. G andG measured at frequencies around 0.1 Hz increased strongly during the first 3 hours of the reaction and then slowly approached a constant value in the range of 10–50 mPa. The rise during polymerisation was more than two orders of magnitude. When a solution of F-actin was subjected to steady shear at a high shear rate for a short period of time and subsequently dynamic measurements were performed, then a considerably smallerG (about 70% smaller) was observed immediately after cessation of the steady shear.G then increased with time and approached its initial value.When measuring the viscosity of F-actin solutions as a function of shear rate, a strong shear thinning effect was seen which did not vanish even at shear rates as low as 0.001 s–1. Even at this low shear rates, a pronounced yield maximum was observed before reaching the steady state. Oscillatory experiments showed a remarkably weak frequency dependence ofG. The results imply that F-actin solutions are largely structured forming a weak temporary network which can be easily destroyed by application of high shear rates. It seems most likely that the destruction occurs by a reversible, shear induced depolymerisation process.  相似文献

建立了一种同时测定阿散酸、亚砷酸根、砷酸根、砷胆碱、砷甜菜碱、一甲基砷和二甲基砷等7种砷形态的高效液相色谱-电感耦合等离子体质谱分析方法。样品采用人工胃液作为提取液进行超声处理,再以碳酸铵溶液和水作为流动相,采用阴离子分析柱将样品提取液进行分离,最后进入电感耦合等离子体质谱测定。各砷形态在1~50 μg/kg范围内线性关系良好,相关系数r2均在0.999以上;在1、2、10 μg/kg 3个添加水平进行了方法学验证,平均回收率为84.3%~106.6%,相对标准偏差为1.4%~4.2%; 7种砷形态的定量限均为1 μg/kg。方法重现性好、灵敏度高、前处理简单,适用于鸡肉和鸡肝中主要有机砷和无机砷残留的分析检测。  相似文献

采用超高效液相色谱-串联质谱(UPLC-MS/MS)在多反应监测(MRM)模式下建立了鱼肉、鱼肝中6种抗雌激素类药物(托瑞米芬、氯米芬、他莫昔芬、雷洛昔芬、阿那曲唑、来曲唑)的检测方法。样品采用乙腈超声提取,上清液中加入适量水稀释提取液,经混合型阳离子交换(MCX)固相萃取柱富集、净化后进行UPLC-MS/MS分析。UPLC分离在ACQUITY UPLCTM BEH C18柱(100 mm×2.1 mm, 1.7 μm)上进行,以乙腈和0.1%甲酸水溶液为流动相进行梯度洗脱。6种抗雌激素药物的定量限(以信噪比为10计)为0.1~0.3 μg/kg; 4个加标水平的平均回收率为84.9%~112.2%,相对标准偏差为0.9%~14.3%。该方法可用于鱼肉、鱼肝中6种抗雌激素药物的痕量分析。  相似文献

We describe a selective pressurized liquid extraction (SPLE) method, followed by gas chromatography–mass spectrometry (GC–MS), for the simultaneous extraction and clean-up of polybrominated diphenyl ethers (PBDEs) and polychlorinated biphenyls (PCBs) in sheep liver tissue samples. The on-line clean-up of liver tissue by SPLE was tested using differing amount of acid-modified silica (sulphuric acid:silica gel, 1:2, w/w), the most effective amount being 20 g. Different extraction solvents (iso-hexane and dichloromethane), either alone or in various combinations, were used to extract these target compounds from spiked liver samples. Variables affecting the SPLE extraction efficiency, including temperature, pressure, number of extraction cycles and static extraction time were studied; the optimum parameters were 80 °C, 10.3 MPa, 2 cycles and 5 min, respectively. The SPLE based method was compared with more traditional Soxhlet, off-line PLE, ultrasonic and heating extraction methods. Overall the mean percentage recoveries for all target chemicals using SPLE were 86–103% (n = 3, SD < 9%), and compared favourably with the Soxhlet (63–109%, n = 3, SD < 8%), off-line PLE (82–104%, n = 3, SD < 18%), ultrasonic (86–99%, n = 3, SD < 11%) and heating (72–102%, n = 3, SD < 21%) extraction methods. The limits of detection of the proposed method were 5–96 pg g−1 and 2–29 pg g−1 for the different PBDE and PCB chemicals studied, respectively. The outputs of the proposed method were linear over the range from 0.02 to 30 ng g−1, for all PCB and PBDE congeners except for PBDE 100 and 153 (0.05–30 ng g−1) and PBDE 183 (0.1–30 ng g−1). The method was successfully applied to sheep liver samples for the determination of the target PBDE and PCB compounds.  相似文献

The development of clinically applicable scaffolds is important for the application of cell transplantation in various human diseases. The aims of this study are to evaluate fibrin glue in a novel protein replacement therapy using proliferative adipocytes and to develop a mouse model system to monitor the delivery of the transgene product into the blood and the fate of the transduced cells after transplantation. Proliferative adipocytes from mouse adipose tissue were transduced by a retroviral vector harboring the human lecithin-cholesterol acyltransferase (lcat) gene, and were subcutaneously transplanted into mice combined with fibrin glue. The lcat gene transduction efficiency and the subsequent secretion of the product in mouse adipocytes were enhanced using a protamine concentration of 500 μg/ml. Adipogenesis induction did not significantly affect the lcat gene-transduced cell survival after transplantation. Immunohistochemistry showed the ectopic enzyme production to persist for 28 days in the subcutaneously transplanted gene- transduced adipocytes. The increased viability of transplanted cells with fibrin glue was accompanied with the decrease in apoptotic cell death. The immunodetectable serum LCAT levels in mice implanted with the fibrin glue were comparable with those observed in mice implanted with Matrigel, indicating that the transplanted lcat gene-transduced adipocytes survived and functioned in the transplanted spaces with fibrin glue as well as with Matrigel for 28 days. Thus, this in vivo system using fibrin is expected to serve as a good model to further improve the transplanted cell/scaffold conditions for the stable and durable cell-based replacement of defective proteins in patients with LCAT deficiency.  相似文献

兔肝VX2肿瘤是一种快速生长的肿瘤模型,可以在多种器官如肝、肺、直肠等快速生长,常用于肿瘤研究。采用可见-近红外高光谱技术对四只兔子的兔肝VX2肿瘤和正常组织进行活体和离体的反射光谱检测,然后采用支持向量机分别实现了二分类(正常肝组织和肝VX2肿瘤组织)和四分类(未出血活体正常肝组织、未出血活体VX2肿瘤组织、出血离体正常肝组织和出血离体肝VX2肿瘤组织)。根据其光谱反射曲线的特征,选择了400~1 800 nm区间的数据为特征变量。为进一步提高分类准确率,分别采用5折交叉验证和遗传算法对支持向量机的核函数参数g和惩罚因子c进行了优化。其中5折交叉验证优化参数和分类结果为:二分类优化的惩罚参数c为4,核函数参数g为0.125 0,其校正集和预测集的准确率都达到了100%;四分类中优化出的参数c为8,g为0.121 1,其校正集和预测集的准确率分别达到了99.242 4%和93.333%。遗传算法优化参数和结果为:二分类中优化的参数c为0.845 6,g为0.062 5,其校正集和预测集的准确率同样都达到了100%;四分类中优化的参数c为5.5307,g为0.068 5,其校正集和预测集的准确率分别达到了99.242 4%和100%。结果显示两种优化方法都取得了很好的效果,遗传算法优化参数对四分类的分类更为精确。为进一步提升算法速度,采用间隔选取变量的方法来不断减少特征变量,最终每隔100 nm谱段选择一个变量,共选择14个谱段作为特征变量。采用遗传算法优化支持向量机参数并对其分类进行了研究,结果表明:二分类和四分类的校正集和预测集结果准确率均为99.242 4%,而且运行时间分别为11.4和20.0 s, 与选择全波段的运行时间:340.3和491.0 s相比, 说明多光谱技术可以进行肝VX2肿瘤组织和正常肝组织的鉴别,且分类准确率可达99%以上,而且运行时间缩短了很多。为未来多光谱技术在未来临床肿瘤诊断中实现肿瘤组织的快速实时在线检测和分类奠定了基础,显示出巨大的应用潜力。  相似文献

应用蚕豆根尖微核技术研究了肝癌高发区的扶绥县饮用水的诱变性与肝癌发病的关系。结果表明,检测32份不同饮水类型的饮用水(饮用塘水、塘边井水、井水、河水和自来水)中有12份饮用塘水和2份塘边井水能明显诱导蚕豆根尖核率增高(P〈0.01),而井水、河水和自来水所诱发的蚕豆根尖微核率与对照组则无显著差异(P〉0.05)。提示该县饮用塘水中存在染色体畸变的物质,此类物质在各种饮水类型的水样中所诱发的蚕豆根尖  相似文献
