A novel, specific, reliable, and accurate capillary zone electrophoretic method was developed and validated for the simultaneous determination of aliskiren hemifumarate, amlodipine besylate, and hydrochlorothiazide in their triple mixture dosage form. Separation was carried out in a fused‐silica capillary (57.0 cm total length and 50.0 cm effective length, 75.6 μm internal diameter) by applying a potential of 17 kV and a running buffer consisting of 40 mM phosphate buffer at pH 6.0 with UV detection at 245 nm. The method was suitably validated with respect to specificity, linearity, LOD, and LOQ, accuracy, precision, and robustness. The method showed good linearity in the ranges 1–10, 2.5–25, and 30–300 μg/mL with LODs of 0.11, 0.33, and 5.83 μg/mL for amlodipine besylate, hydrochlorothiazide, and aliskiren hemifumarate, respectively. The proposed method was successfully applied for the analysis of the studied drugs in their coformulated tablets. The results of the proposed method were statistically compared with those obtained by the RP‐HPLC reference method revealing no significant differences in the performance of the methods regarding accuracy and precision.  相似文献

A zone‐folding (ZF) approach is applied for the estimation of the phonon contributions to thermodynamic properties of carbon‐and ZrS2‐based nanotubes (NTs) of hexagonal morphology with different chiralities. The results obtained are compared with those from the direct calculation of the thermodynamic properties of NTs using PBE0 hybrid exchange‐correlation functional. The phonon contribution to the stability of NTs proved to be negligible for the internal energy and small for the Helmholtz free energy. It is found that the ZF approach allows us an accurate estimation of phonon contributions to internal energy, but slightly overestimates the phonon contributions to entropy. © 2015 Wiley Periodicals, Inc.  相似文献

On-line coupled analytical techniques can be advantageous in the assay of smaller peptides in complex biological matrices such as plasma, cerebrospinal fluid (CSF) and tissues. The present study shows the feasibility of a recently developed system, consisting of a size-exclusion chromatographic (SEC) separation followed by a trapping procedure on an RP18 microcolumn with subsequent elution of the trapped fraction and separation by capillary zone electrophoresis (CZE) for the quantification of structural-related peptides in biological matrices, as demonstrated for a number of enkephalins in CSF. After SEC separation of the enkephalins from large proteins present in CSF a heart-cut of 200 nuL, containing the enkephalin peak, is taken, concentrated on the RP18 microcolumn and, after elution of the enkephalins with 80% acetonitrile, a fraction of the eluate is electrokinetically injected into the CZE system, where stacking and separation is achieved. The degradation of the peptides, caused by endogenous peptidases in the matrix, is sufficiently inhibited with imipramine HCl. The assay has a satisfactory linearity and intraday (9.70-16.3%) precision considering the complexity of this multidimensional separation system. The sensitivity of the method, with a concentration limit of quantification of 2.5 nug/mL, is comparable with other CZE assays for peptides and sufficient for the quantification of peptide drugs in biological matrices.  相似文献

An automatic sampling device has been developed for capillary zone electrophoresis. The sample is introduced to the column electrokinetically by rapidly exchanging buffer for sample in a narrow channel drilled in a plexiglass block. The autosampler is capable, under computer control, of performing multiple sample injections from a large volume sample source (such as a reaction vessel) as the sample concentration changes, and thus presents the possibility of analyzing time-varying processes by CZE. Peak area reproducibility in electropherograms obtained after use of the sampler is less than 1% and efficiency is more than 2.2 × 105 theoretical plates.  相似文献

Capillary zone electrophoresis (CZE) has been utilized to profile the low-density (LDL) particles in human blood serum in this study. A 5 mM sodium phosphate buffer, pH 7.40, was chosen as the most suitable CE buffer and an extensive ultrafiltration (UF) procedure was applied to purify the LDL sample. Two LDL particle species, LDL with lower mobility and LDL- with higher mobility were observed. The electropherograms were highly reproducible with good precision of effective mobilities, corrected peak areas (CPAs) and CPA ratio of LDL-/LDL. LDL particles shown on the electropherogram were also characterized by several procedures. The applications of Sigma HDL cholesterol reagent and CE on-line 2-propanol precipitation indicated that the two particle species shown in the electropherogram belong to LDL. The LDL particles were found to associate with the buoyant LDL fraction and the LDL- particles associate with the dense LDL fraction. This study utilizes CZE for the profiling of LDL isoforms and provides a new analytical method for the resolution of LDL subspecies. It demonstrates a high-mobility LDL particle which circulates in healthy subjects and diminishes in atherosclerotic patients. Diminution of the high-mobility LDL subspecies may be linked to minimal formation of arterial plaque in atherosclerotic patients.  相似文献

Siderophores are low molecular weight non-ribosomal peptides with extremely high affinity by iron. However, other metals present affinity for siderophores but to a smaller degree. Deferoxamine is an example of a bacterial hydroxamic siderophore, which was investigated herein. Capillary zone electrophoresis (CZE) was used as a new approach in the continuous variation method for the characterization of metal-deferoxamine complexes. A set of samples containing both metal (e.g., Fe(III), Fe(II) or Ni(II)) and siderophore with different molar ratios was prepared and analyzed by both CZE and UV-vis spectrophotometry. A phosphate buffer pH 8.0 was used as the background electrolyte in the first case due to best complex and free ligand peaks resolution. The Job's plots obtained from complex peak areas (complex concentration) versus metal molar fraction revealed complexes stoichiometries of M : L of 2 : 3, 1 : 2 and 1 : 1 for Fe(III), Fe(II) and Ni(II) complexes, respectively. Conditional formation constants could also be calculated for Fe(III) and Fe(II) complexes as Kf = 1.03 × 1013 and 2.47 × 104, respectively. UV-visible spectrophotometric analysis confirmed the data obtained for Fe(III)-complex.  相似文献

软弱岩带的变形模量对于工程建设意义重大,当前变形试验方法难以表征原位条件下软弱岩带的变形模量,导致取值偏于保守。本文基于弹性力学推导了下卧软弱岩带变形模量的解析解,建立了一种新的软弱岩带变形模量的取值方法。在现场试验成果的基础上,采用有限元反演,数值模拟成果与试验计算成果较为吻合,证明该计算方法合理可行,较常规方法直接在软弱岩带上开展的试验成果大幅提高。  相似文献

The concepts of subcritical and supercritical flows are introduced for the long-wave approximation model describing stationary free-boundary rotational flows of an ideal incompressible fluid. Shear flows of a fluid layer above an uneven bottom are analyzed. Exact solutions describing different flow regimes are constructed, and the flow properties are studied as a function of the flow regime.
