Apparatus integrating a Fourier transform-infrared (FT-IR) spectrometer and a mid-infrared difference frequency generation (DFG) laser spectrometer was built for the study of the matrix isolation spectrum in solid molecular hydrogen. A 3-cm-long molecular hydrogen crystal was grown in a liquid-helium Dewar, and its infrared absorption spectrum in the 1-5 μm region was recorded to test the system. The W0(0) (ν=0←0, J=6←0) line around 2410 cm−1 of solid hydrogen was investigated with the DFG laser spectroscopy. High-resolution matrix isolation spectrum of CO2 co-deposited with hydrogen on a BaF2 cold plate at liquid-helium temperature was studied.  相似文献

The availability of sufficient quantities of DNA of adequate quality is crucial in polymerase chain reaction (PCR)-based methods for genetically modified food detection. In this work, the suitability of anion-exchange CIM (Convective Interaction Media; BIA Separations, Ljubljana, Slovenia) monolithic columns for isolation of DNA from food was studied. Maize and its derivates corn meal and thermally pretreated corn meal were chosen as model food. Two commercially available CIM disk columns were tested: DEAE (diethylaminoethyl) and QA (quaternary amine). Preliminary separations were performed with standard solution of salmon DNA at different pH values and different NaCl concentrations in mobile phase. DEAE groups and pH 8 were chosen for further isolations of DNA from a complex matrix-food extract. The quality and quantity of isolated DNA were tested on agarose gel electrophoresis, with UV-scanning spectrophotometry, and by amplification with real-time PCR. DNA isolated in this way was of suitable quality for further PCR analyses. The described method is also applicable for DNA isolation from processed foods with decreased DNA content. Furthermore, it is more effective and less time-consuming in comparison with the existing proposed methods for isolation of DNA from plant-derived foods.  相似文献

The products of high-vacuum pyrolysis of 1,1,1-trifluoro-2-bromo-2-chloroethane were studied by matrix IR spectroscopy. The decomposition of 1,1,1-trifluoro-2-bromo-2-chloroethane was shown to occur predominantlyvia two directions: to form the 1,1,1-trifluoro-2-chloroethyl radical and trifluoromethylcarbene isomerizing to trifluoroethylene. The CF3CHCl radical has been detected in the matrix for the first time. The bands observed in the IR spectrum were calculated by the quantum-chemical B3LYP/6-311 G(d,p) method and assigned to normal vibrations of the radical. Translated fromIzvestiya Akademii Nauk. Extracts of Aconitum septentrionale Koelle roots obtained using chloroform, isopropanol, and ethanol were purified using chloroform and basic γ-Al2O3. Ballast materials were selectively adsorbed by γ-Al2O3, increasing the mass fraction of lappaconitine in the extract. The ethanol extract was purified most. The degree of lappaconitine extraction by chloroform was unaffected by the presence of γ-Al2O3. Bromine Superoxide: Generation and Photoisomerization into Bromine Dioxide Flash pyrolysis of a gas mixture containing bromine, oxygen and argon yields bromine superoxide, which can be identified IR- and UV-spectroscopically after trapping the pyrolysate at 12 K. Matrix irradiation transfers bromine superoxide into bromine dioxide. The backreaction can be induced by changing the wavelength. Even at room temperature a detectable amount of bromine superoxide is formed upon preparing a mixture of gaseous bromine, oxygen and argon.  相似文献
