Voriconazole is used to prevent invasive pulmonary aspergillosis. However, little is known about the concentrations of voriconazole in human lung epithelial cells (A549), which is the target for preventing invasive pulmonary aspergillosis. The goal of this study was to develop a high-performance liquid chromatography–tandem mass spectrometry method to quantify voriconazole in A549 cells. A triple-quadrupole mass spectrometer in selected reaction monitoring mode was used with positive electrospray ionization. The total duration of each run was 5?min. The calibration curves fit a least squares model for the voriconazole concentration ranging from 0.625 to 160?ng/mL. Intraday and interday coefficients of variation were less than 10%. Recoveries at the concentrations of the quality control samples where greater than 85%, and the matrix effects showed that the ratios of the peak response exhibited a 15% suppression of the signal in the matrix compared to water. Voriconazole may penetrate A549 cells. However, the voriconazole uptake was slow in A549 cells, reaching a plateau at 2?h, where the dose-dependent intracellular voriconazole concentrations were 1.98?±?0.38, 4.43?±?0.54, and 8.14?±?0.52?ng/mg protein for extracellular voriconazole concentrations of 5, 10, and 20?µg/mL, respectively. The uptake of voriconazole by A549 cells was linear at extracellular concentrations from 0 to 20?µg/mL. This study established a rapid and sensitive method suitable for determining voriconazole in A549 cells and described the kinetic properties of the absorption of voriconazole by A549 cells. 相似文献

探讨老年急性心肌梗塞(AMI)患者QT离散度(QTd)、QTc离散度(QTcd)与泵衰竭的关系。方法61例老年AMI患者分为两组:泵衰竭组(Kilip分级泵功能≥Ⅲ级者,n=21)和对照组(Kilip分级泵功能Ⅰ级者,n=40)。分析患者入院时心电图的QTd和QTcd。结果:QTd、QTcd在泵衰竭组分别为(646±1601)ms和(7733±2060)ms,在对照组分别为(5275±1450)ms和(6098±1593)ms,二组间比较差异有显著性(P<005)。结论:QTd和QTcd增大与AMI患者的泵衰竭有关,QTd和QTcd可作为评估老年人AMI早期泵功能和预后的指标 相似文献

为了探讨乳苣水提取物对体外培养的肺癌细胞SPCA-1的影响,分别用质量浓度为0.5、1、1.25和1.5 g/L(mg/m L)的乳苣水提取物处理SPCA-1细胞,24 h后通过CCK-8法检测发现,与对照组(未处理)相比,处理组细胞增殖活力均显著下降(P0.001).流式细胞仪检测(AnnexinⅤ-FITC/PI染色)发现随着处理浓度的增加,处理组细胞凋亡率都显著提高(P0.05),且呈剂量依赖关系.为了探索乳苣是否对体内肿瘤有抑制作用,构建了SPCA-1细胞的裸鼠皮下瘤模型,通过观察喂食乳苣水提取物后皮下瘤的生长情况发现,与未喂食乳苣水提取物的对照鼠相比,喂食5周后的实验组的瘤体得到明显的抑制.实验结果表明乳苣水提取物能够抑制体外和体内生长的SPCA-1细胞,可以作为一种潜在的预防和治疗肺癌的药物做进一步研究. 相似文献

目的探讨血浆sST2联合NT-proBNP对急诊心衰患者生存状况的预测价值.方法选择急诊心衰患者130例,分为心功能Ⅱ级组、心功能Ⅲ~Ⅳ级组,对照组选择健康的体检者60例.对3组NT-proBNP及血浆sST2水平进行观察对比,对心功能不同分级患者NT-proBNP及血浆sST2水平进行比较,比较NT-proBNP,sST2,NTproBNP联合血浆sST2对心衰患者死亡状况的预测价值.结果随心功能分级递增,NT-proBNP水平逐渐升高,sST2曲线下面积(0.776)及NT-proBNP对死亡判定的ROC曲线与NT-proBNP曲线下面积(0.775)相比较没有明显差异,但二者联合曲线下面积为0.813,与NT-proBNP及sST2曲线下面积相比较差异显著.sST2联合NTproBNP进行心衰患者死亡状况预测时的敏感度为99.0%,特异度为93.2%.结论作为急诊心衰患者诊断、预后和风险评估的主要标志物,sST2联合NT-proBNP对患者生存状况进行评估可以很好地预测患者的生存状况,提高准确度和敏感度,值得在临床上推广应用. 相似文献

目的探讨二维斑点追踪成像(2D-STI)及实时三维超声心动图(RT-3DE)评价AMI患者左心室收缩功能和各节段运动的同步性.方法 50例急性心肌梗死患者为病例组,30例正常体检者为对照组,应用CMQ软件获取各节段径向圆周应变及应变率曲线,然后转变为RT-3DE模式,得到17节段时间-体积曲线(VTC),并获得收缩功能以及非同步收缩参数.结果 1)与对照组相比,心肌梗死组3个水平所有节段径向应变值和圆周应变值均减低;2)心梗组患者Trs-SD,Tcs-SD,Trsr-SD,Tcsr-SD明显高于对照组(P0.05);3)与单支冠状动脉病变心梗组比较,多支冠状动脉病变心梗组LVEDV,LVESV显著增加,LVEF显著减小;4)病例组患者左室Tmsv17-SD,Tmsv17-Dif,Tmsv17-SD%,Tmsv17-Dif%均较对照组增大(P0.05).结论 RT-3DE和2D-STI可以准确地量化左室收缩功能,评价AMI患者左室运动的同步性,左室收缩功能障碍也影响收缩同步性. 相似文献

Reliable methods for the determination of tryptophan and its metabolites are vital to the monitoring of biochemical states during the initiation and progression of cardiovascular disease. In the present study, a single‐run liquid chromatography–tandem mass spectrometry (LC‐MS/MS) method was developed for the simultaneous determination of tryptophan (Trp) and its metabolites, including kynurenine (Kyn), kynurenic acid (KA), xanthurenic acid (XA) and 5‐hydroxytryptamine (5‐HT), in human plasma. The plasma samples were prepared using a protein precipitation approach, and the chromatographic separation was performed by gradient elution on a C18 column within a total analysis time of 3.5 min. The calibration ranges were 40–20,000 ng/mL for Trp, 4–2000 ng/mL for Kyn, 0.2–100 ng/mL for KA, 0.4–200 ng/mL for XA and 1–500 ng/mL for 5‐HT, and the precision and accuracy were acceptable. The evaluation of recovery and internal standard‐normalized matrix effect proved that the sample preparation approach was effective and the matrix effect could be negligible. The newly developed method was successfully applied to the analysis of plasma samples from healthy individuals and myocardial infarction patients. The findings suggested that the plasma concentrations of Trp, Kyn, 5‐HT as well as the concentration ratios of Kyn/Trp and Trp/5‐HT might serve as biomarkers for the monitoring of acute myocardial infarction. 相似文献
