Ab initio calculations ofp-dichlorobenzene molecule were carried out using the Hartree-Fock method in the 6–31 G* valence-split basis set. The molecule was also calculated by the MNDO method in the valence sp-basis set for comparison. The populations of the valent p-orbitals of the C and CI atoms were analyzed. The optimized geometry of the molecule as well as its35Cl NQR frequency and the asymmetry parameter of the electric field gradient at the35CI nuclei calculated using the populations of the less diffuse components of the valent p-orbitals of the Cl atoms are in agreement with the corresponding experimental values for the -modification of 1,4-Cl2C6H4.Translated fromIzvestiya Akademii Nauk. Mass determination by mass spectrometric methods (electrospray ionization mass spectrometry (ESI-MS), matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOFMS)) of sodiumdodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE)-separated proteins is a well known procedure and reliable protocols are available. In our efforts to use the established methods to determine the molecular mass of the disulfide bridged, heterodimeric glycoprotein GP3 and to determine the carbohydrate content of each protein subunit we developed an in-gel chemical deglycosylation method. For this purpose we established experimental conditions that allow maximum extraction of the high molecular mass protein subunits and developed a routine method to apply the HF-pyridine deglycosylation protocol to proteins isolated from polyacrylamide gel pieces. The novel protocol and extraction procedure described can be used to analyze O-glycosylated proteins up to 150 kDa after SDS-PAGE separation.  相似文献

The electron beam process has proved to be an efficient method for the removal of inorganic pollutants from flue gas. Since it simulates natural processes which occur in the atmospheric photochemistry, it appeared attractive to investigate the potential of the e-beam process to clean off-gases which contain hydrocarbon and inorganic trace components. Such emissions arise from industrial processes and from automobile tunnels. Commercial solvents were vaporized in air and irradiated with energetic electrons (300 keV). CO, CO2 and aerosol particles were found as products and were determined quantitatively. The aerosol particles can be collected by a gravel bed filter and can be removed by combustion or biological degradation. From experiments and model calculations it was found that the e-beam process is a very economic tool to remove hydrocarbons from large off-gas volumes at initial concentrations of 50–100 mg C/m3, and that NOx can be removed very efficiently from tunnel off-gas.  相似文献

Effects of water content and carbon and nitrogen sources on the production ofL-glutamate oxidase (GOD) by solid state fermentation (SSF) ofStreptomyces sp. N1 were investigated in a 250-mL shake flask. The results show that in the solid medium containing wheat bran 98% (w/w), KCl 0.2% (w/w), and MgCl2 0.2% (w/w), addition of 2.0-mL water per gram solid medium and 0.4% (w/w) (NH4)2SO4 was the best for GOD production. In this work, we also developed a simple technique forin situ measuring oxygen uptake rate (OUR) and carbon dioxide evolution rate (CER) in SSF in a shake flask based on the principle of Warburg manometer. The method was successfully applied to determine OUR and CER values in SSF ofStreptomyces sp. N1. The results indicate that the largest OUR value was detected about one or two days ahead of the highest GOD activity reached depending on the fermentation conditions, and the OUR may be used as anin situ indicator of GOD production in the SSF process.  相似文献

A fast and sensitive high performance liquid chromatography method for quantitative determination of rosiglitazone in human plasma has been developed. The extraction from plasma was performed using solid-phase extraction (SPE) on C4 silica (100 mg) disposable extraction cartridges (DEC). The separation of rosiglitazone and two metabolites was achieved on a Phenomenex® Synergi 4 µm MAX-RP (150 × 4.6 mm) column, protected by a guard column. The mobile phase was 0.01 M ammonium acetate, pH 7.0 - acetonitrile (65:35, v/v). (3S)-3-OH-quinidine was used as internal standard. The analytes were detected using fluorescence detection. The method was validated. The limit of quantitation was 1 ng mL−1 and the detection limit was 0.25 ng mL−1 for rosiglitazone in human plasma. The recovery was 90% for rosiglitazone. Linearity was observed over a range of 1-1000 ng mL−1 (r2=0.9959). The intra- and inter-day precision (C.V.) did not exceed 8.7 %. Applicability of the method was demonstrated by a clinical pharmacokinetic study. A healthy volunteer received in two separate phases 4 mg and 8 mg rosiglitazone maleate as a single oral dose. Plasma concentrations were measured for 24 h in both phases.  相似文献

A series of methyl methacrylate polymers have been prepared containing sucrose-based crosslinkers and additives. Thermogravimetry and long-term aging studies at 200°C show that sucrose-based alkyl and allyl ethers provide unprecedented thermal stability to linear, as well as crosslinked, poly (methyl methacrylate) or PMMA. Linear PMMA and PMMA crosslinked with trimethylolpropane trimethacrylate (TMPTMA) both degrade at 284°C. PMMA containing octa-O-crotylsucrose (1 mol %) degraded at 322°C. Depending on concentration, PMMA containing octa-O-allylsucrose (0.1-1.0 mol % and higher) degraded between 334 and 354°C, and PMMA containing 1′,6,6′-trimethacryloyl-2,3,3′,4,4′-penta-O-methylsucrose (0.1-1.0 mol %) degraded between 309 and 320°C. PMMA containing (1 mol % each) sucrose-based esters, ester-ether derivatives, all degraded at or below the degradation temperature of pure PMMA. Long-term air aging studies revealed that PMMA containing penta-O-methylsucrose trimethacrylate, octa-O-allylsucrose, and octa-O-crotylsucrose did not flow or sag after heating for 24 h at 200°C, but the polymers did show yellowing. While linear and crosslinked samples of PMMA containing compounds other than sucrose ethers lost more than 50% of their original weight within 15 h at 200°C, PMMA containing sucrose-based ethers lost about 8 and 20% of their original weight after 1 and 8.5 days, respectively. Herein we propose a unique mechanism by which saccharide ethers may be imparting this unprecedented thermal stabilization to PMMA. While tertiary hydrogens alpha to oxygens in saccharide ethers are stable to chain transfer during normal polymerization temperatures, they readily chain transfer at 200°C where PMMA is unstable. Chain transfer of these hydrogens is followed by fragmentation to produce alkyl, allyl or crotyl radicals, which combine with the macroradicals and terminate depropagation. © 1995 John Wiley & Sons, Inc.  相似文献

The proton NMR spectra of N-[2-pyridyl-N-oxide]-derivatives of primary and secondary ethylamines, containing a substituent R on the C atom bearing the amino function, have been completely analysed in terms of the fundamental NMR parameters. The preferred conformations of the compounds investigated were established by the indications from NOE experiments as well as: (1) the long range coupling across the five bond between the aminic hydrogen and the proton in 4-position of the pyridine-N-oxide ring (5JmH,NH ~ 0·5 c/s), (2) the value of the vicinal coupling constant in the fragment CHNH (3JNHCH ~ 7–9 c/s), (3) the large deshielding (Δτ ~ 1–1·5 ppm) observed for the resonance position of the proton on the asymmetric C atom in secondary amine derivatives with respect to the corresponding primary ones, and (4) the diamagnetic shielding produced on protons in position 3 and 4 of the pyridine-N-oxide ring by different aromatic groups introduced in the R substituent.The NMR data confirmed the preferred rotamers previously suggested on the basis of ORD and CD measurements.  相似文献

Zusammenfassung Der Aufschluß von Nahrungsmitteln tierischen Ursprungs mit Salpetersäure-Schwefelsäure in einer Polyäthylenflasche bei 60° C und mit Salpetersäure in einem Bombengefäß bei 150° C wurde untersucht. Beide Verfahren liefern befriedigende Ergebnisse. Im zweiten Fall besteht wegen kleinerer Reagensmengen jedoch eine geringere Gefahr des Einschleppens von Blindwerten. Blei, Cadmium und Quecksilber können aus der Aufschlußlösung durch Adsorption an dem Austauscherharz Chelex 100 oder durch Fällung mit Ammonium-pyrrolidindithiocarbamat unter Zusatz von Kupfer als Träger abgetrennt werden. Arsen und Tellur lassen sich anschließend zusammen mit Kupferträger durch Schwefelwasserstoff ausfällen. Die Ausbeuten für Aufschluß und Abtrennung liegen bei 90% oder darüber. — Die quantitative Bestimmung wurde mit Hilfe der energiedispersiven Röntgenfluorescenzanalyse durchgeführt, wobei 109Cd bzw. 241Am oder eine Röntgenröhre mit Zirkon oder Molybdän als Sekundärstrahler zur Anregung verwendet wurden. Beim Blei und Quecksilber konnten noch 1 g, beim Arsen und Tellur noch 0,5 g quantitativ bestimmt werden.
Investigation of a method for the determination of lead, cadmium, mercury, arsenic and tellurium in food by means of X-ray fluorescence analysis
The decomposition of food of animal origin with a mixture of nitric acid and sulphuric acid in a polyethylene flask at 60° C and with nitric acid in a steel vessel at 150° C was investigated. Both procedures give satisfactory results. The advantage of the second procedure lies in the small amounts of reagents needed, thus keeping blank values low. Lead, cadmium and mercury can be separated from the decomposition solution by the chelating resin Chelex 100 or by precipitation with ammonium pyrrolidine dithiocarbamate, using copper as carrier. Arsenic and tellurium may be precipitated in the filtrate by hydrogen sulphide, using copper as carrier. The overall yields are about 90% or more. — The quantitative determination was performed by means of the energy dispersive X-ray fluorescence analysis, using 109Cd or 241Am sources or an X-ray tube with zirconium or molybdenum as secondary target for the excitation. 1 g of lead and mercury and 0.5 g of arsenic and tellurium could still be determined.

In on-column or in splitless injection with recondensation of the solvent, the length of the flooded zone in the column inlet depends primarily on the wettability of the internal wall of the inlet. For columns with a coated inlet this explains why peak distortion due to band broadening in space by a certain sample volume is pronounced in one case and hardly observable in another. Glass or fused silica capillaries silylated with diphenyltetramethyldisilazane were found to give optimal retention gaps. They combine thorough and thermostable deactivation with good wettability and low retention power. On the other hand it is very easy to deactivate fused silica capillaries with Carbowax. The resulting retention gaps are suitable for a wide range of applications and are particularly attractive for the analysis of dirty samples which require frequent replacement of the inlet.  相似文献
