Upon the study of small-molecules binding to proteins, the traditional methods for calculating dissociation constants (Kd and Ki) have shortcomings in dealing with the single binding site models. In this paper, two equations have been derived to solve this problem. These two equations are independent of the total concentration or initial degree of saturation of receptor and the activity of the competitive molecule. Through nonlinear fitting against these two equations, Kd value of a probe can be obtained by binding assay, and Ki value of a ligand can be obtained by competitive assay. Moreover, only the total concentrations of receptor([R]t), ligand([L]t) and probe([P]t) are required for the data fitting. In this work, Ki values of some typical ligands of PPARγ were successfully determined by use of our equations, among which the Ki value of PPARγ-LY171883 was reported for the first time. 相似文献

Apparent equilibrium constants and calorimetric enthalpies of reaction have been measured for the reaction L-tryptophan(aq) + H2O(l) = indole(aq) + pyruvate(aq) + ammonia(aq) which is catalyzed by L-tryptophanase. High-pressure liquid-chromatography and microcalorimetery were used to perform these measurements. The equilibrium measurements were performed as a function of pH, temperature, and ionic strength. Preparation, 19F NMR Spectroscopic Evidence and Study of the Formation of Metal-Mixed Cluster Anions [(Mo6?nWnCl )F ]2?, n = 0?6 The complete system of metal-mixed octahedral cluster ions [(Mo6?nWnCl)F]2?, n = 0?6, is prepared by tempering Mo powder with WCl6 at 600°C. A mixture containing inclusively the geometric isomers (n = 2, 3, 4) all ten possible species is transferred into the tetra-n-butylammonium salts (TBA)2[(Mo6?nWnCl)F]. In the 19F nmr spectrum the 24 expected signals are observed, assigned on the basis of their chemical shifts, multiplicities and intensities, and confirmed by a 2D-19F-19F COSY spectrum. From the integrated intensities the distribution of the different components is derived revealing a non-statistical formation, in that isomers with Mo…?Mo or W…?W atoms in trans-positions in comparision to those with mixed Mo…?W axes are favoured, and that especially the homoleptic compounds Mo6 and W6 are present to an over-average extent. Evaluation of 19F chemical shifts reveals that F bound to W which is in antipodal position to Mo resonates at higher field compared to F bound to W in a W…?W arrangement, caused by an increased shielding, which is synonymous to a positive antipodal-effect by Mo. Vice versa F bound to Mo with an antipodal W resonates at lower field compared with F bound to Mo in an Mo…?Mo arrangement caused by an increased deshielding and synonymous a negative antipodal-effect by W. The chemical shifts, resulting from antipodal-effects, are different for the compounds within the [(Mo6?nWnCl)F]2? - system. The difference of the antipodal effect of successive substitution products results in characteristic values designated as antipodal shift constants, depending on the kind of substituents, which is valid for other cluster systems, too. 相似文献

Hydrogen abstraction reaction, H C2H4 --H2 C2H2 was studied by using A initio SCF method. Ge-ometries were fully optimized at SCF level and energies were computed at STO-3G basis set for reactants and transition state. Vibrational analysis was performed thereupon. Finally, the rate constant calculations were carried out at different temperatures for all range of reaction temperature according to Eyring's sbwlute reaction rate theory. The calculated activation energy is 12. 68 kcal/mol, lower than observed value (H. S kcal/mol) by 1. 82 kcal/mol only. In condensed matter, optical properties can be described by a dielectric function (DF), and the structures observed in spectra are then related to the poles and zeros of the DF. As an example, model functions are calculated by a fit to measured spectroscopic data for polystyrene and silica. The first material shows weak, narrow bands and the latter strong, broad bands and a negative real part of the DF.Based on these model DFs, spectra are simulated which are expected to be obtained by “conventional” methods such as transmittance or reflectance measurements, or by “unconventional” methods such as reflectance at oblique incidence, diffuse reflectance, photoacoustic spectroscopy and attenuated total reflectance. A variety of simulated, typical spectra are plotted as a small “atlas”. Conditions are discussed that allow a straightforward procedure for interpreting the spectra quantitatively, i.e., the evaluation of the resonance frequency and the concentration of the oscillators under consideration.It is shown that for systems characterized by weak, narrow oscillator lines, mostly an intuitive interpretation is possible, looking only at the position and strength of “lines” in the spectra. Materials showing strong polar vibrations, however, require more sophisticated procedures for interpreting the spectra. 相似文献

The cloud point extraction behaviors of lanthanoids(III) (Ln(III) = La(III), Eu(III) and Lu(III)) with and without di(2-ethylhexyl)phosphoric acid (HDEHP) using Triton X-100 were investigated. It was suggested that the extraction of Ln(III) into the surfactant-rich phase without added chelating agent was caused by the impurities contained in Triton X-100. The extraction percentage more than 91% for all Ln(III) metals was obtained using 3.0 × 10−5 mol dm−3 HDEHP and 2.0% (v/v) Triton X-100. From the equilibrium analysis, it was clarified that Ln(III) was extracted as Ln(DEHP)3 into the surfactant-rich phase. The extraction constant of Ln(III) with HDEHP and 2.0% (v/v) Triton X-100 were also obtained. 相似文献
