We discuss two possible approaches for extracting kinetic information from single-molecule experiments. The first approach is based on computing correlation functions from measured fluorescence signals, and the second on studying the statistics of on and off times of the same fluorescence signal. We show that in both cases it is possible to extract kinetic information about the nature of intramolecular fluctuations of the single molecule. We show that for single-molecule kinetics the intramolecular fluctuations produce stochastic memory effects which lead to new dynamic features that do not exist in traditional chemical kinetics. In particular, we investigate a new type of chemical oscillations in correlation functions observed experimentally by Edman and Rigler (Proc. Natl. Acad. Sci. USA 2000, 97, 8266).  相似文献

A solvent extraction separation of uranium, in the presence of thorium, cerium and lanthanides with a new calix[4]resorcinarene bearing eight hydroxamic acid groups (C4RAHA) is described. Quantitative extraction of uranium is possible in ethyl acetate solution of C4RAHA at pH 8.0. The lambda(max) and molar absorptivity (varepsilon) for uranium is 356nm and 8352Lmol(-1)cm(-1). The Binding ratio of uranium with C4RAHA as evaluated by Job's method is 4:1. The system obeys Beer's law over the range 0.075-6.0mugml(-1) of uranium with Sandell sensitivity 0.0284mugcm(-2). A preconcentration factor of 142 was achieved by directly aspirating the extract for GF-AAS measurements. The two-phase stability constant evaluated at 25 degrees C for uranium is 15.91. The complexation is characterized by favorable enthalpy and entropy changes. A liquid membrane transport study of uranium was carried out from source to the receiving phase under controlled conditions and a mechanism of transport is proposed. Uranium has been determined in standard and environmental samples.  相似文献

Two of the 12 possible oxide-bridged phenylmorphans, were synthesized, rac-(3R,6aS,11aS)-2-methyl-1,3,4,5,6,11a-hexahydro-2H-3,6a-methanobenzofuro[2,3-c]azocine-10-ol (7) (the ortho-c compound), and rac-(3R,6aS,11aS)-2-methyl-1,3,4,5,6,11a-hexahydro-2H-3,6a-methanobenzofuro[2,3-c]azocine-8-ol (8) (the para-c compound). Single-crystal X-ray diffraction studies indicated that the dihedral angle between the least squares planes through the phenyl ring and the atoms C1, C11a, C12, and C3 in the piperidine ring in both 7·CHCl3 and 8·HBr was 6.9°. The C12-C6a-C6b-C10a torsion angle was found to be 139.3° for both compounds. The angular relationship between the phenolic ring and the piperidine ring in phenylmorphans that interact with specific opioid receptors as agonists or antagonists is of considerable theoretical interest.  相似文献

The viscoelastic properties of single poly(ethylene glycol) (PEG) molecules were measured by analysis of thermally and magnetically driven oscillations of an atomic force microscope (AFM) cantilever/molecule system. The molecular and monomer stiffness and friction of the PEG polymer were derived using a simple harmonic oscillator (SHO) model. Excellent agreement between the values of these two parameters obtained by the two approaches indicates the validity of the SHO model under the experimental regimes and the excellent reproducibility of the techniques. A sharp minimum in the monomeric friction is seen at around 180 pN applied force which we propose is due to a force induced change in the shape of the energy landscape describing the conformational transition of PEG from a helical to a planar state, which in turn affects the timescale of the transition and therefore modifies the measured internal friction. A knowledge of the viscoelastic response of PEG monomers is particularly important since PEG is widely used as a linker molecule for tethering groups of interest to the AFM tip in force spectroscopy experiments, and we show here that care must be exercised because of the force-dependent viscoelastic properties of these linkers.  相似文献

A series of [tetra-α-(2,2,4-tirmethyl-3-pentoxy) phthalocyaninato] metal complexes(M=Co, Ni, Cu, Zn)were prepared by cyclotetramerization of 3-(2,2,4-tirmethyl-3-pentoxy)phthalonitrile with corresponding anhydrous metal salts, using 1.8-diazabicyclo[5.4.0] undec-7-ene (DBU) as the catalyst. HPLC analysis shows that one isomer predominates in the product of nickel complex, while at least two main constitutional isomers exist in the product of other metal complexes. The complexes (in the form of mixture of constitutional isomers) were characterized by elemental analyses, MS, IR and UV-Vis spectroscopy.  相似文献

N"-Substituted isonicotinic hydrazides of the general formula Py—C(=O)—N(H)-N"=C(H)—R, where R is o- (1), m- (2), or p-nitrophenyl (3), were studied by IR spectroscopy and X-ray diffraction analysis. The position of the nitro group in these compounds has no effect on the type of the crystal structure. The crystal packings are based on stacks consisting of antiparallel planar molecules. The molecules from the adjacent stacks are linked to each other via the N—H...NPy hydrogen bonds. Depending on the position of the nitro group, the N...NPy distance increases in the series 3 > 1 > 2 and the energy of the hydrogen bonds decreases (according to the IR spectroscopic data) from 3.9 to 3.1 kcal mol–1. Analysis of the IR spectra demonstrated that the intensity of absorption in the (C—H) stretching region of the pyridine ring increases substantially as the the N—H...NPy hydrogen bond is strengthened. Some regularities of the changes, which are observed for the (NO2) bands in the spectra of the nitrophenyl-containing conjugated molecules in solutions, persist in the crystalline state.  相似文献

Abstract

Impregnated thin-layer chromatography (TLC) layers based on silica gel are presented. Impregnating agents such as metal cations, inorganic ions, chelating agents, chiral selectors, surfactants, ion-pairing reagents, and ionic liquids are discussed. The role of impregnated TLC layers in medicinal chemistry is highlighted. The historical overview of TLC separations on physically coated layers is given and some future prospects ahead of this technique are discussed.  相似文献

A label- and immobilization-free approach to detecting the reversible formation of complexes between nucleic acids and proteins at the single-molecule level is described. The voltage-driven translocation of individual oligoribonucleotides through a nanoscale protein pore is observed by single-channel current recordings. The oligoribonucleotide 5'-C25A(25)-3' gives rise to current blockades with an average duration of approximately 0.5 ms. In the presence of the RNA-binding ATPase P4, a viral packaging motor from bacteriophage phi8, longer events of tens to hundreds of milliseconds are observed. Upon addition of ATP the long events disappear, indicating the dissociation of the P4RNA complex. The frequency of events also depends on the concentration of P4 and the length of the oligoribonucleotide, thereby confirming the specificity of the P4RNA events. This study shows that single-channel current recordings can be used to monitor RNA-protein complex formation, thus opening up a new means to examine the motor activity of RNA- or DNA-processing enzymes.  相似文献

The novel methacrylic monomer, 4-nitro-3-methylphenyl methacrylate (NMPM) was synthesized by reacting 4-nitro-3-methylphenol dissolved in ethyl methyl ketone (EMK) with methacryloyl chloride in the presence of triethylamine as a catalyst. The homopolymer and copolymers of NMPM with glycidyl methacrylate having different compositions were synthesized by free radical polymerization in EMK solution at 70 ± 1 °C using benzoyl peroxide as free radical initiator. The homopolymer and the copolymers were characterized by FT-IR, 1H NMR and 13C NMR spectroscopic techniques. The solubility tests were tested in various polar and non-polar solvents. The molecular weight and polydispersity indices of the copolymers were determined using gel permeation chromatography. The glass transition temperature of the copolymers increases with increase in NMPM content. The thermogravimetric analysis of the polymers performed in air showed that the thermal stability of the copolymer increases with NMPM content. The copolymer composition was determined using 1H NMR spectra. The monomer reactivity ratios were determined by the application of conventional linearization methods such Fineman-Ross (r1 = 1.862, r2 = 0.881), Kelen-Tudos (r1 = 1.712, r2 = 0.893) and extended Kelen-Tudos methods (r1 = 1.889, r2 = 0.884).  相似文献

Aniline was polymerized directly into polyaniline‐sulfate salt without using protonic acid in this work. Polyaniline‐sulfate salt was prepared by emulsion and aqueous polymerization pathways. The dopant i.e. sulfate ion in polyaniline‐sulfate salt was generated from ammonium persulfate which was used for oxidizing aniline. Ammonium persulfate acts both as oxidizing agent as well as protonating agent in the polymerization process of aniline to polyaniline salt. The efficiency of oxidizing and protonating power of ammonium persulfate is increased by the use of surfactant. The activity of ammonium persulfate is further increased by the use of sulfuric acid as protonic acid. It may be necessary to consider the effect of sulfate ion which is generated during the oxidation process of aniline in the chemical polymerization of aniline to polyaniline salt by ammonium persulfate either aqueous or emulsion polymerization pathway in the presence of protonic acid/functionalized protonic acid.
