The discrimination of complex mixtures, especially those with very similar compositions, remains a challenging part of chemical analysis. In this paper, a single cataluminescence (CTL) sensor constructed using MgO nanomaterials in a closed reaction cell (CRC) was used to identify vinegars. It may provide an archetype of this type of highly multicomponent system. By scanning the CTL spectra, which were distributed in 15 wavelengths during the reaction period, the spectral array patterns of the vinegars were obtained. These functioned as their fingerprints. The CTL signals of the array were then normalized and identified through linear discrimination analysis (LDA). Nine types and eight brands of vinegars and an additional series of artificial samples were tested; the new technique was found to distinguish between them very well. This single sensor demonstrated excellent promise for analysis of complex mixtures in real-world applications and may provide a novel method for identifying very similar complex analytes.  相似文献

The lateral-flow immunoassay (LFA) is an inexpensive and rapid paper-based assay that can potentially detect infectious disease biomarkers in resource-poor settings. Despite its many advantages that make it suitable for point-of-care diagnosis, LFA is limited by its inferior sensitivity relative to sophisticated laboratory-based assays. Our group previously introduced the use of a micellar aqueous two-phase system (ATPS), comprised of the nonionic Triton X-114 surfactant, to concentrate biomarkers in a sample and enhance their detection with LFA. However, achieving complete phase separation and target concentration using the Triton X-114 system required many hours, and the concentrated sample needed to be manually extracted and applied to LFA. Here, we successfully integrated the concentration and detection steps into a single step that occurs entirely within a portable paper-based diagnostic strip. In a novel approach, we applied the micellar ATPS to a 3-D paper design and effectively reduced the macroscopic phase separation time from 8 h to approximately 3 min. The 3-D design was integrated with LFA to simultaneously concentrate and detect Plasmodium lactate dehydrogenase (pLDH), a malaria biomarker, in both phosphate-buffered saline and fetal bovine serum within 20 min at room temperature. Compared to a conventional LFA setup with a pLDH detection limit of 10 ng μL−1, our single-step diagnostic successfully detected pLDH at 1.0 ng μL−1, demonstrating a 10-fold detection limit improvement and resulting in a sensitive and user-friendly assay that can be used at the point-of-care. The integration of a micellar ATPS and LFA represents a new platform that can improve and promote the use of paper-based diagnostic assays for malaria and other diseases within resource-poor settings.  相似文献

A sensitive and rapid on-line immunoassay for the determination of ciguatoxin CTX3C was developed based on a capillary mixing system, which was integrated with capillary electrophoresis (CE) separation and electrochemical (EC) detection. In the sandwich immunoassay system, anti-CTX3C-functionalized magnetic nanoparticles were used as immunosensing probes, and horseradish peroxidase (HRP) and anti-CTX3C antibody were bound onto the surface of gold nanoparticles (AuNPs) and used as recognition elements. Online formation of immunocomplex was realized in capillary inlet end with an external rotating magnetic field. Compared with classical HPLC-MS and ELISA, the assay adopting AuNPs as multienzyme carriers and online sandwich immunoassay format with rotating magnetic field exhibited higher sensitivity and shorter assay time. The linear range of the assay for CTX3C was from 0.6 to 150 ng/L with a correlation coefficient of 0.9948 (n = 2), and the detection limit (S/N = 3) was 0.09 ng/L. The developed assay showed satisfying reproducibility and stability, and it was successfully applied for the quantification of CTX3C in fish samples.  相似文献

β‐Agonists are a group of illegal feed additives. In this paper, it was found that the light emission produced by the oxidation of luminol by potassium ferricyanide was enhanced by the β‐agonists (ractopamine, salbutamol, and terbutaline). Based on chemiluminescence phenomenon, a novel, rapid, and sensitive microflow injection analysis system on a microfluidic glass chip was established for determination of the β‐agonists. The chip was fabricated from two glass plates (64 mm × 32 mm) with microchannels of 200 μm width and 100 μm depth. The detection limits were achieved at 2.0 × 10?8 mol/L of ractopamine, 1.0 × 10?8 mol/L of terbutaline and 5.0 × 10?7 mol/L of salbutamol. In this report, our method was applied for determination of the β‐agonists in swine hair from three different sources with satisfactory results.  相似文献

根据竞争法免疫层析试条的检测原理,本研究将其分为TwA和TnA两种模式,结合对流扩散方程分别建立其数学模型,并运用COMSOL软件对试条的动态反应过程进行仿真,得到检测线和质控线上各复合物浓度关于各影响因素的关系曲线.在TwA模式中,分析了待测物浓度[A0]=0~20 mol/L,标记物浓度[P0]=0.01~100 mol/L,检测线位于5~20 mm位置等因素对于检测结果的影响;在TnA模式中,分析了[A0]=0~20 mol/L, [P0]=0.01~100 mol/L, [A0]和[P0]两种物质浓度及孔隙率等因素对于检测结果的影响.结果表明,本研究建立的竞争法模型与仿真能探究各参数对于检测结果的影响,优化试条性能,从而提高试条检测灵敏度和实现定量检测.  相似文献

A novel chemiluminescence (CL) performance of CdTe/CdS/ZnS quantum dots (QDs) with periodate (KIO4) was studied. Effects of concentration and pH on the CL system were investigated. Electron spin resonance (ESR) and the effects of radical scavenger analysis were employed for identification of intermediate species. The CL spectra for this system showed only one maximum emission peak centered around 620 nm, which was similar with photoluminescence (PL) spectra of CdTe/CdS/ZnS QDs. The CL of CdTe/CdS/ZnS QDs was induced by direct chemical oxidation and the possible mechanism could be explained by radiative recombination of injected holes and electrons. This investigation not only provided new sight into the optical characteristics of CdTe/CdS/ZnS QDs, but also broadened their potential optical utilizations.  相似文献

建立了间接竞争酶联免疫吸附法检测食品中细交链格孢酮酸(Tenuazonic Acid,TA)残留,并研制快速检测试剂盒。采用肟化法对TA进行衍生化,活泼酯法偶联半抗原和载体蛋白得到人工抗原TAO-BSA和TAO-KLH,以TAO-KLH作为免疫原免疫雌性Balb/c小鼠制备特异性抗体。对包被浓度、包被时间、封闭液类型、抗体工作浓度、二抗稀释度、底物显色时间等参数进行研究,建立了TA残留间接竞争酶联免疫检测方法并进行了配套试剂盒的研制。该试剂盒半抑制浓度Ic_(50)为1.48ng/mL,检测线性范围为0.06~35.95ng/mL(R~2=0.9941);检测限为0.02ng/mL;加标样品平均回收率大于88.50%;试剂盒的批间和批内平均变异系数分别为2.84%和9.49%,与食品中常见真菌毒素交叉反应率均小于1%。  相似文献

本文基于岩白菜素对鲁米诺-牛血清白蛋白体系化学发光信号显著的抑制作用,建立了快速灵敏测定岩白菜素的流动注射-化学发光分析新方法。实验发现,化学发光强度的降低值与岩白菜素质量浓度对数值呈良好的线性关系,方法的线性范围为3.0~5.0×10~5 pg/mL,检出限(3σ)为1.0pg/mL。当溶液流速为2.0mL/min时,完成一次分析过程仅需30s,采样频率120/h。本法用于片剂、人血清和尿液中岩白菜素的含量测定,回收率为98.1%~102.7%,相对标准偏差小于2.0%(n=7)。同时对化学发光反应机理进行了探讨。  相似文献

基于头孢唑啉钠能抑制磷钼钒杂多酸氧化鲁米诺产生化学发光,结合流动注射分析技术,提出了测定头孢唑啉钠的化学发光新体系。头孢唑啉钠在3.0×10-7~8.0×10-5 g/mL质量浓度范围内与化学发光信号呈线性关系;检出限为8.0×10-8 g/mL;对5.0×10-6 g/mL的头孢唑啉钠进行11次连续测定,相对标准偏差为3.8%。该方法快速、灵敏,已经被成功地应用于头孢唑啉钠针剂的测定。  相似文献
