The behavior of (ferrocene)amylose (FA), in the presence of amylolytic depolymerases (α-amylase from Aspergillus oryzae and human saliva), has been investigated by cyclic voltammetry at a rotating disk electrode (CVA/RDE). Growth of the limiting current with time in the presence of the enzymes is proportional to the amount of enzyme introduced. The quantitative data treatment to assay the endoamylolytic activity of enzymes at CVA/RDE involves plotting (idt/id0)4.5 against time; the slope of the linear plot being equal to (rate) Mn0C−1, where idt and id0 are the limiting currents at time t and 0, respectively, (rate) is the enzymatic activity, Mn0 is the number averaged molecular weight of FA at t = 0, and c is its concentration. The comparison of CVA/RDE with the 3,5-dinitrosalicylic acid and the Somogyi–Nelson reducing saccharides procedures shows advantages of the former, especially in assaying small quantities of enzymes. Also the CVA/RDE approach is simpler and takes place under much milder conditions. The main disadvantage of CVA/RDE is the inhibiting effect of Triton X-100 in the reaction between FA and the amylases which is not observed in the case of native, ferrocene-free amylose. In general, CVA/RDE appears to be an attractive analytical method for monitoring diverse enzymatic depolymerization reactions.  相似文献

A semi-analytical solution is obtained for a rotating stress-free edge disk of constant thickness and density. In the plastic range, the Hill’s quadratic orthotropic yield criterion is adopted. In the elastic range, the Hooke’s law holds with thermal effects included. The analysis of singularities performed may be used for correct implementation of numerical codes and preliminary engineering design.  相似文献

Non-linear internal damping in rotating cylindrical shafts leads to isotropic non-linear circulatory and dissipative force fields. Orbital instability in such class of systems arises when the regenerative work due to circulatory forces exceeds the dissipative work done over an orbit. In this paper, stability condition for rotor shafts with polynomial type non-linear internal damping is derived and also it is shown that distortion of the critical orbit, which results in larger change in orbital path as compared to the change in enclosed area, may lead to stable orbit at normally unstable operating conditions. This principle is applied to stabilize a simple rotor with discontinuous stiffness characteristics, which come into effect on exceeding some threshold displacement of the rotor itself.  相似文献

The axisymmetric spreading of a thin liquid drop under the influence of gravity and rotation is investigated. The effects of the Coriolis force and surface tension are ignored. The Lie group method is used to analyse the non-linear diffusion-convection equation modelling the spreading of the liquid drop under gravity and rotation. A stationary group invariant solution is obtained. The case when rotation is small is considered next. A straightforward perturbation approach is used to determine the effects of the small rotation on the solution given for spreading under gravity only. Over a short period of time no real difference is observed between the approximate solution and the solution for spreading under gravity only. After a long period of time, the approximate solution tends toward a dewetting solution. We find that the approximate solution is valid only in the interval t∈[0,t∗), where t∗ is the time when dewetting takes place. An approximation to t∗ is obtained.  相似文献

The heat transfer from a rotating cylinder in an air-cross flow was investigated by purely optical measuring techniques. Flow velocities were measured by a two-dimensional LDV both in the vicinity of the cylinder and in the boundary layer. A new optical device based on light-deflection in a temperature field was developed to examine local temperature gradients in the boundary layer of the rotating cylinder. Finally, a Michelson-interferometer was installed to produce real-time pictures of isothermal lines around the heated cylinder. The impact of rotation on flow patterns, boundary layer behaviour and heat transfer could be clearly identified. It appears that the velocity-ratio acts like an independent parameter, in that flow patterns correspond to this dimensionless number. Furthermore, it seems that rotation dominates over cross flow, both fluid-dynamically and thermally above = 2.This work was carried out at the University of the Federal Armed Forces in Munich/Germany.  相似文献

Numerical calculations have been carried out for flow in a truncated cone generated by rotation of one endwall. For both convergent (radius increasing with approach to the rotating endwall) and divergent geometries, vortex breakdown is suppressed beyond a certain angle of inclination of the sidewall. At the same time Moffat eddies of increasing strength and extent appear in the corner between the sidewall and the non-rotating endwall. For the divergent geometry, a zone of recirculation appears on the sidewall and eventually merges with the Moffat eddies. The flow phenomena identified from streamline patterns are consistent with the calculated variation of pressure around the periphery of the computational domain.  相似文献

This study investigates the rotating magnetohydrodynamic (MHD) flow of a third-grade fluid in a porous space. Modified Darcy's law has been utilized for the flow modeling. The Hall effects are taken into consideration. The basic equations governing the flow are reduced to a highly nonlinear ordinary differential equation. This equation has been solved analytically by employing the homotopy analysis method (HAM). The effects of the various interesting parameters on the velocity distribution have been discussed.  相似文献

Analytical solutions for the stress distribution in rotating parabolic solid disks are obtained. The analysis is based on Tresca's yield criterion, its associated flow rule and linear strain hardening. It is shown that, the deformation behavior of the convex parabolic disk is similar to that of the uniform thickness disk, but in the case of concave parabolic solid disk, it is different. In the latter, the plastic core consists of three different plastic regions with different mathematical forms of the yield criteria. Accordingly, three different stages of elastic–plastic deformation occur. All these stages of elastic–plastic deformation are studied in detail. It is also shown mathematically that in the limiting case the parabolic disk solution reduces to the solution of rotating uniform thickness solid disk.  相似文献

This paper presents accurate elastic solutions for the rotating variable-thickness and/or uniform-thickness orthotropic circular cylinders. The present circular cylinder may contain a uniform-thickness solid core of rigid or homogeneously isotropic material. Different cases of rotating cylinders of various cores are investigated. These cylinders include completely isotropic solid cylinder, uniform-thickness orthotropic cylinder containing an isotropic core, variable-thickness orthotropic cylinder containing an isotropic core, uniform-thickness orthotropic cylinder containing a rigid core, and variable-thickness orthotropic cylinder containing a rigid core. For all cases studied, exact elastic solutions are obtained and numerical results are presented. The results include the radial, hoop, and axial stresses and radial displacement of the five cylinder configurations. The distributions of displacement and stresses through the radial direction of the rotating cylinder are obtained and comparisons between different cases are made at the same angular velocity.  相似文献

This paper deals with the non-linear viscoelastodynamics of three-dimensional rotating structure undergoing finite displacement. In addition, the non-linear dynamics is studied with respect to geometrical and mechanical perturbations. On part of the boundary of the structure, a rigid body displacement field is applied which moves the structure in a rotation motion. A time-dependent Dirichlet condition is applied to another part of the boundary. For instance, this corresponds to the cycle step of a helicopter rotor blade. A surface force field is applied to the third part of the boundary and depends on the time history of the structural displacement field. For example, this might corresponds to general unsteady aerodynamics forces applied to the structure. The objective of this paper is to model the non-linear dynamic behavior of such a rotating viscoelastic structure undergoing finite displacements, and to allow small geometrical and mechanical (mass, constitutive equations) perturbations analysis to be performed. The model is constructed by the introduction of a reference configuration which is deduced from the non-linear steady boundary value problem. A constitutive equation deduced from the Coleman and Noll theory concerning the viscoelasticity in finite displacement is used. Thereafter, the weak formulation of the boundary value problem is constructed and discretized using the finite element method. In order to simplify the mathematical study of the equations, multilinear forms are introduced in the algebraic calculation and their mathematical properties are presented.  相似文献
