Several studies indicate that substances synthesized by granulosa cells are capableof regulating oocyte activity. We have studied the effect of factors synthesized by gran-ulosa cells on tPA activity of denuded oocytes using a co- culture system. The resultsshow that an FSH- dependent factor(s) synthesized by granulosa cells (but not by theca-interstitial cells) is capable of stimulating tPA activity of denuded oocytes. Prion‐like transcellular spreading of tau in Alzheimer's Disease (AD) is mediated by tau binding to cell surface heparan sulfate (HS). However, the structural determinants for tau–HS interaction are not well understood. Microarray and SPR assays of structurally defined HS oligosaccharides show that a rare 3‐O‐sulfation (3‐O‐S) of HS significantly enhances tau binding. In Hs3st1?/? (HS 3‐O‐sulfotransferase‐1 knockout) cells, reduced 3‐O‐S levels of HS diminished both cell surface binding and internalization of tau. In a cell culture, the addition of a 3‐O‐S HS 12‐mer reduced both tau cell surface binding and cellular uptake. NMR titrations mapped 3‐O‐S binding sites to the microtubule binding repeat 2 (R2) and proline‐rich region 2 (PRR2) of tau. Tau is only the seventh protein currently known to recognize HS 3‐O‐sulfation. Our work demonstrates that this rare 3‐O‐sulfation enhances tau–HS binding and likely the transcellular spread of tau, providing a novel target for disease‐modifying treatment of AD and other tauopathies. 相似文献

Carbon corrosion at high anodic potentials is a major source of instability, especially in acidic electrolytes and impairs the long‐term functionality of electrodes. In‐depth investigation of carbon corrosion in alkaline environment by means of differential electrochemical mass spectrometry (DEMS) is prevented by the conversion of CO2 into CO32?. We report the adaptation of a DEMS system for online CO2 detection as the product of carbon corrosion in alkaline electrolytes. A new cell design allows for in situ acidification of the electrolyte to release initially dissolved CO32? as CO2 in front of the DEMS membrane and its subsequent detection by mass spectrometry. DEMS studies of a carbon‐supported nickel boride (NixB/C) catalyst and Vulcan XC 72 at high anodic potentials suggest protection of carbon in the presence of highly active oxygen evolution electrocatalysts. Small‐molecule probes for the in vitro imaging of KCa3.1 channel‐expressing cells were developed. Senicapoc, showing high affinity and selectivity for the KCa3.1 channels, was chosen as the targeting component. BODIPY dyes 15 – 20 were synthesized and connected by a CuI‐catalyzed azide–alkyne [3+2]cycloaddition with propargyl ether senicapoc derivative 8 , yielding fluorescently labeled ligands 21 – 26 . The dimethylpyrrole‐based imaging probes 25 and 26 allow staining of KCa3.1 channels in NSCLC cells. The specificity was shown by removing the punctate staining pattern by pre‐incubation with senicapoc. The density of KCa3.1 channels detected with 25 and by immunostaining was identical. The punctate structure of the labeled channels could also be observed in living cells. Molecular modeling showed binding of the senicapoc‐targeting component towards the binding site within the ion channel and orientation of the linker with the dye along the inner surface of the ion channel. 相似文献

设计了一种新的电化学氢化物发生装置, 用于测定不同价态的硒. 通过对传统的平板电解池的改进, 设计了全新的圆盘电解池并采用螺纹密封的方式, 极大地缩短了电解池的装配时间, 提高了电解池的使用寿命和密封性能. 结合热水浴在线还原技术, 将 Se(Ⅵ)在高浓度HCl介质中120 s内还原成Se(Ⅳ), 通过电化学氢化物发生技术生成氢化物, 成功的进行硒的在线价态分析. 对各种实验参数进行了详细的研究, Se(Ⅳ) 和Se(Ⅵ)的相对标准偏差分别为2.6% 和 3.1% (n=11);Se(Ⅳ)和总硒的检出限分别为0.32和0.27 μg/L (3σ). 相似文献

Pt-based alloy nanoporous structures have attracted a lot of attention because of their high activity and stability toward alcohol oxidation reactions. Especially, Pt alloying with Earth-abundant metal can lower the cost of catalyst. Here, we introduce a one-pot approach to synthesize bimetallic PtCu and Ni-doped PtCu nanoalloy with porous structure. The as-synthesized Ni-doped Pt60Ni3Cu37 nanoalloys exhibit excellent electrocatalytic properties toward methanol oxidation in acidic medium. The mass activity of the as-synthesized Pt60Ni3Cu37 nanoalloys is 3.6 times and 5.3 times that of Pt55Cu45 nanoalloys and commercial Pt black for methanol oxidation in 0.2?M methanol solution. Besides, the stability of the as-synthesized Pt60Ni3Cu37 nanoalloys was much better than Pt55Cu45 nanoalloys and commercial Pt black. After 3600?s chronoamperometry test, the remaining values of the Pt60Ni3Cu37 nanoalloys are 3.7 times and 11.0 times that of Pt55Cu45 nanoalloys and commercial Pt black. And it is the first time to report that small amount of Ni dopants can boost the activity and stability of PtNiCu alloys toward methanol oxidation. 相似文献
