采用分子动力学模拟和拉伸分子动力学模拟方法, 结合分子力学-广义玻恩表面积(MM-GB/SA)方法进行自由能计算和结构交互指纹分析, 研究了模拟过程中非特异性底物(对氧磷/内酯)分别与嗜热磷酸三酯酶样内酯酶(SsoPox)野生型和突变体(W263F/W263T)结合的构象变化, 分析了Loop8中重要残基Trp263的突变提高SsoPox非特异性底物活力的原因, 发现其能够影响门控残基Phe229的构象变化, 导致活性口袋入口变宽(Phe229与Tyr99之间的距离变大), 使对氧磷和内酯更容易结合到蛋白质的活性位点上; Asp256和Arg223形成盐桥的几率高于野生型(WT)SsoPox, 在Arg223(位于Loop7)的协助下质子更加高效地从活性中心的Asp256(位于Loop8)传递到溶剂中去, 因而能够提高SsoPox水解底物的效率. 通过比较2个野生型复合物的结构稳定性和结合自由能差异, 发现在模拟过程中SsoPox与内酯的复合物体系更加稳定并且具有更低的结合自由能, 有利于SsoPox识别底物并使其埋在活性部位的疏水环境中, 促进氢氧化物亲核进攻底物的亲电中心. 因此, 底物与酶稳定的相互作用可能是SsoPox具有天然内酯酶活性的原因之一. 相似文献

<正>In this study,advanced oxidation processes(AOPs) such as anodic oxidation(AO),UV/H_2O_2 and Fenton processes(FP) were investigated for the degradation of salicylic acid(SA) in lab-scale experiments.Boron-doped diamond(BDD) film electrodes using Ta as substrates were employed for AO of SA.In the case of FP and UV/H_2O_2,most favorable experimental conditions were determined for each process and these were used for comparing with AO process.The study showed that the FP was the most effective process under acidic conditions,leading to the highest rate of SA degradation in a very short time interval.However,the results showed that Ta/BDD films had high electrocatalytic activity for complete degradation of SA;even if it employs more time for complete elimination of the SA respect to FP.Additionally,AO led to a sixfold acceleration of the oxidation rate compared with the UV/H_2O_2 process.Finally a rough comparison of the specific energy consumption shows that AO process reduced the energy consumption by at least 90%compared with the UV/H_2O_2 process. 相似文献

Chito-oligosaccharides (COSs) were encapsulated by the film-ultrasonic method into three nano-liposomes, which were uncoated liposomes (COSs-Lip), chitosan-coated liposomes (CH-COSs-Lip), and sodium alginate (SA)/chitosan (CH)-coated liposomes (SA/CH-COSs-Lip). The physicochemical and structural properties, as well as the stability and digestive characteristics, of all three nano-liposomes were assessed in the current study. Thereafter, the characteristics of intestinal absorption and transport of nano-liposomes were investigated by the Caco-2 cell monolayer. All nano-liposomes showed a smaller-sized distribution with a higher encapsulation efficiency. The ζ-potential, Z-average diameter (Dz), and polydispersity index (PDI) demonstrated that the stability of the SA/CH-COSs-Lip had much better stability than COSs-Lip and CH-COSs-Lip. In addition, the transport of the nano-liposomes via the Caco-2 cell monolayer indicated a higher transmembrane transport capacity. In summary, the chitosan and sodium alginate could serve as potential delivery systems for COSs to fortify functional foods and medicines. 相似文献

Nowadays, time-resolved spectroscopy data can be routinely and accurately collected in UV-vis femtosecond transient absorption spectroscopy. However, the data analysis strategy and the postulation of a physically valid model for this kind of measurements may be tackled with many different approaches ranging from pure soft-modeling (model-free) to hard-modeling, where the elaboration of a parametric spectro-temporal model may be required. This paper reviews methods that are used in practice for the analysis of femtosecond transient absorption spectroscopy data. Model-based methods, common in photochemistry, are revisited, and soft-modeling methods, which originate from the chemometrics field and that recently disseminated in the photo(bio)chemistry literature, are presented. These soft-modeling methods are designed to suit the intrinsic nature of the multivariate (or multi-way) measurement. Soft-modeling tools do not require a priori physical or mechanistic models to provide a decomposition of the data on the time and wavelength dimensions, the only requirement being that these two (or more) dimensions are separable. Additionally, Bayesian data analysis, which provides a probabilistic framework for data analysis, is considered in detail, since it allows uncertainty quantification and validation of the model selection step. 相似文献

It is known that polymethine dyes (PD) can form complexes with biomacromolecules, in which, as a rule, fluorescence buildup is observed. In addition, PD possess a unique property to form ordered aggregates of different types (dimers, H- and J-aggregates) both in the free state and on biomacromolecules as templates. This creates a basis for application of PD as spectral-fluorescent probes for biomacromolecules, which is a matter of this review. The nonlinear absorption properties of organic materials in nanosecond (ns), picoseond (ps) and femtosecond (fs) regime were theoretically investigated by rate equations based on multilevel organic molecular system, respectively. The simulation results show that the RSA is mainly due to the absorption contribution of the triplet first excited state in the ns regime and that of the singlet first excited state in the ps regime, both of which are a third-order nonlinear phenomena. 本研究分析MRI测量胫骨结节-股骨滑车凹间距(tibial tuberosity-trocholear distance,TT-TG)、髌骨高度(patellar height,PH)、股骨滑车沟角(sulcus angle,SA)、滑车沟深度(sulcus depth,SD),与一次性髌骨外侧脱位(transient lateral patellar dislocation,TLPD)患者诊断及分型的关系。将2018年1月~2019年2月我院收治的50例TLPD患者作为观察组,50例膝关节正常者作为对照组。采用1.5 T MRI测量受试者TT-TG、PH、SA、SD水平,分析各指标单独应用及联合应用的诊断价值,并探讨其与临床分型的关系。结果显示,观察组TT-TG、PH、SA水平均明显高于对照组(P<0.05),观察组SD水平明显低于对照组(P<0.05);TT-TG、PH、SA及SD指标的联合应用可明显提高TLPD诊断的敏感度、特异度及准确度(P<0.05),且联合应用时AUC面积明显大于各指标单独应用,差异存在统计学意义(P<0.05);随着患者Dejour分型恶化,患者TT-TG、PH及SA水平均明显升高,SD水平随之明显降低(P<0.05);TT-TG、PH、SA及SD是影响TLPD患者临床分型的独立性影响因素(P<0.05)。研究结果表明,采用1.5 T MRI对TLPD患者的TT-TG、PH、SA、SD进行测量可有效提高临床诊断价值,且TT-TG、PH、SA、SD是影响患者临床分型的独立性影响因素。 相似文献
