Manipulation of Single‐Stranded DNA by Using an Artificial Site‐Selective DNA Cutter Composed of Cerium(IV)/EDTA and Phosphonate–Oligonucleotide Conjugates

An artificial site‐selective DNA cutter to hydrolyze single‐stranded DNA at a desired site was prepared from Ce^IV/ethylenediamintetraacetic acid (EDTA) and two ethylenediamine‐N,N,N′,N′‐tetrakis(methylenephosphonic acid)–oligonucleotide conjugates. By using this cutter, the sense strand of a blue fluorescent protein (BFP) gene was selectively cut at a predetermined site in the chromophore‐coding region. The upstream fragment obtained by the site‐selective scission was ligated with the downstream fragment of the closely related green fluorescent protein (GFP) gene so that the 5′‐ and 3′‐end portions of the chromophore came from the BFP fragment and the GFP fragment, respectively. The recombinant gene was successfully expressed in E. coli and the chimeric chromophore emitted green fluorescence as expected.     

Microcalorimetric study on expression of foreign genes in Bacillus thuringiensis

The thermogenic curves of the aerobic metabolism of the three strains of Bacillus thuringiensis B.t. A, B.t. B and B.t. C have been determined by using an LKB-2277 BioActivity Monitor. B.t. A was the host bacterium without foreign gene. B.t. B and B.t. C were constructed by transforming different foreign genes into the host B.t. A, respectively. B.t. B expressed erythromycin resistant gene, while B.t. C expressed both erythromycin resistant gene and tyrosinase gene. The heat flow rate of these strains is B.t. A> B.t. B >B.t. C. These results indicated that there is obvious interrelation between expression of foreign genes and heat flow rate of B.t. strains. 受乳糖启动子控制的基因表达过程的优化

借助于ＳＤＳ－聚丙烯酰胺凝胶电泳分析方法，对乳糖诱导的由乳糖启动子控制的大肠杆菌ＲＮＡ聚合酶σ７０因子基因表达过程进行了优化研究．结果表明，含此融合基因的大肠杆菌菌株ＢＬ２１（ＤＥ３）ｐＧＥＭＤ的生长量达到ＯＤ６００ｎｍ＝１．０时，用终浓度为０．０８％的乳糖进行连续５ｈ的诱导，可使大肠杆菌ＲＮＡ聚合酶σ７０因子基因的表达量达到最大，并达到ＩＰＴＧ同样的诱导效果．流加低浓度的乳糖溶液能增加σ７０因子的合成量，但是促进作用不很显著．     

TRANSFER OF THE ATRAZINE-RESISTANT GENE OF BLACK NIGHTSHADE TO SOYBEAN CHLOROPLAST GENOME AND ITS EXPRESSION IN TRANSGENIC PLANTS

The atrazine-resistant psbA gene of black nightshade was transferred to the chloroplast genome of atrazine-susceptible soybean by means of ovary microinjection during the stage of zygote. The identification was carried out by using the methods of spraying the leaves directly with atrazine solution, examining the change of leaf fluorescence kinetics under a brighter light induction, molecular hybridization, etc. The experimental results show that the transgenic soybean plants do have been obtained for the first time.     

B组轮状病毒WH-1株nsp2基因的原核表达与抗体制备

根据B组轮状病毒(GBRV)WH-1株nsp2基因的全序列,设计引物,用PCR的方法扩增得到nsp2基因的编码区.将其片段克隆到原核表达载体pGEX-KG上,经IPTG诱导,在E.coliDH5α菌株中得到高效表达.表达的GST融合蛋白大小为61×103,经SDS-PAGE分离纯化,免疫小白鼠制备了多克隆抗体.抗体经1∶3 000倍稀释后用于Western Blot分析,获得特异性显色信号.所表达的蛋白和制备的抗体可用于蛋白结构和功能的进一步研究.     

介导GAD基因的人泡沫病毒载体的构建及其应用

构建了两种新的重组人泡沫病毒载体pGPSNI—hGAD67和pGPSNI—hGAD65，分别携带人符氨酸脱羧酶（GAD）的两种同工酶GAD65和GAD67基因，并研究其在帕金森病（PD）模刭鼠丘脑底核中的表达及对帕金森病的治疗作用．RT—PCR结果表明，人泡沫病毒载体成功地介导了GAD65基因和GAD67基因在PD模型鼠丘脑底核中有效表达．动物行为检测表明，GAD65基因导入组PD模型鼠行为与对照组相比得到明显的改善（n=12，由〈0．01）．     

分子标记与QTL间连锁检测与估计的方差分析法及其应用

拓展了适用于多种作图群体的检测数量性状基因座位（ＱＴＬ）与分子标记（ＭＬ）间连锁关系的方差分析模型，并提出利用该方差分析之期望均方，在一定的遗传假定前提下，能够用于估计ＭＬ与ＱＴＬ间的重组值．文章最后给出了一个分析实例，将水稻广亲和基因进行了定位．
