The aggregation interaction between reduced-denatured egg white lysozymes during refolding procedure in urea solution was studied by means of reducing and non-reducing protein electrophoreses. Results of non-reducing sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) of the supernatant and aggregate precipitate formed in refolding process show that except being refolded to native egg white lysozymes, the reduced-denatured lysozymes can also form the aggregates with molecular weights (MW) being separately about 30.0 and 35.0 kD, while the reducing SDS-PAGE and the refolding results in the presence of sodium dodecyl sulphate show that these aggregates are formed chiefly through the misconnection of disulfide bonds between the reduced-denatured lysozymes, and the aggregate precipitates are formed through the non-covalent interactions between the aggregates with molecular weight being about 30.0 kD. From the results of electrophoresis and size-exclusion chromatographic analyses, it can be inferred that the aggregates with molecular weights being about 30.0 and 35.0 kD are bi-molecular and tri-molecular egg white lysozyme aggregates, respectively. And finally, a suggested refolding mechanism of reduced-denatured egg white lysozymes in urea solution was presented.  相似文献

In the presence of white phosphorus the redox potentials of the copper ions change and the potential of the reduction wave of CuI/Cu0 shifts noticeably toward more positive values. The CuI—P4 complex is characterized by a lower value of the electrochemical gap, that is, higher polarizability and reactivity compared to those of the free CuI cation. Phosphorus esters can be synthesized from P4 and ethanol. The latter is in the composition of the copper(ii) complexes, which act as a catalyst-charge mediator.  相似文献

Starting from earlier work by Baudler we introduce a chemical heuristic for the systematic deduction and classification of covalent partial structures of phosphorus in polycyclic phosphanes, phosphorus-rich polycyclic phosphides, and allotropes of phosphorus except the black forms. This approach is used to direct ab initio techniques (which also confirm the rules) in the quest for as yet unknown forms of molecular or macromolecular phosphorus. Based on calculated stabilities of systematically generated structural alternatives we rationalize the stabilities of Hittorf's phosphorus and of molecular P4, confirm the possible existence of at least one other crystalline allotropic form of phosphorus, and provide insight into the probable structure of amorphous red phosphorus. In total, the combined approach of chemical heuristics and large scale ab initio calculations presented in this work supplies a coherent chemical understanding of covalent polyphosphorus structures.  相似文献

Only noisy nonlinear observations taken on the point location (state) are available toM. The velocityv is of the formv(y)= u (u 1,u 2) y (du), wherey denotes the value of the observed signal,U is the range of the velocity, and, for eachy, y is a probability measure on (U). Using the available observations, the pointM wishes to steer itself into a given target set by choosing a randomized strategy ={ y :y 2}. Sufficient conditions on weak optimal randomized strategies are derived. In the present work, corrosion resistance of surface-coated galvanized steel was quantitatively determined by an analysis of the alternating current (AC) impedance spectra measured on the salt-spray-tested specimen. To evaluate the corrosion resistance of the surface-coated galvanized steel, AC impedance spectroscopy was performed on the salt-spray-tested specimen previously exposed to salt-sprayed corrosive environment. From the analysis of the impedance spectra, the area fraction transient of white rust θ 2(t) was theoretically derived from the equivalent circuit equation by using two fitting parameters. The values of the two fitting parameters were determined by fitting the empirical transient equations to the area fraction of the resin coating layer and to the total resistance obtained from the impedance spectra measured, respectively. From the analyses of θ 2(t) for four kinds of surface-coated galvanized steels with various resin coating layers, it is indicated that as the values of the two fitting parameters decrease in the order of CP, GI, OD and OM (commercial trade names) specimens, the corrosion resistance increases in that order as well. Furthermore, from the quantitative comparison of the two fitting parameters with the polarization resistance of the upper resin coating layer R p determined from the potentiodynamic polarization curve, it is suggested that the two fitting parameters decrease in value as well with increasing R p.  相似文献

 建立了用高效阳离子交换分离纯化蛋清中溶菌酶的新方法 ,讨论了纯化的工艺条件。蛋清样品匀浆后 ,用氯化钠初步纯化 ,然后用弱阳离子交换柱XIDACE WCX分离。结果表明 ,被纯化的溶菌酶和杂蛋白得到很好分离。经活性检测 ,溶菌酶过柱后的活性回收率为 10 7% ,蛋白的比活为 15 4 6 7U/mg ,纯化倍数提高了 5 6倍。用尺寸排阻 (SEC)鉴定 ,得到的溶菌酶呈均一性。该法较传统软基质低压离子交换分离速度快 ,纯化效率高。  相似文献

The effect of carbon and nitrogen sources, lignocellulosic substrates, and metal ions on lignin peroxidase (LiP) activity of Aspergillus sp., which was isolated from a mangrove area, was studied. Glucose (1%) was found to be the best carbon source. Among the various lignocellulosic substrates used, coir pith at 3% concentration increased LiP activity twofold on the second day of incubation. Peptone and KNO3 completely inhibited the enzyme synthesis while (NH4)2SO4 at 12.5 mM produced maximum activity. Since seawater contained all the requisite metal ions, any added ions had a negative effect on activity. Cu2+ had the most inhibiting effect while K+ the least. When all the optimized conditions were provided, in nitrogen- and carbon-sufficient medium, a maximum LiP activity of 345 U/mL was obtained on the second day of incubation.  相似文献

Abstract

The paper deals with the influence of high pressure treatment of fresh egg white on its properties and protein composition (individual amino-acids predicted as a function of pressure and time levels). The rheological properties are changed by high pressure from Newtonian to non-Newtonian behaviour, with increasing apparent viscosity as the pressure and time increased. The pH, whipping ability, foam stability, gel strength of heat induced gels after treatment and the whole protein content, were also predicted.

The results showed that the foam stability is increased with increasing pressure and time of processing. The foam volume is also increased with pressure. The pH did not change with pressure or time of processing. Composition of proteins as indicated by individual amino-acids did not exhibit statistically important changes. Gel strength of heat induced gels prepared from previously pressured liquid whites showed no important change of values with pressure or time of treatment. The modulus of elasticity showed a decrease for samples pressured to 400 MPa for 5 up to 15 minutes.  相似文献
