The demand for more efficient methods of establishing the undetermined stereochemistries of peptidic natural products continues unabated. A new method for microscale stereochemical determination was devised by integrating solid-phase synthesis, split-and-mix randomization, 18O/16O-encoding of d /l -configurations, tandem mass spectrometry, and biological evaluation. Here we applied gramicidin A as the molecule for a blind test. Gramicidin A and its 31 diastereomers were randomly prepared in microgram scale with 18O/16O-stereochemical encoding and subjected to MS/MS-structural determination and cytotoxicity assay. Only the parent gramicidin A was selected from among the 32 stereoisomers, validating the high reliability of the present strategy.  相似文献

This study describes the application of a common analytical procedure adapted for compound-specific stable carbon isotope analyses of riverine contaminants. To evaluate the sensitivity of the analytical method and the precision of the isotopic data obtained, a set of numerous substances at different concentration levels were measured. For most of the anthropogenic contaminants investigated (including chlorinated aliphatics and aromatics, musk fragrances, phthalate-based plasticizers and tetrabutyl tin) acceptable carbon isotope analyses could be obtained down to amounts of approximately 5?ng absolutely applied to the gas chromatograph. These amounts correspond to concentrations in water samples at a natural abundance level of approximately 50–200?ng?L?1 (low to medium contaminated river systems). However, it has to be considered that the precision and the sensitivity of the analytical method depend partially on the chemical properties of the substances measured. Five recovery experiments were conducted to assess changes in carbon isotope ratios during sample preparation and measurement. The compounds selected for these experiments are known riverine contaminants. Isotopic shifts or higher variations of the isotope ratios as a result of the analytical procedures applied were observed only for a couple of contaminants. Furthermore, compound-specific carbon isotope analyses were performed on eight water extracts of the Rhine river. By comparing the variation of the data of several individual compounds with the deviations obtained from the recovery experiments, it was possible to differentiate contaminants with unaffected isotope ratios and substances with significant alterations of the δ13C-values.  相似文献

Multivariate statistical analyses were applied on the measured physico-chemical (Cd, Pb, Cu, Zn, Mg, Ca, O2, alkalinity, temperature, pH, SAS, DOC and DIC) and isotopic parameters (δ13C and δ18O) to estimate and distinguish anthropogenic from natural influences to the water system of the Krka River. Analyses were conducted on the data collected during six years from twelve sampling sites. On the basis of orientation, positioning and grouping of parameters arranged by biplots, four main hypotheses were defined and finally statistically confirmed. Thereof, two main and distinct processes occurring in the Krka River could be highlighted: (i) upstream pollution, caused by the inflow of untreated waste-waters of city of Knin and (ii) downstream self-purification, caused by the sedimentation and/or co-precipitation of pollutants coupled by the inflow of clean subterranean water (groundwater recharge). Grouping of (i) hydrological and carbon cycle connected parameters, and (ii) anthropogenically influenced correlated parameters were proposed as a result of statistical analysis. Regarding the pH, it is shown that a stream section influenced by the subterranean inflow of Zrmanja River is statistically significantly different for all sampling campaigns during six years, being lower for about 0.5 pH unit.  相似文献

A facile method of producing stable aqueous dispersion of graphene/polyaniline (PANI) composite is described, which involves the in situ polymerization of aniline on the surface of graphene with the aid of polystyrene sulfonic acid (PSS). The prepared aqueous graphene/PANI composite dispersion was very stable and no aggregation or precipitation was observed for several weeks. The excellent aqueous dispersibility and stability of the graphene/PANI composite is attributed to the cooperative interactions of π stacking interaction between PSS, PANI, and the graphene basal planes, and the electrostatic repulsions between negatively charged PSS bound on graphene/PANI composite. Fourier transform‐infrared spectrometry (FTIR), ultraviolet‐visible spectra (UV–vis), and Raman spectra confirmed the interaction of PANI and graphene in the composite, which effectively delocalize the electrons. In addition, the composite showed three orders of magnitude of conductivity increase compared with pure PANI. This new approach is simple, fast, and straightforward, representing a significant improvement in the processing of graphene/PANI composites. © 2012 Wiley Periodicals, Inc. Stable conformations of 12-crown-O3N and its Li complexes in aqueous solution were investigated. To calculate the free energy differences of conformers of 12-crown-O3N and its Li+ complex, our procedure was to make use of two programs, CONFLEX and BOSS. The former generates conformers, and the latter calculates the differences in free energy of solvation between two conformers in aqueous solution. It was confirmed that the present procedure is applicable in solving the question of what is the most stable conformation of 12-crown-O3N in aqueous solution. Results of the calculations suggest that the order of stability for conformers in a vacuum is different from that in aqueous solution. It was also confirmed that the coordination geometry of solvent waters to Li+ changes depending on the distance between the cation and the crown ring.  相似文献

We present the access to [5-19F, 5-13C]-uridine and -cytidine phosphoramidites for the production of site-specifically modified RNAs up to 65 nucleotides (nts). The amidites were used to introduce [5-19F, 5-13C]-pyrimidine labels into five RNAs—the 30 nt human immunodeficiency virus trans activation response (HIV TAR) 2 RNA, the 61 nt human hepatitis B virus ϵ (hHBV ϵ) RNA, the 49 nt SAM VI riboswitch aptamer domain from B. angulatum, the 29 nt apical stem loop of the pre-microRNA (miRNA) 21 and the 59 nt full length pre-miRNA 21. The main stimulus to introduce the aromatic 19F–13C-spin topology into RNA comes from a work of Boeszoermenyi et al., in which the dipole-dipole interaction and the chemical shift anisotropy relaxation mechanisms cancel each other leading to advantageous TROSY properties shown for aromatic protein sidechains. This aromatic 13C–19F labeling scheme is now transferred to RNA. We provide a protocol for the resonance assignment by solid phase synthesis based on diluted [5-19F, 5-13C]/[5-19F] pyrimidine labeling. For the 61 nt hHBV ϵ we find a beneficial 19F–13C TROSY enhancement, which should be even more pronounced in larger RNAs and will facilitate the NMR studies of larger RNAs. The [19F, 13C]-labeling of the SAM VI aptamer domain and the pre-miRNA 21 further opens the possibility to use the biorthogonal stable isotope reporter nuclei in in vivo NMR to observe ligand binding and microRNA processing in a biological relevant setting.  相似文献

In this study, we demonstrate that an Mn-doped ultrathin Ni-MOF nanosheet array on nickel foam (Mn0.1-Ni-MOF/NF) serves as a highly capacitive and stable supercapacitor positive electrode. The Mn0.1-Ni-MOF/NF shows an areal capacity of 6.48 C cm−2 (specific capacity C: 1178 C g−1) at 2 mA cm−2 in 6.0 m KOH, outperforming most reported MOF-based materials. More importantly, it possesses excellent cycle stability to maintain 80.6 % capacity after 5000 cycles. An asymmetric supercapacitor device utilizing Mn0.1-Ni-MOF/NF as the positive electrode and activated carbon as the negative electrode attains a high energy density of 39.6 Wh kg−1 at 143.8 Wkg−1 power density with a capacitance retention of 83.6 % after 5000 cycles.  相似文献

The synthesis of bench‐stable α,α‐bis(trimethylsilyl)toluenes and tris(trimethylsilyl)methane is described and their use in stereoselective Peterson olefinations has been achieved with a wide substrate scope. Product stereoselectivity was poor with carbonyl electrophiles (E/Z ~1:1 to 4:1) though this was significantly improved by employing the corresponding substituted N‐benzylideneaniline (up to 99:1) as an alternative electrophile. The olefination byproduct was identified as N,N‐bis(trimethylsilyl)aniline and could be easily separated from product by aqueous acid extraction. Evidence for an autocatalytic cycle has been obtained.  相似文献
