Many pathologies affecting muscles (muscular dystrophies, sarcopenia, cachexia, renal insufficiency, obesity, diabetes type 2, etc.) are now clearly linked to mechanisms involving oxidative stress. In this context, there is a growing interest in exploring plants to find new natural antioxidants to prevent the appearance and the development of these muscle disorders. In this study, we investigated the antioxidant properties of Arctium lappa leaves in a model of primary human muscle cells exposed to H2O2 oxidative stress. We identified using bioassay-guided purification, onopordopicrin, a sesquiterpene lactone as the main molecule responsible for the antioxidant activity of A. lappa leaf extract. According to our findings, onopordopicrin inhibited the H2O2-mediated loss of muscle cell viability, by limiting the production of free radicals and abolishing DNA cellular damages. Moreover, we showed that onopordopicrin promoted the expression of the nuclear factor-erythroid-2-related factor 2 (Nrf2) downstream target protein heme oxygenase-1 (HO-1) in muscle cells. By using siRNA, we demonstrated that the inhibition of the expression of Nrf2 reduced the protective effect of onopordopicrin, indicating that the activation of the Nrf2/HO-1 signaling pathway mediates the antioxidant effect of onopordopicrin in primary human muscle cells. Therefore, our results suggest that onopordopicrin may be a potential therapeutic molecule to fight against oxidative stress in pathological specific muscle disorders.  相似文献

In the confetti percolation model, or two‐coloured dead leaves model, radius one disks arrive on the plane according to a space‐time Poisson process. Each disk is coloured black with probability p and white with probability . In this paper we show that the critical probability for confetti percolation equals 1/2. That is, if p > 1/2 then a.s. there is an unbounded curve in the plane all of whose points are black; while if then a.s. all connected components of the set of black points are bounded. This answers a question of Benjamini and Schramm [1]. Considerable attention has been paid to the study of green tea leaves because of their high consume and beneficial effects on human health. In this work, an appropriate strategy is proposed to investigate and resolve the major metabolites extracted from Camellia sinensis tea leaves. Statistical design mixtures of ethanol, ethyl acetate, dichloromethane, and chloroform were used to study the effects of different solvents and their mixtures on the extraction of the secondary metabolites of C. sinensis tea leaves from two different harvest seasons. Extracted samples were analyzed by high performance liquid chromatography‐diode array detection‐electrospray ionization mass spectrometer allowing the resolution of a large amount of tea metabolites with high relative abundances, especially when their extraction was performed in pure ethanol and with solvent mixtures with ethanol. Resolution of the more relevant metabolites was achieved by the simultaneous analysis of the fused diode array detection and mass spectrometer detectors data from the same samples using the multivariate curve resolution‐alternating least squares chemometric method. Peak areas finally resolved were further analyzed by orthogonal signal correction and partial least squares‐discrimination analysis to discriminate among C. sinensis tea samples. Using the Variable Importance in Projection variable selection method, epigallocatechin and caffeine were finally selected as the two more important chemical constituents of tea leaves that were discriminating more between the tea samples from two different harvest seasons. The chemical composition of the essential oil from the leaves of Eucalyptus camaldulensis, Eucalyptus camaldulensis var. obtusa and Eucalyptus gomphocephala grown in northern Egypt was analysed by using GC-FID and GC–MS techniques. The antibacterial (agar disc diffusion and minimum inhibitory concentration methods) and antioxidant activities (2,2′-diphenypicrylhydrazyl) were examined. The main oils constituents were 1,8-cineole (21.75%), β-pinene (20.51%) and methyleugenol (6.10%) in E. camaldulensis; spathulenol (37.46%), p-cymene (17.20%) and crypton (8.88%) in E. gomphocephala; spathulenol (18.37%), p-cymene (19.38%) and crypton (16.91%) in E. camaldulensis var. obtusa. The essential oils from the leaves of Eucalyptus spp. exhibited considerable antibacterial activity against Gram-positive and Gram-negative bacteria. The values of total antioxidant activity were 70 ± 3.13%, 50 ± 3.34% and 84 ± 4.64% for E. camaldulensis, E. camaldulensis var. obtusa and E. gomphocephala, respectively. The highest antioxidant activity value of 84 ± 4.64% could be attributed to the high amount of spathulenol (37.46%).  相似文献

Sixteen subsequent fractions were prepared from the ethyl acetate fraction of Zanthoxylum bungeanum leaves after bio-guided chromatographic separation. The HPLC profiles and antioxidant activity of the various fractions indicated that the content of eight phenolic compounds (chlorogenic acid, epicatechin, rutin, hyperoside, trifolin, quercitrin, afzelin and quercetin) and antioxidant activity vary significantly, and high concentrations of a combination of eight phenolic compounds would result in an increase of the antioxidant activity. These results suggested that the eight compounds could be used as chemical markers for quality assessment of Z. bungeanum leaves. Correlation between chromatographic fingerprint and antioxidant activity of the fractions showed that quercitrin and hyperoside play crucial roles in the antioxidant activity, and they can be seen as the milestone for quality control. The findings also suggested that five obtained fractions (E-3-3, E-2-4, E-7, E-5 and E-4) could become useful supplements for functional food ingredients and health-related products.  相似文献

A method was developed for quantifying 17 amino acids in tobacco leaves by using an A300 amino acid analyzer and chemometric resolution. In the method, amino acids were eluted by the buffer solution on an ion‐exchange column. After reacting with ninhydrin, the derivatives of amino acids were detected by ultraviolet detection. Most amino acids are separated by the elution program. However, five peaks of the derivatives are still overlapping. A non‐negative immune algorithm was employed to extract the profiles of the derivatives from the overlapping signals, and then peak areas were adopted for quantitative analysis of the amino acids. The method was validated by the determination of amino acids in tobacco leaves. The relative standard deviations (n = 5) are all less than 2.54% and the recoveries of the spiked samples are in a range of 94.62–108.21%. The feasibility of the method was proved by analyzing the 17 amino acids in 30 tobacco leaf samples.  相似文献

开展了杨树叶片的飞秒激光等离子体丝诱导击穿光谱研究, 定性比较分析了长春市区的第一汽车厂、火车站、净月潭公园及长春理工大学四个地理区域的杨树叶片中重金属元素Ca, Fe和Cr. 实验结果表明, 通过分析杨树叶片中Ca Ⅱ 393.37 nm和Fe Ⅰ 422.87 nm光谱谱线可知叶茎中Ca和Fe元素浓度均高于叶肉. 比较长春四个地理区域的杨树叶的飞秒激光等离子体丝诱导击穿光谱, 发现汽车厂附近的杨树叶内重金属Ca, Fe和Cr 元素浓度最高, 净月公园的杨树叶重金属浓度最低. 由于飞秒激光等离子体丝光强度的“光学钳箍”效应, 对于杨树叶片这种表面不平整样品, 仍可获得稳定性较好的等离子体光谱. 飞秒激光等离子体丝诱导光谱技术有望在环境污染在线检测具有广泛的潜在应用.  相似文献

In this paper, we prove that an m-connected graph G on n vertices has a spanning tree with at most k leaves (for k ≥ 2 and m ≥ 1) if every independent set of G with cardinality m + k contains at least one pair of vertices with degree sum at least nk + 1. The aim of this study was to analyse the effect of spontaneous microbial maceration on the release and extraction of the flavonoids and phenolics from olive leaves. Bioprofiling based on thin-layer chromatography effect-directed detection followed by ATR-FTIR spectroscopy proved to be a reliable and convenient method for simultaneous comparison of the extracts. Results show that fermentation significantly enhances the extraction of phenolic compounds and flavonoids. The polyphenolic content was increased from 6.7 µg GAE (gallic acid equivalents) to 25.5 µg GAE, antioxidants from 10.3 µg GAE to 25.3 µg GAE, and flavonoid content from 42 µg RE (rutin equivalents) to 238 µg RE per 20 µL of extract. Increased antioxidant activity of fermented ethyl acetate extracts was attributed to the higher concentration of extracted flavonoids and phenolic terpenoids, while increased antioxidant activity in fermented ethanol extract was due to increased extraction of flavonoids as extraction of phenolic compounds was not improved. Lactic acid that is released during fermentation and glycine present in the olive leaves form a natural deep eutectic solvent (NADES) with significantly increased solubility for flavonoids.  相似文献
