吡啶二甲酸镓、铟配合物的合成及生物活性 总被引:1,自引:0,他引:1
吡啶二甲酸能与过渡、非过渡、镧系和锕系金属离子螯合形成稳定的配合物,而且配位方式不拘一格,既可以是双齿,三齿,也可以是桥式配位。吡啶二甲酸的另一重要意义在于它们的生物活性。由于镓具有抗癌,抗肿瘤的活性,吡啶二甲酸镓配合物可能会是一种非常有潜力的新型药物。基于上述事实,本文合成了六种新型的吡啶二甲酸(2,3-,2,5-,2,6-)镓(Ⅲ)、铟(Ⅲ)配合物,并进一步测试了三种镓配合物的生物活性。 相似文献

Characteristic features of the kinetics of solid-state cage reactions with distributed parameters of the relaxing matrix were
considered. Depending on the ratio of the constants of the reaction rate and relaxation of environment, the kinetics of chemical
conversions can be either exponential or nonexponential. Plausible reasons for the unsteady-state character of the kinetics
of the processes of two types,viz., the reactions of alkyl radicals in amorphous alcohol matrices and conversions in biological systems, were discussed. The
main reason for the unsteady-state character of the reactions of the first type is a dispersion of the equilibrium distances
between the reagents. Kinetics of the reactions of the second type, such as rebinding of the ligands in the heme-containing
proteins (e.g., in myoglobin), is determined by the distances in the pairs of reagents and the relaxation transitions.

Yinlan lipid regulatory capsule (YL) is a composite traditional Chinese medicine (TCM) new drug to alleviate hyperlipidemia, while its therapeutic mechanism in vivo was not clarified with nontargeted metabolomics investigation. An animal model was established in rats fed a high-fat diet, and their body weights, body mass index (BMI) and blood cholesterol levels were measured. Serum, liver and kidney tissue samples were also extracted for PXR-CYP3A4-ABCB1-FXR signaling pathway research using PCR and UHPLC–MS. The obtained plasma samples were analyzed by UHPLC-Q-TOF-MS metabolomic investigation, which revealed PXR-CYP3A4-related metabolites and changes induced by YL. Finally, the key metabolites were chosen as index components, and their levels in the serum, liver, small intestine and bile were used for simultaneous UHPLC–MS-MS determination. The results indicated that YL was effective in rebalancing blood TG and TC levels (compared to controls). With respect to the PXR-CYP3A4-ABCB1 pathway, as a result of YL’s effect, gene expression or activity of the two targets decreased significantly in both the liver and kidney. The same trend was observed in the serum samples mentioned above. Metabolomics screening and data revealed that 44 metabolites can be regarded as biomarkers related to hyperlipidemia, fatty acids synthesis, and body energy consumption, as well as synthesis, transportation and exertion of cholesterol. YL’s treatment focused on 26 of them, primarily bile acids, indicating that the antihyperlipidemic effect of this drug lies in its inhibitory activity of cholesterol metabolism. Subsequent analysis of those in vivo components revealed that significant increases (compared to the model group) occurred in the blood, liver, small intestine and bile in groups that received medium and high doses of YL (while the low dose was relatively unchanged). Those target components exhibit a close relationship with PXR and/or CYP3A4. The use of YL repressed PXR expression and subsequently decreased CYP3A4 activity. As a result, synthesis of related bile acids increased, while cholesterol levels decreased, consequently leading to the attenuation of hyperlipidemia. This study comprehensively investigated the antihyperlipidemia mechanism of YL based on its repression of PXR-CYP3A4 activity and related metabolite yield, establishing an accurate method for evaluating the therapeutic effect of YL. 相似文献

Summary In this paper we investigate the potential of alkyl-bonded silica monolithic columns for the isolation and identification
of drug-related components in biological fluids. Up to 6 columns have been connected in series to produce a chromatographic
system with up to 40,000 plates. This high-resolution chromatography system has been coupled to both MS and NMR to enable
efficient detection and characterisation of drug-related components in biological fluids. The use of six coupled columns has
been shown to give enhanced resolution over a high quality silica particulate column packed with 3 μm material which exhibits
the same back pressure. The effect of volume and mass load on the performance of monolithic columns for semi-preparative chromatography
of biological fluids has also been investigated. In these studies it was possible to inject up to 100 mL of neat urine with
no loss of chromatographic performance. Furthermore, upon re-testing, the columns showed similar chromatographic performance.

A new isoprenyl phenyl ether, 3-hydroxy-4-(3-methylbut-2-enyloxy)benzoic acid methyl ester (1), together with 4-hydroxybenzoic acid (2), 2-hydroxy-6-methylbenzoic acid (3), and 4-hydroxy-3-methoxybenzoic acid (4) were isolated from Mangrove fungus (No. B60) from the South China Sea. The structures of the compounds were established
on the basis of NMR spectroscopic and mass spectrometric data. In the preliminary bioassay, compound 1 exhibited antibacterial and antifungal activities. Compound 1 also inhibited cytotoxicity to the hepG2 cell line with an IC50 value of 10.0 μg/mL.

Pectinic substances from the aerial part of Mentha x piperita were isolated and characterized and found to be a mixture of β-(1→4)-glucogalactan (MPG) and two α-(1→4)-rhamnopolygalacturonans (MPP′-1 and MPP′-2). It was shown that the pectin and its components exhibited membrane-stabilizing,
antiatherogenic, and antioxidant activity. Hemicelluloses from M. piperita were a lignocarbohydrate complex.
