A path-based timing optimization algorithm for buffer insertion and simultaneous sizing is proposed. Firstly, candidate buffer insertion location and buffer size for each branch in a given routing path were obtained via localized timing optimization. Then, through evaluating each potential insertion against design objectives, potential optimal buffer insertion locations and sizes for the whole routing tree were determined. Flavin-containing monooxygenase (FMO), like cytochrome P450 (CYP), is a monooxygenase that uses the reducing equivalents of NADPH to reduce one atom of molecular oxygen to water, while the other atom is used to oxidize the substrate. Recently, it was shown that some CYP isoforms have been subject to positive selection. However, it is unknown whether the highly conserved phase I detoxification enzyme, FMO, has undergone similar positive Darwinian selection. We used maximum-likelihood models of codon substitution, evolutionary fingerprinting, and cross species comparison to investigate the occurrence of adaptive evolution in FMO sequences. We used recent genomic data from a range of species, including vertebrates and invertebrates. We present the evidence for the occurrence of adaptive evolution in mammalian FMO 3, 4, 5, and fugu FMOs but not in mammalian FMO 1, FMO 2, frog FMOs, other fish FMOs and invertebrate FMOs. The sites under adaptive evolution were significantly associated with the insertion domain in mammalian FMO 5. We identified specific amino acid sites in FMOs 3–5 that are likely targets for selection based on the patterns of parallel amino acid change. The most likely role of adaptive evolution is the repair of mutations that permitted optimal NADP+ binding and improved catalytic efficiency. The occurrence of positive selection during the evolution of phase I detoxification enzymes such as FMOs 3–5 and fugu FMO suggests the occurrence of both high selection pressure acting on species within their unique habitats and significant changes in intensity and direction (forms of xenobiotics and drugs) resulting from changes in microhabitat and food. 相似文献

In the present work we investigated a novel triplet ground-state germylenoid HB=GeLiF as well as its insertion reactions with RH(R = F,OH and NH2) using the DFT B3LYP and QCISD methods for the first time.Geometry optimization calculations show that the triplet HB=GeLiF has three equilibrium structures,in which the four-membered ring structure is the most stable with the lowest energy.All mechanisms of the three insertion reactions of germylenoid HB=GeLiF with RH(R = F,OH,and NH2) are identical to each other.Based on the calculated results,it is concluded that under the same conditions the insertion reactions should occur easily in the order of H-F > H-OH > H-NH2.In THF solvent the insertion reactions get more difficult than in the gas phase. 相似文献

Neutral phosphidozirconocene complexes [Cp2Zr(PR2)Me] (Cp=cyclopentadienyl; 1a : R=cyclohexyl (Cy); 1b : R=mesityl (Mes); 1c : R=tBu) undergo insertion into the Zr?P bond by non‐enolisable carbonyl building blocks (O=CR′R′′), such as benzophenone, aldehydes, paraformaldehyde or CO2, to give [Cp2Zr(OCR′R′′PR2)Me] ( 3 – 7 ). Depending on the steric bulk around P, complexes 3 – 7 react with B(C6F5)3 to give O‐bridged cationic zirconocene dimers that display typical frustrated Lewis pair (FLP)/ambiphilic ligand behaviour. Thus, the reaction of {[Cp2Zr(μ‐OCHPhPCy2)][MeB(C6F5)3]}2 ( 10a ) with chalcone results in 1,4 addition of the Zr+/P FLP, whereas the reaction of {[Cp2Zr(μ‐OCHFcPCy2)][MeB(C6F5)3]}2 ( 11a ; Fc=(C5H4)CpFe) with [Pd(η3‐C3H5)Cl]2 yields the unique Zr?Fe?Pd trimetallic complex 13a , which has been characterised by XRD analysis. 相似文献
