A surgical site infection (SSI) is an infection that occurs after surgery in the part of the body where the surgery took place. An SSI may range from a spontaneously limited wound discharge within 7–10 days of an operation to a life‐threatening postoperative complication, such as a sternal infection after open heart surgery. Most SSIs are caused by contamination of an incision with microorganisms from the patient's own body during surgery. From the analytical point of view, the complex nature of these samples as well as the low concentrations of analytes require a system with high sensitivity and efficiency. Such situation requires a technique such as CE, which is a powerful and versatile separation technique that promises to rival HPLC when applied to the separation of both charged and neutral species. During the study, it has been demonstrated that CZE identifies characteristics of such groups of pathogens such as bacteria Gram (+) and different species of bacteria Gram (?), and also develops weekly individual profiles for patients after application of antibiotics. This was done in order to show the impact of antibiotic therapy in change “numbers” of bacteria present in the wound after surgery. The method proved to be the ideal straight specificity in the case of Escherichia coli (100%). Finally, analysis of the spectra and the second derivatives of the UV‐Vis spectra confirmed the similarity in the profiles and showed that the CZE is a great method for fast screening test in bacterial infection. 相似文献

采用毛细管电泳分析方法对维甲酸异构体进行了分离。比较了区带电泳和胶束电动色谱两种模式对维甲酸异构体的分离效果。考察了缓冲液组成、pH值、乙腈含量以及十二烷基硫酸钠(SDS)浓度等对分离的影响。采用胶束电动色谱法可完全分离13-顺-和全反式-维甲酸,最佳的缓冲溶液为(20 mmol/L Tris-H3BO3(pH 8.5)-25 mmol/L SDS)-乙腈(体积比为80∶20)。该法首次应用于加标血浆中的维甲酸分析,获得了较好的结果。 相似文献

A new method using capillary zone electrophoresis was developed for the rapid quantification of two common uronic acids, galacturonic acid and glucuronic acid, based on utilization of an alkaline background electrolyte with reversed electroosmotic flow (EOF) within 16 min. The method relies on in‐capillary reaction and direct UV detection at the wavelength 270 nm. The optimum electrolyte solution was prepared of 130 mm sodium hydroxide, 36 mm disodium hydrogen phosphate dihydrate and 0.5 mm cetyltrimethylammonium bromide. EOF was reversed to detect uronic acids and to improve the separation of neutral sugars. The established method was validated and the results showed good linearity, high precision and satisfactory sensitivity. The newly developed method was successfully applied to analyze galacturonic acid and glucuronic acid content in Forsythia suspensa polysaccharides. A novel two-dimensional electrophoretic system for the control of electroosmosis in capillary zone electrophoresis has been developed and evaluated for rapid separations of proteins. The system comprises uncoated and polyether-coated fused silica capillaries coupled in series. An equation relating the average electroosmotic flow velocity in the coupled capillaries to the intrinsic electroosmotic velocities of the connected segments and their corresponding lengths has been derived and verified experimentally. This approach has the advantage of enabling the electroosmotic flow to be tuned independently of the applied voltage. As a consequence, rapid protein analysis at relatively low field strength was achieved without sacrificing the high separation efficiencies obtained with surface-modified capillaries. 相似文献

Summary Capillary zone electrophoresis using cyclodextrins and a chiral crown ether as buffer constituents was studied for the enantiomeric separation of drugs and amino acids. Based on results obtained from separation of racemic -amino acids both chiral selectors are compared with respect to resolution, efficiency and retention time. For (±)-Quinagolide effects of buffer composition and temperature are examined using -cyclodextrin as chiral agent. Optimum conditions were pH 2.5 at 30 mmol L–1 -cyclodextrin. A linear dependence of retention on -cyclodextrin concentration allowed calculation of formation constants of the host-guest complexes. Buffer concentration and temperature also influence resolution. The application of a chiral crown ether to the separation of optical isomers in capillary zone electrophoresis is described for the first time. Chiral recognition of solutes depends on the formation of protonated alkyl amines and separation is attributed to the formation of diastereomeric host-guest complexes with different interactions for each enantiomer. A rapid method of capillary zone electrophoresis is described to determine the oxidized (GSSG) and reduced (GSH) form of glutathione in plant tissue. In order to separate both analytes in a fused-silica capillary, the pH and composition of the electrolyte solution were optimized. The electrolyte composition was 100 mmol/L, borate 25 mmol/L Tris, and 0.2% w/v metaphosphoric acid (MPA), pH 8.2. Some instrumental conditions used to run the samples were hydrostatic injection for 30 s, 30 kV applied voltage, and UV detection (185 nm) at 25 degrees C. Linearity and useful range obtained for the calibration curves were optimum, with correlation coefficients about 0.999 in the 0-120 micromol/L range. The migration time was highly reproducible, less than 5 min being afforded to run a sample. Electrolyte buffer and samples required a careful pH control for optimal separation of both analytes. This aspect constitutes a critical analytical step when acids are used in the procedure for sample preparation. Simultaneous analysis of GSH and GSSG may provide a useful tool for comparative studies of plants in order to select those species with a potential capacity for detoxification from toxic elements or those appearing promising from phytoremediation for these elements. 相似文献

Abstract A capillary zone electrophoresis (CZE) method for the simultaneous determination of the β-blocker drugs atenolol and chlorthalidone in pharmaceutical formulations has been developed. The CZE separation was performed under the following conditions: capillary temperature, 25°C; applied voltage, 25 kV; 20 mM H3PO4–NaOH running buffer (pH 9.0); and detection wavelength, 198 nm. Phenobarbital was used as internal standard. The method was validated and showed not only good precision and accuracy but also good robustness. The method has been successfully applied to the simultaneous determination of both atenolol and chlorthalidone in pharmaceutical tablets. 相似文献
