氯化血红素的催化显色反应及分析应用

本文研究了以氯化血红素替代辣根过氧化物酶催化过氧化氢与4-氨基安替比林和苯酚的催化显色反应条件,拟订了测定过氧化氢的高灵敏催化分光光度法,做了多种离子对测定过氧化氢的干扰实验。并通过与葡萄糖氧化酶联用,拟订了水介质和血清中葡萄糖的分光光度测定方法,试用于测定血清样品中的葡萄糖含量得到了较满意的结果。     

Chiral discrimination ofN-(dansyl)-dl-amino acids on human serum albumin stationary phase: Effect of a mobile phase modifier

Summary The effect of perchlorate anion as mobile phase modifier on the separation factor, α, forN-(dansyl)-dl-norvaline andN-(dansyl)-dl-tryptophan on a human serum albumin (HSA) column was studied by varying the concentration,c, of the chaotropic agent and the column temperatureT. Gibbs-Helmholtz parameters Δ(ΔH) and Δ(ΔS) between thed andl enantiomers were determined from linear van't Hoff plots of lnα against 1/T. Thermodynamic results indicated that the enhancement of the separation factor observed asc was increased was enthalpically controlled owing to stereoselective H-bonding interactions. Such behavior was used to optimize the chromatographic conditions for separation ofN-(dansyl)-amino acids on HSA.     

Chiral separation of bupivacaine enantiomers by capillary electrophoresis partial-filling technique with human serum albumin as chiral selector

Capillary electrophoresis (CE) is a powerful technique for enantiomer separations due to its intrinsic high separation efficiencies, speed of analysis, low reagent consumption and small sample requirements. However, some chiral selectors present strong background UV absorption providing high detection limits. The present paper deals with the application of the partial-filling technique to the separation of bupivacaine enantiomers by capillary electrophoresis using human serum albumin (HSA) as chiral selector. In this procedure the cationic surfactant cetyltrimethylammonium bromide (CTAB) was used as a dinamic capillary coating in order to reduce the electro-osmotic flow and detect both bupivacaine enantiomers out of the chiral selector plug. Several experimental conditions such as CTAB concentration, pH, HSA concentration and plug length, background electrolyte concentration, temperature and voltage were studied. Under the selected conditions it is possible to detect the separated enantiomers out of the HSA plug in less than 4 min using 50 mM Tris pH 8 as background electrolyte with 50 microM CTAB, at 30 degrees C and using a separation voltage of 25 kV. The proposed methodology was then validated for analytical purposes and applied to the analysis of pharmaceutical preparations commercially available. The results obtained with the proposed methodology were in good agreement with those declared by the manufacturers. The simplicity, sample throughput, accuracy, reproducibility and low cost of the proposed method make it suitable for the control of the enantiomeric composition of bupivacaine in pharmaceuticals.     

中药血清药理学研究金银花的抑菌活性

采用最低抑菌浓度(MIC)测定方法和抑菌活性试验,考察不同产地的金银花以及含药血清的抑菌效果。试验结果为:含药血清对大肠杆菌和金黄色葡萄球菌的MIC分别为0.5μg绿原酸/mL和0.25μg绿原酸/mL;含药血清抑菌活性为高敏。     

Determination of pefloxacin in serum and pharmaceutical forms by derivative spectrophotometry

A method for the direct determination of pefloxacin in serum and pharmaceutical forms (tablets and ampoules) has been developed, based on the use of second-order derivative ultraviolet spectra. Spectrophotometric assay of pefloxacin in tablets and ampoules was carried out in 0.1 mol/L NaOH, while in serum it was performed in 0.1 mol/L NaOH with the addition of sodium dodecylsulfate, in 337–347 nm wavelength range. Linear calibration curves were obtained in the concentration ranges 2–30g/mL pefloxacin for tablets and ampoules and 0.12–5 g/ mL for serum samples. Relative error of determination, as criterion for accuracy, was less than 1%, while the precision was better than 4 ng/ml. The minimum detectable concentration of pefloxacin in serum was 15 ng/mL.     

用铍试剂测定人血清白蛋白的分光光度法

在pH3.6的缓冲溶液中，铍试剂Ⅲ与人血清白蛋白（HSA）能形成复合物，吸光度与HSA的含量呈线性关系；复合物的最大吸收峰波长λmax=460nm,比铍试剂本身紫移80nm左右；HSA标准曲线线性范围为20－140mg/L；实验表明该反应选择性、重复性很好，操作简便，适用测定含量范围宽，测定结果与传统的考马斯亮蓝法结果相吻合。     

偶氮胭脂红B与血清蛋白质相互作用的光度法研究及其分析应用

在酸性介质中，偶氮胭脂红B（ABX）与蛋白质在室温下能迅速结合生成复合物，其最大吸收波长为570nm，比ABX红移了50nm。本文用光度法研究了该结合反应的最佳条件，并在此基础上建立了一个测定蛋白质的新方法。在570nm处各蛋白质的浓度至少在2.5～120mg/L范围内与吸光度成正比，对测定牛血清蛋白质（BSA）、人血清蛋白（HSA）及γ-球蛋白（IgG）的桑德尔灵敏度分别为0.173、0.173和0.199μg/cm^2。除阴阳离子表面活性剂外，基余大部分物质不干扰蛋白质的测定。可见方法具有选择性好、灵敏度高且快速、稳定的优点，用于尿液及人血清样品中总蛋白的测定，所得结果与考马斯亮蓝方法测得结果基本一致。     

On-line SFE-GC for determination of PCBs in human milk and blood serum

On-line coupled supercritical fluid extraction and gas chromatography (SFE-GC) has been utilized for the determination of PCBs and other organochlorine compounds in human milk and blood serum. The procedure involved preconcentration of the sample on C₁₈-silica sorbent in an extraction cell: after precipitation of the proteins up to 20 ml of human milk was concentrated on 0.5 g of sorbent. Serum (up to 5 ml) was applied to the C₁₈ material without pretreatment. Basic alumina was utilized as a selective adsorbent for lipids in the on-line SFE-GC system. The method was used to analyze milk and serum spiked with 0.5 and 10 ng of Aroclor 1260 and the results compared with those obtained by liquid–liquid extraction of serum.     

Immobilization of a Monolayer of Bovine Serum Albumin on Gold Nanoparticles for Stereo‐specified Recognition of Dansyl‐norvaline

The assembly strategy to prepare a monolayer of bovine serum albumin on the surface of silica gel supported gold nanoparticles is described. The stereo‐specific recognition ability of this material was evaluated by enantioresolution of Dansyl‐norvaline. For enantiomeric separation, the influences of buffer concentration and the concentration of organic modifier on the separation performance were investigated. A better separation in terms of enantioresolution and peak shape was found with the phosphate concentration at 30 mM. Moreover, the peak shape and resolution can be improved by the addition of methanol solution. Enantioresolution of Dansyl‐norvaline was obtained from this material at optimized conditions. It appears that the immobilization of a monolayer of bovine serum albumin on gold nanoparticles as the chiral selector of Dansyl‐derivative amino acid is promising.
