根据要因实验原则,选择影响因子较大的因素进行实验,初步研究了根霉Rhizopussp.TC1的产酶条件.经实验确定优化产酶条件为:稻草∶麸皮=1∶1;固液比=1∶3;接种量9 mL种子液/100 g湿基;温度28℃,在此条件下发酵96 h,FPA和CMC酶活分别达到17.8 IU/g、187.0 IU/g,较优化前的12.3 IU/g、131.6 IU/g分别提高了45%和42%. 相似文献

用硫酸铵沉淀,Q-Sepharose HP离子交换层析,Superdex 200凝胶过滤层析和Mono-Q离子交换层析方法,对从大连潮间带繁茂膜海绵中分离得到的1株假单胞菌DEH138A(Pseudomonas sp.DEH138A)中的脱卤酶进行分离纯化和酶学性质表征。结果显示,假单胞菌菌株DEH138A中含有一种能降解L-2-氯丙酸的L-2-卤代酸脱卤酶(L-DEX)。经纯化后,L-DEX亚基分子量为27.8kDa,全酶分子量为42.5kDa,推测其为一个二聚体蛋白。L-DEX最适pH值及最适反应温度分别为10.0和30℃,Km值为0.63mmol/L。L-DEX能够作用于多种2-卤代酸,而且对单溴乙酸的脱卤效果最好。DTT和EDTA对L-DEX无抑制作用,Cu2+和Co2+对L-DEX有明显的抑制作用。L-DEX具有立体选择性等独特的酶学特性,在环境修复、精细化工等领域具有潜在的应用价值。 相似文献

A bacterial strain, SWU-4, capable of using benzothiophene (BT) as a sole carbon and energy source was isolated from a petroleum-contaminated
site in Thailand and identified by 16S rRNA gene sequence analysis to be in the genus of Mycobacterium. The strain was Gram-positive, nonspore former, and grew at 50° C. Colonies of the strain on nutrient agar were rod-shaped,
smooth with a convex surface, slightly mucoid, and yellow pigmented. The thermophilic Mycobacterium sp. strain SWU-4 rapidly degraded 2% (w/v) BT at 50°C. Interestingly, this strain was able to degrade a wide variety of organosulfur
compounds including thiophene, bromo(α)thiophene, and 3-methylthiophene in liquid minimum medium at 50°C, which will be beneficial

Physicochemical and biologic analysis of dairy wastewater showed that the effluent had a high organic load (chemical oxygen
demand [COD]: 5095 mg/L), an acidic pH (6.4), and a high probability of coliforms (most probable number [MPN]>1100). The various

Streptomyces omiyaensis SSM 5670 was characterized by its ability to use compression moulded samples of poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) as its sole carbon source. Biodegradation of PHBV in liquid mineral salts medium was investigated using scanning electron microscopy, gravimetric measurements, capillary viscometry, tensile testing and wide angle X-ray spectroscopy. The biodegradation of PHBV proceeds via surface erosion mechanism, resulting in the formation of pits by microbial attack. PHBV specimens lost about 45% of their original weight after 45 days of exposure. During the degradation process the elastic modulus reduces less than 10%. The formation of pores and microcracks initiated at the degraded pits determines the reduction of the elongation and stress at break. However, the true stress at break is practically independent of the degradation time. No significant changes of PHBV molecular weight or crystallinity were observed during biodegradation. The polymer chain cleavage occurred only at the specimen surface and does not discriminate between crystalline and amorphous states. 相似文献

分离得到产抗菌聚氨基酸--ε-聚赖氨酸菌株淀粉酶产色链霉菌TUST2,从中纯化了ε-聚赖氨酸降解酶,并对其性质进行了研究.结果表明,该酶为膜结合蛋白.为提取该降解酶,先收集菌体细胞并用超声波破碎,细胞膜部分用1.0 moL/L NaSCN溶液溶解.将粗酶液进行Sephadex G100凝胶柱层析分离.用100mmol/L磷酸缓冲液洗脱,收集活性部分.纯化后的样品用SDS-PAGE检测,酶亚基分子量约为54700.酶活力在pH=6.0~9.0间稳定,最适宜pH=7.0.酶的最适温度为30℃,在10~50℃水浴30 min酶活力未见明显下降.研究了不同金属离子对酶活力的影响,结果表明,Zn~(2+),Cu~(2+)和Fe_(3+)可分别提高酶活力29.72%,15.85%和15.08%;但Ag~(+),Hg~(2+),Co~(2+)和Mn~(2+)对酶活力有强烈的抑制作用.Ca2~(2+),K~+和Ba~(2+)对酶活力没有影响.添加4%Tween-80能提高酶活力10%,但EDTA能强烈抑制酶活力.研究结果表明,此降解酶的性质与白色链霉菌产生的ε-聚赖氨酸降解酶的性质相似. 相似文献
