HPLC with fluorescence detection assay of perampanel,a novel AMPA receptor antagonist,in human plasma for clinical pharmacokinetic studies

Perampanel (Fycompa®), a novel α‐amino‐3‐hydroxy‐5‐methyl‐4‐isoxazolepropionic acid (AMPA) receptor antagonist, is registered for the adjunctive treatment of patients (aged ≥12 years) with refractory partial‐onset seizures. To support therapeutic drug monitoring, a simple high‐performance liquid chromatography (HPLC) assay with fluorescence detection was developed to determine perampanel concentrations in human plasma and validated to support clinical trials. Human plasma samples (1.0 mL) were processed by liquid extraction using diethyl ether, followed by chromatographic separation on a YMC Pack Pro C₁₈ column (150 × 4.6 mm i.d., 5 µm) with isocratic elution of acetonitrile–water–acetic acid–sodium acetate (840:560:3:1.8, v/v/v/w) at a flow rate of 1.0 mL/min. Column eluent was monitored at excitation and emission wavelengths of 290 and 430 nm, respectively. The assay was linear (range 1.0–500 ng/mL) and this could be extended to 25 µg/mL by 50‐fold dilution integrity. No endogenous peaks were detected in the elution of analytes in drug‐free blank human plasma from six individuals and no interference was observed with co‐medications tested. Intra‐ and inter‐batch reproducibility studies demonstrated accuracy and precision within the acceptance criteria of bioanalytical guidelines. Integral Kernel Operators on Fock Space – Generalizations and Applications to Quantum Dynamics

A general theory of operators on Boson Fock space is discussed in terms of the white noise distribution theory on Gaussian space (white noise calculus). An integral kernel operator is generalized from two aspects: (i) The use of an operator-valued distribution as an integral kernel leads us to the Fubini type theorem which allows an iterated integration in an integral kernel operator. As an application a white noise approach to quantum stochastic integrals is discussed and a quantum Hitsuda–Skorokhod integral is introduced. (ii) The use of pointwise derivatives of annihilation and creation operators assures the partial integration in an integral kernel operator. In particular, the particle flux density becomes a distribution with values in continuous operators on white noise functions and yields a representation of a Lie algebra of vector fields by means of such operators.     

Improving the rate of convergence of Newton methods on Banach spaces with a convergence structure and applications

In this note, we use inexact Newton-like methods to find solutions of nonlinear operator equations on Banach spaces with a convergence structure. Our technique involves the introduction of a generalized norm as an operator from a linear space into a partially ordered Banach space. In this way, the metric properties of the examined problem can be analyzed more precisely. Moreover, this approach allows us to derive from the same theorem, on the one hand, semilocal results of Kantorovich-type, and on the other hand, global results based on monotonicity considerations. By imposing very general Lipschitz-like conditions on the operators involved, on the one hand, we cover a wider range of problems, and on the other hand, by choosing our operators appropriately, we can find sharper error bounds on the distances involved than before. Furthermore, we show that special cases of our results reduce to the corresponding ones already in the literature. Finally, several examples are being provided where our results compare favorably with earlier ones.     

Algebra matrix and similarity classification of operators

In this paper, by the Gelfand representation theory and the Silov idempotents theorem, we first obtain a central decomposition theorem related to a unital semi-simple n-homogeneous Banach algebra, and then give a similarity classification of two strongly irreducible Cowen-Douglas operators using this theorem.

An efficient numerical scheme for precise time integration of a diffusion-dissolution/precipitation chemical system

A numerical scheme based on an operator splitting method and a dense output event location algorithm is proposed to integrate a diffusion-dissolution/precipitation chemical initial-boundary value problem with jumping nonlinearities. The numerical analysis of the scheme is carried out and it is proved to be of order 2 in time. This global order estimate is illustrated numerically on a test case.
