A simple lab-on-chip approach with time-based detection is proposed. A platform is made from a piece of acrylic differently shaped channels for introducing sample and reagent(s) using flow manipulation. Time-based changes involving migration of the reaction zone are monitored. The changes can be visually monitored by using a stop-watch with naked eyes observation. This article proposes a new hybrid visualization technique that integrates a frequency-based model and a generalized parallel coordinate plot (GPCP), thus mitigating the visual cluttering of GPCP. In the new technique, a GPCP’s profile lines (or curves) with similar frequencies are detected and saturated with appropriate color intensity corresponding to the frequencies. The technique may be employed to enhance a family of visualization tools—the Andrews plot and scatterplot matrix, for example. In addition to the new technique’s efficiency in reducing visual clutter in the multivariate data visualization techniques, it is computationally feasible, easy to implement, and has important mathematical and statistical properties. The reliability and accuracy of the technique are demonstrated through extensive experiments on challenging datasets, both simulated and real. These datasets are high in dimensions and large so that they cannot be explored with GPCP or frequency-based techniques alone. This paper presents a novel approach for accelerating the popular reciprocal nearest neighbors (RNN) clustering algorithm, i.e. the fast-RNN. We speed up the nearest neighbor chains construction via a novel dynamic slicing strategy for the projection search paradigm. We detail an efficient implementation of the clustering algorithm along with a novel data structure, and present extensive experimental results that illustrate the excellent performance of fast-RNN in low- and high-dimensional spaces. For definitive diagnosis of thalassemia carriers and patients, as well as for prenatal diagnosis, genotype analysis is of fundamental importance. We report a dry-reagent, lateral flow dipstick test that enables visual genotyping (detection by naked eye) of 15 mutations common in Mediterranean populations in the beta-globin gene (HBB). The method comprises 3 simple steps: (i) PCR amplification of a single 1896 bp segment of the beta globin gene flanking all 15 mutations; (ii) a multiplex (10-plex and/or 30-plex) primer extension reaction of the unpurified amplification product using allele-specific primers. Biotin is incorporated in the extended product; (iii) a dry-reagent multi-allele (10-plex) dipstick assay for visual detection of the primer extension reaction products within minutes. The total time required for PCR, primer extension reaction and the dipstick assay is ∼2 h. The method was evaluated by genotyping 45 DNA samples of known genotypes and 54 blind samples. The results were fully concordant with reference methods. The method is simple, rapid, and cost-effective. Detection by the dipstick assay does not require specialized instrumentation or highly qualified personnel. The proposed method could be a particularly useful tool in laboratories with limited resources and a basis for point-of-care diagnostics especially in combination with PCR amplification from whole blood. 相似文献

Due to the poor road markings and irregular driving behaviors, not every vehicle is positioned in the center of the lane. The deviation from the center can cause discomfort to drivers in the neighboring lane, which is referred to as lateral discomfort (or lateral friction). Such lateral discomfort can be incorporated into the driver stimulus–response framework by considering the visual angle and its changing rate from the psychological viewpoint. In this study, a two-lane visual angle based car-following model is proposed and its stability condition is obtained through linear stability theory. Further derivations indicate that the neutral stability line of the model is asymmetry and four factors including the vehicle width and length, the lateral separation and the sensitivity regarding the changing rate of visual angle have large impacts on the stability of traffic flow. Numerical simulations further verify these theoretical results, and demonstrate that the behaviors of diverging, merging and lane changing can break the original steady state and cause traffic fluctuations. However, these fluctuations may be alleviated to some extent by reducing the lateral discomfort. 相似文献
