Antigen retrieval on formaldehyde-fixed paraffin sections: its potential drawbacks and optimization for double immunostaining

It is well known that the major artifact induced by formaldehyde fixation is the masking of tissue antigens due to cross-linking of protein amino acid residues. Recently many antigen retrieval techniques have been devised to unmask the hidden antigen epitopes and recover immunoreactivity. In this study, some practical problems of two common unmasking techniques, i.e. heat-induced epitope retrieval and enzyme digestion have been reviewed in immunostaining of proliferating cell nuclear antigen (PCNA) on formaldehyde-fixed paraffin-embedded sections. As the heating conditions became more severe, false-positive staining and/or nonspecific background staining occurred. Based on the principle of protein inactivation/denaturation and the possible mechanisms of antigen retrieval, it has been suggested that the antigen retrieval itself can also denature proteins in tissues, just as many other protein inactivation processes. Thus, the total magnitude of protein conformational change caused by the overall unmasking procedure is in practice crucial. To prove this hypothesis and to overcome such undesirable drawbacks after antigen retrieval, a new combination technique of a mild heating condition (microwaved at 80°C for 15–20 min) and pepsin digestion was devised. This technique led to a strong specific immunoreactivity of PCNA, without any undesirable false positive or background staining. The procedure was also adapted for double immunostaining of PCNA together with -actin, bromodeoxyuridine, keratin, type IV collagen and vimentin.     

CD40L对B淋巴细胞表面分子及分泌IL-12的影响

在体外使用CD40L作用于B淋巴细胞使之活化,通过测定B淋巴细胞表面分子及分泌细胞因子的变化,探讨B淋巴细胞作为抗原递呈细胞在抗肿瘤免疫中的优势,为B淋巴细胞特异性活化细胞毒T淋巴细胞(cytotoxic T lymphocyte,CTL)进而发挥其肿瘤细胞杀伤效应奠定基础.取7名健康成人外周静脉血,经淋巴细胞分离液分离人外周血单个核细胞(PBMC),分为对照组和实验组,实验组培养液中加入加入CD40L和IL-4,共培养21 d.观察B淋巴细胞生长状况,并在第7 d、第14 d和第21 d,用流式细胞仪(FCAS)测定其表面分子CD80、CD86和CD19,在第21 d,使用酶联免疫吸附法(ELISA)测定培养液中的IL-12水平.结果显示,在CD40L的作用下,B淋巴细胞呈克隆样增殖,并高表达表面分子CD80/CD86和CD19,和对照组相比,B淋巴细胞分泌IL-12水平升高(P＜0.05).上述实验结果说明通过CD40L的刺激,B淋巴细胞得到了活化,提示通过该途径活化的B淋巴细胞具备了作为抗原递呈细胞而发挥其抗肿瘤免疫治疗的能力.     

禽流感病毒H5亚型特异性抗原捕捉ELISA检测方法的研究

 在获得禽流感病毒多克隆抗体及H5亚型特异性单克隆抗体的基础上,研究建立H5亚型特异性抗原捕捉ELISA检测方法,用于检测H5亚型禽流感病毒.优化反应条件,确定包被抗体、检测抗体及酶结合物的最佳工作浓度,进行敏感性、特异性、重复性及稳定性分析,并与RT-PCR方法比较.同时使用该方法对野外样品进行检测.结果表明:该方法敏感、特异,具有良好的重复性和稳定性,可用于检测临床样品、鸡胚培养物及细胞培养物中的H5亚型禽流感病毒.     

Vanishing structure set of 3-manifolds

In this short note we update a result proved in Roushon (2007) [17]. This will complete our program of Roushon (2000) [13] showing that the structure set vanishes for compact aspherical 3-manifolds.

Congruence of Siegel modular forms and special values of their standard zeta functions

In this paper, we consider the relationship between the congruence of cuspidal Hecke eigenforms with respect to Sp _n (Z) and the special values of their standard zeta functions. In particular, we propose a conjecture concerning the congruence between Saito-Kurokawa lifts and non-Saito-Kurokawa lifts, and prove it under certain condition. Delta shock wave formation in the case of triangular hyperbolic system of conservation laws

We describe δ-shock wave generation from continuous initial data in the case of triangular conservation law system arising from “generalized pressureless gas dynamics model.” We use smooth approximations in the weak sense that are more general than small viscosity approximations.     

Eigenfunctions of transfer operators and cohomology

The eigenfunctions with eigenvalues 1 or −1 of the transfer operator of Mayer are in bijective correspondence with the eigenfunctions with eigenvalue 1 of a transfer operator connected to the nearest integer continued fraction algorithm. This is shown by relating these eigenspaces of these operators to cohomology groups for the modular group with coefficients in certain principal series representations.     

The blow-up curve of solutions of mixed problems for semilinear wave equations with exponential nonlinearities in one space dimension, I

In this paper, we consider mixed problems with a spacelike boundary derivative condition for semilinear wave equations with exponential nonlinearities in a quarter plane. Results similar to those obtained earlier by Caffarelli-Friedman for Cauchy problems and power nonlinearities are proved in the present situation, namely we show that solutions either are global or blow up on a spacelike curve. Weaker results are also obtained if the boundary vector field is tangent to the characteristic which leaves the domain in the future. A Multidimensional Generalization of Shukla's 8ψ8 Summation

   Abstract. We give an r -dimensional generalization of H. S. Shukla's very-well-poised ₈ ψ ₈ summation formula. We work in the setting of multiple basic hypergeometric series very-well-poised over the root system A _r-1 or, equivalently, the unitary group U(r) . Our proof, which is already new in the one-dimensional case, utilizes an A _r-1 nonterminating very-well-poised ₆ φ ₅ summation by S. C. Milne, a partial fraction decomposition, and analytic continuation.
