An investigation of heparinase immobilization   总被引:1,自引:0,他引:1  
A systematic investigation of the parameters that affect the efficiency of immobilizing heparinase onto cyanogen bromide activated crosslinked 8% agarose beads was conducted. Two experimental measures, the “fraction bound” and the “fraction retained,” were used to monitor the coupling efficiency. The fraction bound is the portion of the total initial enzyme that is bound to the agarose gel. The fraction retained is the fraction of bound enzyme that is active. The product of the two measures indicates the coupling efficiency. The activity of the immobilized heparinase was measured under conditions free of both internal and external mass transfer limitations, and thus, the fraction retained represents the true immobilized enzyme activity. Increasing the degree of activation of the beads results in an increase in the fraction bound, the fraction retained, and consequently, the coupling efficiency. As the ratio of enzyme solution to gel volume increases from 1.5 to 2.2, the fraction bound remains constant but the fraction retained decreases (heparinase concentration; 0.15 mg/mL and degree of activation; 9.5 μmol of cyanate esters/g of gel). At volume ratios greater than 2.2, both the fraction bound and the fraction retained decline continuously. Changing the heparinase concentration in the coupling solution changes the coupling efficiency in a manner similar to that of the volume ratio change. When heparin is added during the coupling process, the fraction bound declines as the heparin concentration increases, whereas the fraction retained increases up to a heparin concentration of 12 mg/mL and decreases thereafter. When arginine, lysine, and glycine are used to block the unreacted cyanate ester groups after the coupling process, the immobilized heparinase shows different pH optima of 6.5, 6.9, and 7.2, respectively. Based upon these findings, a protocol to optimize heparinase immobilization is developed.  相似文献

Fair bandwidth allocation (FBA) has been studied in optical burst switching (OBS) networks, with the main idea being to map the max-min fairness in traditional IP networks to the fair-loss probability in OBS networks. This approach has proven to be fair in terms of the bandwidth allocation for differential connections, but the use of the ErlangB formula to calculate the theoretical loss probability has made this approach applicable only to Poisson flows. Furthermore, it is necessary to have a reasonable fairness measure to evaluate FBA models. This article proposes an approach involving throughput-based-FBA, called TFBA, and recommends a new fairness measure that is based on the ratio of the actual throughput to the allocated bandwidth. An analytical model for the performance of the output link with TFBA is also proposed.  相似文献

An efficient red phosphorescent organic light emitting diode (PhOLED) has been realized by utilizing a composite hole transporting layer comprised of all-inorganic cesium lead halide perovskite CsPbBr3 via spin-coating and 1,3-bis(9-carbazolyl) benzene (mCP) by vacuum depositing, in which CsPbBr3 film is used as a hole transporting layer and mCP plays a dominant role in electron and exciton blocking. And this PhOLED shows a saturated red emission coordinated at CIE (0.65, 0.33) driven at 7.5 V, a maximum brightness of 20,750 cd/m2, and a maximum current efficiency of 10.64 cd/A, which is as 1.87 times as that 5.68 cd/A of the reference PhOLEDs based on traditional small organic molecular hole transporting material N,N′-bis(naphthalen-1-yl)-N,N′-bis(phenyl)-benzi (NPB). The electroluminescent (EL) spectra and the energy level alignment of different PhOLEDs are investigated. The enhanced EL performances are ascribed to improved hole injecting and transporting behaviors, and better electron and exciton confinements by introducing the composite hole transporting layer CsPbBr3/mCP.  相似文献

In this letter, lateral GaN-based Light Emitting Diodes (LEDs) with a SiO2 current blocking layer (CBL) buried in the indium tin oxide (ITO) film and highly reflective metal materials have been proposed. Compared with the conventional CBL structure which was inserted between ITO film and p-type GaN, simulation results showed that LEDs with a buried CBL in the ITO film effectively facilitated current spreading under the CBL. We demonstrated that buried CBL was beneficial for suppressing current crowding (CC) effect around the edge of CBL and may facilitate higher LED efficiency. Furthermore, experimental results showed that LEDs with the buried structure we proposed showed lower working voltage and higher light output power (LOP) compared with those with conventional CBL structure. These results further confirmed that the buried CBL scheme was effective to reduce current crowding (CC) effect. In addition, highly reflective metal materials of Cr/Al/Pt/Au were employed to reduce light absorption and achieve high light extraction efficiency.  相似文献

The adaptive modulation and coding (AMC) technique, which has been adopted by advanced mobile telecommunication systems, supports a flexible response to the random radio behaviour. As a result, the attained transmission rate over a wireless link is time varying. Hence, resource demands are not deterministic but fluctuating even for calls with constant bit rate service requirements. Consequently, constant bit rate calls are susceptible to a forced call termination because of insufficient resources not only in a target cell during inter‐cell handoffs but also in a serving cell during radio link deterioration. Furthermore, call blocking and dropping probabilities depend on radio propagation conditions among other factors and therefore they are dissimilar throughout a service area. The latter leads to unfairness problems. We analytically measure the impact of AMC on fixed‐rate service with hard delay constraints such as voice for different signal, mobility and traffic conditions. We consider a reference case (call requests are admitted into the system provided there are enough free resources) and two classes of admission control approaches: traditional (only inter‐cell handoffs are prioritised) and modified (all ongoing calls are prioritised). This paper presents a channel reservation and preemption (CRP) model using overlapping regions in a cellular network with multiple sectors. To fully exploit and reuse the frequencies, directional antennas are installed on base stations (BSs) to divide the coverage into a number of equal‐sized sectors. When traffic is unevenly distributed across the sectors in a BS, channel utilization in every sector may become very different; low‐traffic sectors may be underutilized while high‐traffic sectors may be overutilized. A CRP scheme is thus proposed to more efficiently utilize free channels among sectors. CRP aims at reducing the dropping probabilities of handoff calls. Specifically, when free channels in a sector are not available, a handoff call, instead of being dropped, is allowed to preempt an ongoing call residing in the overlapping region of two adjacent sectors or two neighbor cells. Under CRP, the preempted ongoing call will not be disconnected, because it can switch over its service to the BS of a neighbor cell or to another directional antenna of an adjacent sector. For the purpose of evaluation, we build an analytical model for the proposed CRP using six‐tuple Markov chains. Analytical results show that the proposed CRP can significantly reduce the dropping probabilities of inter‐sector handoff calls, particularly when traffic between two sectors is not evenly distributed. Several analytical models have been proposed to study the blocking probability for personal communications service networks or mobile phone networks. These models cannot accurately predict the blocking probability because they do not capture two important features. First, they do not capture the busy-line effect. Even if a cell has free channels, incoming and outgoing calls must be dropped when the destination portable is already in a conversation. Second, they do not capture the mobility of individual portables. In these models, mobility is addressed by net hand-off traffic to a cell, which results in traffic with a smaller variance to a cell compared with the true situation. We propose a new analytic model which addresses both the busy-line effect and individual portable mobility. Furthermore, our model can be used to derive the portable population distribution in a cell. The model is validated against the simulation experiments. We indicate that the previously proposed models approximate a special case of our model where the number of portables in a cell is 40 times larger than the number of channels.  相似文献

针对波分复用(WDM)光网络在实现端到端业务链接时的波长连续性限制问题,提出了对光网络进行稀疏多光纤配置的方法。通过分析发现:稀疏多光纤配置能够使整个网络的性能受波长连续性限制的影响得到明显改善;具有稀疏多光纤配置的光网络长链路建链阻塞概率在全网阻塞率中的比例要高于使用相同数目波长转换器的光网络,这说明,波长转换器对于光网络的长链建链仍具有一定的优势。  相似文献
