A high-performance liquid chromatographic (HPLC) method is described for determination of the ratio of beta-d-mannuronic acid to alpha-l-guluronic acid (M/G ratio) in dietary fibre of edible seaweeds. Total dietary fibre (TDF) content was determined gravimetrically. The TDF fraction was hydrolysed with 12 m and 1 m H(2)SO(4), then neutralized with AG 4 x 4 resin. The uronic acids were separated in a Tracer Extrasil SAX 5 micro m column (25 cm x 4 mm) at 35 degrees C, with 2 mm KH(2)PO(4) containing 5% methanol as mobile phase at a fl ow rate of 1.5 mL/min. The detection wavelength was UV 210 nm. The chromatographic identifications of beta-d-mannuronic acid and alpha-l-guluronic acid were confirmed by liquid chromatography-mass spectrometry (LC-MS). The method precision was 1.4% for beta-d-mannuronic acid and 3.5% for alpha-l-guluronic acid. The method was used to determine M/G ratio in canned seaweeds (Saccorhiza polyschides and Himanthalia elongata) and in dried seaweeds (H. elongata, Laminaria ochroleuca, Undaria pinnatifida, Palmaria sp. and Porphyra sp.). 相似文献

The complexation of 2,3-anthracenedicarboxylate (ADC) by- and-cyclodextrins in water containing an organic solvent has been studied by induced circular dichroism. It has been shown that an increase of organic solvent ratio causes the degradation of the 1:1 ADC:-CD complex and the liberation of one guest molecule from the 2:1 ADC:-CD complex in water. The Schiff base of all-trans-retinal was investigated in organic solution by 1H- and 13C-NMR. at high field. Complete assignment of the 1H-NMR. peaks of N-butyl-(all-trans-retinylidene)amine ( 2 ) and the N-butyl-(all-trans-retinylidene)-ammonium ion ( 3 ) was achieved by INDOR (internuclear double resonance). The vicinal proton coupling constants of the polyene chain show that the π-bond orders remain unchanged in N-butyl-(all-trans-retinylidene)amine relative to all-trans-retinal ( 1 ), but change towards larger π-delocalization in the N-butyl-(all-trans-retinylidene)ammonium ion. At ?-61° only one isomer of N-butyl-(all-trans-retinylidene)ammonium was observed. This was shown to be trans at the imine linkage and independent of the solvent. The trifluoroacetic acid counter-ion can approach the positive charge of the N-atom in the weakly polar solvent dichloromethane but not in the leveling solvent methanol. In dichloromethane the nature of the 1:1 complex is a H-bonded (O?…H-N+) ion-pair whose rate of breaking and forming is rapid at RT. Strong stabilization of the ion-pair resulted from homo-conjugation with a second molecule of trifluoroacetic acid. Excess of acid efficiently diminished the isomerization rate at the C,N-bond. 相似文献

The geometries of the 2-aminoethyl cation and the isomeric protonated aziridine have been optimized using ab initio molecular orbital calculations employing the split-valence shell 4-31G basis set. The protonated aziridine is computed to be the more stable ion by 46.5 kcal/mole (4-31G level) and 44.9 kcal/mole (double-zeta basis set).
