从人胎脑cDNA文库中克隆到一条长3479bp的cDNA,它含一个长903bp的开放阅读框架,拟编码一个300个氨基酸的蛋白质,其分子质量为35397u,等电点为5.35,与酵母MAK16蛋白的同源性为41%,该编码蛋白有一个双侧核定位信号motif和一个RNA结合motif,将这一新cDNA序列推导的蛋白命名为RNA结合基序蛋白13(RBM13),基因定名为RBM13,用RBM13基因的cDNA探针进行Northern杂交,检测到3.6kb和2.2kb二种长度的转录本。在心脏,骨骼肌,肾脏和肝脏中有较高表达。  相似文献

A cyclic bimetallic metal-organic complex [Zn(C)(H-fmpdc)(H2O)]2·2H2O (fmpdc = 4-(furan-2-yl)-2,6-dimethylpyridine-3,5-dicarboxylate) was synthesized and characterized by single-crystal X-ray analysis. The compound crystallizes in orthorhombic, space group Pbca with a = 12.905(2), b = 14.774(3), c = 16.833(3)A, V= 3029.4(10)A^3 Z = 4, Dc = 1.644 g/cm^3, F(000) = 1616, R = 0.0347 and wR = 0.0956 (I 〉 2σ(I)). There exist diverse supramolecular motifs (1-D, 2-D and 3-D) from distinct hydrogen bonds in the crystal structure of the title compound. The furanyl group has obvious contribution to the red-shift in the photoluminescent spectrum of the H2fmpdc ligand. The title compound 1 shows strong photoluminescence with emission maximum at 2 = 402 nm (λex.max = 367 nm).  相似文献

为了解学龄前儿童智商与血中微量元素含量之间的关系本文测定了佛山市幼儿园576名4-6岁儿童的IQ值及其血中六种元素(Zn、Fe、Cu、Ca、Mn、Pb)的含量,并对幼儿血中微量元素含量与IQ值的关系进行了探讨。结果表明,低智商儿童血中锌含量显著低于高智商儿童血中锌含量,而血铅则显著高于高智商儿童的血铅。说明学龄前儿童智商高低与机体锌及铅的含量有密切关系。  相似文献

Short guanine(G)‐repeat and cytosine(C)‐repeat DNA strands can self‐assemble to form four‐stranded G‐quadruplexes and i‐motifs, respectively. Herein, G‐rich and C‐rich strands with non‐G or non‐C terminal bases and different lengths of G‐ or C‐repeats are mixed selectively in pH 4.5 and 6.7 ammonium acetate buffer solutions and studied by electrospray ionization mass spectrometry (ESI‐MS). Various strand associations corresponding to bi‐, tri‐ and tetramolecular ions are observed in mass spectra, indicating that the formation of quadruplex structures is a random strand by strand association process. However, with increasing incubation time for the mixtures, initially associated hybrid tetramers will transform into self‐assembled conformations, which is mainly driven by the structural stability. The melting temperature values of self‐assembled quadruplexes suggest that the length of G‐repeats or C‐repeats shows more significant effect on the stability of quadruplex structures than that of terminal residues. Accordingly, we can obtain the self‐associated tetrameric species generated from the mixtures of various homologous G‐ or C‐strands efficiently by altering the length of G‐ or C‐repeats. Our studies demonstrate that ESI‐MS is a very direct, fast and sensitive tool to provide significant information on DNA strand associations and stoichiometric transitions, particularly for complex mixtures. Due to its unique role as a hydrogen‐bond donor and its positive charge, the guanidine group is an important pharmacophoric group and often used in synthetic ligands. The chemical modification of the guanidine group is often considered to destroy its function. Herein, we show that the N‐methylation, N‐alkylation, or N‐acylation of the guanidine group can be used to modify the receptor subtype specificity of the integrin ligand cilengitide. Using the αvβ6/α5β1‐biselective ligand c(isoDGRkphg) and the αvβ6‐specific ligand c(FRGDLAFp(NMe)K(Ac) as examples, we show that the binding affinities of the ligands can be fine‐tuned by this method to enhance the selectivity for αvβ6. Furthermore, we describe a new strategy for the functionalization of integrin ligands. By introducing longer N‐alkylguanidine and N‐acylguanidine groups, we are able to simultaneously identify a hitherto unknown anchoring point and enhance the subtype selectivity of the ligand.  相似文献

本研究从鸡cDNA文库中,通过菌落PCR筛选获得单核细胞趋化激活因子(MCP-1)基因的cDNA,将该cDNA中的ORF插入到pET-28a( )质粒(携带T7/lac启动子序列和His-Tag标签序列)中,得到pET-mcp-1质粒;重组质粒转化E.coliBL21(DE3)后,经IPTG诱导表达得到MCP-1和His-Tag的融合蛋白(HisTag-MCP-1)。该基因不仅具有CC趋化因子家族Cys-Cys氨基酸模序,并且与结构和功能相关的氨基酸高度保守。  相似文献

This paper attempts to reconstruct the phylogeny of azoreductase enzyme from different organisms and compare it with the small subunit rRNA-based phylogeny of the organisms. The two phylogenies were found to be incongruent, indicating several events of lateral transfer of azoreductase gene between phylogenetically diverse organisms. However, the phylogenetic analysis methods have several limitations and a single method may not give the true pattern. Hence, it is necessary to corroborate the results with other complementary analysis tools. We used several tools to test our hypothesis of lateral transfer and found that it was supported not only by the analysis of the whole sequences, but also by the conserved motifs detected in these sequences. There were ample evidences for lateral transfer of azoreductase gene among enteric bacteria. There were also indications that azoreductase probably evolved in prokaryotes and then it was laterally transferred to eukaryotes in multiple events, resulting in some sequence variation among eukaryotic azoreductases. Finally, profile HMMs and conserved motifs extracted from these azoreductase sequences were found to provide sensitive tools for identifying potential azoreductases from the database. The analysis techniques used in this study can be extended to other gene trees to verify their evolutionary histories.  相似文献

The natural occurrence of the human telomeric G-quadruplex or i-motif in vivo has not been demonstrated and the biological effects of the induction of these structures need to be clarified. Intracellular environments are highly crowded with various biomolecules and in vitro studies under molecular-crowding conditions will provide important information on how biomolecules behave in cells. Here we report that cell-mimic crowding can increase i-motif stability at acid pH and cause dehydration. However, crowding can not induce i-motif formation at physiological pH. Intriguingly, single-walled carbon nanotubes (SWNTs) can drive i-motif formation under cell-mimic crowding conditions and cause more water to be released. To our knowledge, there is no report to show how SWNTs can influence DNA under cell-mimic crowding conditions. Our results indicate that SWNTs may have the potential to modulate the structure of human telomeric DNA in vivo, like DNA B-A transitions and B-Z changes on SWNTs in live cells, which demonstrates potential for drug design and cancer therapy.  相似文献
